A special three-layer step-index fiber for building compact STED systems.

ABSTRACT: Up to now, most of stimulated-emission-depletion (STED) systems were lens-based bulky systems. Exchanging some spatial light paths with optical fiber components will make the systems more flexible and will benefit various fields. A big problem to achieve this goal is that the STED beam generated by the traditional method of bulky systems cannot be maintained in an optical fiber due to its birefringence. In this article, we will introduce a type of special optical fiber. With the special fiber, a dark hollow beam with doughnut-shaped focal spot and a concentric beam with Gaussian-shaped focal spot can be generated at the same time. Parameters of a sample and a compact STED system based on it are demonstrated.

Flexible biodegradable citrate-based polymeric step-index optical fiber.

Project description:Implanting fiber optical waveguides into tissue or organs for light delivery and collection is among the most effective ways to overcome the issue of tissue turbidity, a long-standing obstacle for biomedical optical technologies. Here, we report a citrate-based material platform with engineerable opto-mechano-biological properties and demonstrate a new type of biodegradable, biocompatible, and low-loss step-index optical fiber for organ-scale light delivery and collection. By leveraging the rich designability and processibility of citrate-based biodegradable polymers, two exemplary biodegradable elastomers with a fine refractive index difference and yet matched mechanical properties and biodegradation profiles were developed. Furthermore, we developed a two-step fabrication method to fabricate flexible and low-loss (0.4 db/cm) optical fibers, and performed systematic characterizations to study optical, spectroscopic, mechanical, and biodegradable properties. In addition, we demonstrated the proof of concept of image transmission through the citrate-based polymeric optical fibers and conducted in vivo deep tissue light delivery and fluorescence sensing in a Sprague-Dawley (SD) rat, laying the groundwork for realizing future implantable devices for long-term implantation where deep-tissue light delivery, sensing and imaging are desired, such as cell, tissue, and scaffold imaging in regenerative medicine and in vivo optogenetic stimulation.

Coherent Mid-IR Supercontinuum Generation using Tapered Chalcogenide Step-Index Optical Fiber: Experiment and modelling.

Project description:Mid-infrared region of electromagnetic spectrum has increased a lot of scientific and technical interest because of its utility to figure out the molecular fingerprints. Current mid-infrared light sources including quantum cascade lasers, thermal-emitters, and synchrotron radiation are not suitable for various potential applications where we require coherent, portable and broadband light sources. During the current decade, several efforts have been put forwarded to extend the spectral range of the supercontinuum. However, the coherent mid-infrared supercontinuum spectrum in the mid-infrared region has been demonstrated rarely. Here, we demonstrate a coherent mid-infrared supercontinuum using a tapered chalcogenide fiber pumped at various wavelength ranging from 2?µm to 2.6?µm. Experimental observations show that the supercontinuum spectrum extending from ~1.6?µm to 3.7?µm can be achieved using a 3?cm long tapered chalcogenide step-index optical fiber pumped with femtosecond laser pulses at 2.6?µm. To the best of our knowledge, using short pump wavelengths at 2?µm to 2.6?µm in an all-normal dispersion engineered chalcogenide glass fiber, the coherent supercontinuum spectrum has been reported first time. Such coherent broadband light source has its key prominence for the various prospective applications in the fields of bio-medical, sensing, and multiplex coherent anti-Stokes Raman scattering microspectroscopy.

Modular Two-Step Access to π-Extended Naphthyridine Systems-Potent Building Blocks for Organic Electronics.

Project description:Efficient synthetic approaches for the incorporation of nitrogen into polyaromatic compounds (PACs) in different patterns as stabilising moiety for π-extended systems and modification tool for optoelectronic properties remain a challenge until today. Herein, we developed a new versatile pathway to napthyridine-based PACs as non-symmetric and regioisomeric pendant to pyrazine-based PACs. A combination of a gold-catalysed synthesis of 2-aminoquinolines and the development of an in situ desulfonation and condensation of these precursors are the key steps of the protocol. The shape and type of attached functional groups of the PACs can be designed in a late stage of the overall synthetic procedure by the chosen anthranile and backbone of the ynamide introduced in the gold-catalysed step. Single-crystal X-ray diffraction and the investigation of electronic properties of the compounds show the influence of the attached substituents. All naphthyridine-based PACs show halochromic behaviour implying their use as highly sensitive proton sensor in non-protic solvents.

Association of Retinal Nerve Fiber Layer Thickness, an Index of Neurodegeneration, With Depressive Symptoms Over Time.

Project description:ImportanceWhether neurodegeneration contributes to the early pathobiology of late-life depression remains incompletely understood.ObjectiveTo investigate whether lower retinal nerve fiber layer (RNFL) thickness, a marker of neurodegeneration, is associated with the incidence of clinically relevant depressive symptoms and depressive symptoms over time.Design, setting, and participantsThis is a population-based cohort study from the Netherlands (The Maastricht Study) with baseline examination between 2010 and 2020 and median (IQR) follow-up of 5.0 (3.0-6.0) years. Participants were recruited from the general population. Individuals with type 2 diabetes were oversampled by design. Data analysis was performed from September 2020 to January 2021.ExposuresRNFL, an index of neurodegeneration, assessed with optical coherence tomography.Main outcomes and measuresDepressive symptoms were assessed with the Patient Health Questionnaire (PHQ)-9 (continuous score, 0-27) at baseline and over time via annual assessments. The presence of clinically relevant depressive symptoms was defined as a PHQ-9 score of 10 or higher.ResultsWe used data from 4934 participants with depressive symptoms over time (mean [SD] age, 59.7 [8.4] years; 2159 women [50.8%]; 870 had type 2 diabetes [20.5%]). Lower RNFL thickness was associated with higher incidence of clinically relevant depressive symptoms (per 1 SD, hazard ratio 1.11; 95% CI, 1.01-1.23) and more depressive symptoms over time (per 1 SD, rate ratio, 1.04; 95% CI, 1.01-1.06), after adjustment for demographic, cardiovascular, and lifestyle factors.Conclusions and relevanceThe findings of this study suggest that lower RNFL thickness is associated with higher incidence of clinically relevant depressive symptoms and more depressive symptoms over time. Hence, neurodegeneration may be associated with the early pathobiology of late-life depression.

A compact fiber-optic probe-based singlet oxygen luminescence detection system.

Project description:This paper presents a novel compact fiberoptic based singlet oxygen near-infrared luminescence probe coupled to an InGaAs/InP single photon avalanche diode (SPAD) detector. Patterned time gating of the single-photon detector is used to limit unwanted dark counts and eliminate the strong photosensitizer luminescence background. Singlet oxygen luminescence detection at 1270 nm is confirmed through spectral filtering and lifetime fitting for Rose Bengal in water, and Photofrin in methanol as model photosensitizers. The overall performance, measured by the signal-to-noise ratio, improves by a factor of 50 over a previous system that used a fiberoptic-coupled superconducting nanowire single-photon detector. The effect of adding light scattering to the photosensitizer is also examined as a first step towards applications in tissue in vivo.

The electrochemical-step asymmetry index.

Project description:The development of oxygen-evolution reaction (OER) electrocatalysts has been spurred by thermodynamic considerations on the free-energy landscape. Most commonly, electrocatalytic activity is approximated by the analysis of the free-energy changes among the mechanