The fluid-mosaic model posits a liquid-like plasma membrane, which can flow in response to tension gradients. It is widely assumed that membrane flow transmits local changes in membrane tension across the cell in milliseconds, mediating long-range signaling. Project description:The migration-selection balance often governs the evolution of lineages, and speciation with gene flow is now considered common across the tree of life. Ecological speciation is a process that can facilitate divergence despite gene flow due to strong selective pressures caused by ecological differences; however, the exact traits under selection are often unknown. The transition from freshwater to saltwater habitats provides strong selection targeting traits with osmoregulatory function. Several lineages of North American watersnakes (Nerodia spp.) are known to occur in saltwater habitat and represent a useful system for studying speciation by providing an opportunity to investigate gene flow and evaluate how species boundaries are maintained or degraded. We use double digest restriction-site associated DNA sequencing to characterize the migration-selection balance and test for evidence of ecological divergence within the Nerodia fasciata-clarkii complex in Florida. We find evidence of high intraspecific gene flow with a pattern of isolation-by-distance underlying subspecific lineages. However, we identify genetic structure indicative of reduced gene flow between inland and coastal lineages suggesting divergence due to isolation-by-environment. This pattern is consistent with observed environmental differences where the amount of admixture decreases with increased salinity. Furthermore, we identify significantly enriched terms related to osmoregulatory function among a set of candidate loci, including several genes that have been previously implicated in adaptation to salinity stress. Collectively, our results demonstrate that ecological differences, likely driven by salinity, cause strong divergent selection which promotes divergence in the N. fasciata-clarkii complex despite significant gene flow.

Project description:White blood cells (WBCs) constitute about 0.1% of the blood cells, yet they play a critical role in innate and adaptive immune responses against pathogenic infections, allergic conditions, and malignancies and thus contain rich information about the immune status of the body. Rapid isolation of WBCs directly from whole blood is a prerequisite for any integrated immunoassay platform designed for examining WBC phenotypes and functions; however, such functionality is still challenging for blood-on-a-chip systems, as existing microfluidic cell sorting techniques are inadequate for efficiently processing unprocessed whole blood on chip with concurrent high throughput and cell purity. Herein we report a microfluidic chip for continuous-flow isolation and sorting of WBCs from whole blood with high throughput and separation efficiency. The microfluidic cell sorting chip leveraged the crossflow filtration scheme in conjunction with a surface-micromachined poly(dimethylsiloxane) (PDMS) microfiltration membrane (PMM) with high porosity. With a sample throughput of 1 mL h(-1), the microfluidic cell sorting chip could recover 27.4 ± 4.9% WBCs with a purity of 93.5 ± 0.5%. By virtue of its separation efficiency, ease of sample recovery, and high throughput enabled by its continuous-flow operation, the microfluidic cell sorting chip holds promise as an upstream component for blood sample preparation and analysis in integrated blood-on-a-chip systems.

Project description:An experimental study of momentum transfer in nanoporous polymeric track-etched membranes with pore diameters ranging from 100 to 1300 nm and nanochannel lengths of 12-20 μm was performed using He, N2, CO2, and SF6 propellants in a wide range of plenum and background pressures. Mass flux through the membranes was elaborated as a combination of Knudsen diffusion and viscous flow at Knudsen numbers above 0.1 and become choked at lower Knudsen numbers. The discharge coefficient for the membranes attained was 0.6, making the permeation rate similar to that of thin orifices. The effect is attributed to the mirror reflection of the molecules from the pore walls at low angles of incidence. The exhaust gas velocity is found to be dependent on the plenum to background pressure ratio and channel length-to-diameter ratio, reaching 0.9 of the velocity of the gas expanded to vacuum (up to 2 M). Close to an isothermal expansion occurs in nanochannels of all sizes. A general quantitative description for gas expansion in nanochannels is provided. The highest thrust is generated in the choked flow regime with the SF6 propellant and a value of 4.5 N cm-2 is attained at a propellant consumption of 0.165 kg (cm2 s)-1 for the membranes with 1300 nm nanochannels. The specific impulse of 138 s is reached for helium. The results show the prospects of the utilization of nanoporous membranes in cold gas propulsion systems.

Project description:The development of chemically stable and high conductive membranes is one of the most important issues to improve the performance of vanadium flow batteries (VFBs). Herein, poly(vinylidene fluoride) (PVDF)/graphene composite nanoporous membranes were easily fabricated by manipulating crystallization processes. The graphene was used to enhance membrane selectivity and conductivity. In the nanoscale channels of the membranes, the graphene nanosheets reduced the apertures among the crystal grains, thus restraining vanadium ions crossover due to the size exclusion effect. Moreover, the oxygen groups on the graphene improved the surface hydrophilicity and formed hydrogen bonds with the PVDF polymer chains, which facilitated the proton transport. The composite membranes, with a 0.15 wt % graphene loading, showed a selectivity of 38.2 and conductivity of 37.1 mS/cm. The single cell exhibited a coulomb efficiency of 94.7%, a voltage efficiency of 88.5%, and an energy efficiency of 83.8%, which was 13% higher than that of the pristine PVDF membranes. The composite membranes showed excellent stability during 100 charge-discharge cycles. All these results indicate that the PVDF/graphene composite membrane is a promising candidate for VFB applications.

Project description:Redox flow batteries such as the all-vanadium redox flow battery (VRFB) are a technical solution for storing fluctuating renewable energies on a large scale. The optimization of cells regarding performance, cycle stability as well as cost reduction are the main areas of research which aim to enable more environmentally friendly energy conversion, especially for stationary applications. As a critical component of the electrochemical cell, the membrane influences battery performance, cycle stability, initial investment and maintenance costs. This review provides an overview about flow-battery targeted membranes in the past years (1995-2020). More than 200 membrane samples are sorted into fluoro-carbons, hydro-carbons or N-heterocycles according to the basic polymer used. Furthermore, the common description in membrane technology regarding the membrane structure is applied, whereby the samples are categorized as dense homogeneous, dense heterogeneous, symmetrical or asymmetrically porous. Moreover, these properties as well as the efficiencies achieved from VRFB cycling tests are discussed, e.g., membrane samples of fluoro-carbons, hydro-carbons and N-heterocycles as a function of current density. Membrane properties taken into consideration include membrane thickness, ion-exchange capacity, water uptake and vanadium-ion diffusion. The data on cycle stability and costs of commercial membranes, as well as membrane developments, are compared. Overall, this investigation shows that dense anion-exchange membranes (AEM) and N-heterocycle-based membranes, especially poly(benzimidazole) (PBI) membranes, are suitable for VRFB requiring low self-discharge. Symmetric and asymmetric porous membranes, as well as cation-exchange membranes (CEM) enable VRFB operation at high current densities. Amphoteric ion-exchange membranes (AIEM) and dense heterogeneous CEM are the choice for operation mode with the highest energy efficiency.

Project description:Electrophoresis, the motion of charged species through liquids and pores under the influence of an external electric field, has been the principle source of chemical pumping for numerous micro- and nanofluidic device platforms. Recent measurements of ion currents through single or few carbon nanotube channels have yielded values of ion mobility that range from close to the bulk mobility to values that are two to seven orders of magnitude higher than the bulk mobility. However, these experiments cannot directly measure ion flux. Experiments on membranes that contain a large number of nanotube pores allow the ion current and ion flux to be measured independently. Here, we report that the mobilities of ions within such membranes are approximately three times higher than the bulk mobility. Moreover, the induced electro-osmotic velocities are four orders of magnitude faster than those measured in conventional porous materials. We also show that a nanotube membrane can function as a rectifying diode due to ionic steric effects within the nanotubes.

Project description:Background: The family Trypanosomatidae encompasses parasitic flagellates, some of which cause serious vector-transmitted diseases of humans and domestic animals. However, insect-restricted parasites represent the ancestral and most diverse group within the family. They display a range of unusual features and their study can provide insights into the biology of human pathogens. Here we describe Vickermania, a new genus of fly midgut-dwelling parasites that bear two flagella in contrast to other trypanosomatids, which are unambiguously uniflagellate.Results: Vickermania has an odd cell cycle, in which shortly after the division the uniflagellate cell starts growing a new flagellum attached to the old one and preserves their contact until the late cytokinesis. The flagella connect to each other throughout their whole length and carry a peculiar seizing structure with a paddle-like apex and two lateral extensions at their tip. In contrast to typical trypanosomatids, which attach to the insect host's intestinal wall, Vickermania is separated from it by a continuous peritrophic membrane and resides freely in the fly midgut lumen.Conclusions: We propose that Vickermania developed a survival strategy that relies on constant movement preventing discharge from the host gut due to intestinal peristalsis. Since these parasites cannot attach to the midgut wall, they were forced to shorten the period of impaired motility when two separate flagella in dividing cells interfere with each other. The connection between the flagella ensures their coordinate movement until the separation of the daughter cells. We propose that Trypanosoma brucei, a severe human pathogen, during its development in the tsetse fly midgut faces the same conditions and follows the same strategy as Vickermania by employing an analogous adaptation, the flagellar connector.

Project description:Salt marshes provide valuable ecosystem services including coastal protection by reducing wave loading on dikes and seawalls. If the topsoil is erosion resistant to fast-flowing water, it may also reduce breach depth if a dike fails. In this experiment, we quantified the topsoil erosion resistance from marshes and bare tidal flats with different soil types to understand the extent to which they can help reduce breach depth. Intact soil samples were collected from 11 locations in the Netherlands at different tidal elevations and then exposed for 3 h to 2.3 m/s currents. To the samples that remained stable after flow exposure, an artificial crack was made to test their stability following soil disturbance. All samples from the tidal flats were completely eroded, regardless of sediment type. In contrast, all samples from well-established marsh plateaus were stable as long as no disturbances were made, including those with sandy subsoils. After creating artificial cracks, samples with a thin cohesive top layer on top of sandy subsoil collapsed, while marshes with silty subsoils remained stable. Pioneer marshes on sandy substrate without a cohesive top layer were the only vegetated soils that completely eroded. The lower erosion of marshes with either sandy or silty soils compared to bare tidal flats was best explained by the presence of a top layer with belowground biomass, high organic content, high water content, and low bulk density. When analyzing the erodibility of marshes only, fine root density was the best predictor of erosion resistance. This study demonstrates the importance of preserving, restoring, or creating salt marshes, to obtain a topsoil that is erosion resistant under fast-flowing water, which helps reduce breach dimensions if a dike fails. The probability of topsoil erosion in established marshes with sandy subsoil is higher than in silty marshes. A silty layer of cohesive sediment on top of the sand provides extra erosion resistance as long as it does not break. Pioneer marshes that have not developed a cohesive top layer are erosion sensitive, especially in sandy soils. For future marsh creations, using fine-grained sediments or a mixture of sand with silt or clay is recommended.

Project description:In recent years, polybenzimidazole (PBI) membranes have been proposed for vanadium redox flow batteries (VRFBs) as an alternative to perfluoroalkylsulfonic acid membranes such as Nafion™. Despite their excellent capacity retention, PBI membranes tend to suffer from a low ionic conductivity. The formation of a polybenzimidazolium through an N-alkylation of the benzimidazole core is shown to improve the ionic conductivity of the membrane, with this class of materials having found uses in alkaline fuel cell and water electrolysis systems. However, much less is known about their incorporation into a VRFB. This article describes the use of hexamethyl-p-terphenyl polybenzimidazolium (HMT-PMBI) membranes for a vanadium redox flow battery, with the membrane characteristics in acidic media being related to their performance in a single-cell VRFB setup. A change of the degree of methylation from 56 to 65, 75, and 89% leads to an increase in ionic conductivity, correlated with an increased fraction of free water in the ionomer. The corresponding increase in cell performance is, however, accompanied by a drop in capacity retention. The membrane with a degree of methylation of 65% shows balanced properties, with a 5% higher efficiency and a two times improved capacity retention compared to Nafion™ NR212 over 200 charge-discharge cycles at 200 mA cm-2.

Project description:The membrane is a crucial component of Zn slurry-air flow battery since it provides ionic conductivity between the electrodes while avoiding the mixing of the two compartments. Herein, six commercial membranes (Cellophane™ 350PØØ, Zirfon®, Fumatech® PBI, Celgard® 3501, 3401 and 5550) were first characterized in terms of electrolyte uptake, ion conductivity and zincate ion crossover, and tested in Zn slurry-air flow battery. The peak power density of the battery employing the membranes was found to depend on the in-situ cell resistance. Among them, the cell using Celgard® 3501 membrane, with in-situ area resistance of 2 Ω cm2 at room temperature displayed the highest peak power density (90 mW cm-2). However, due to the porous nature of most of these membranes, a significant crossover of zincate ions was observed. To address this issue, an ion-selective ionomer containing modified poly(phenylene oxide) (PPO) and N-spirocyclic quaternary ammonium monomer was coated on a Celgard® 3501 membrane and crosslinked via UV irradiation (PPO-3.45 + 3501). Moreover, commercial FAA-3 solutions (FAA, Fumatech) were coated for comparison purpose. The successful impregnation of the membrane with the anion-exchange polymers was confirmed by SEM, FTIR and Hg porosimetry. The PPO-3.45 + 3501 membrane exhibited 18 times lower zincate ions crossover compared to that of the pristine membrane (5.2 × 10-13 vs. 9.2 × 10-12 m2 s-1). With low zincate ions crossover and a peak power density of 66 mW cm-2, the prepared membrane is a suitable candidate for rechargeable Zn slurry-air flow batteries.
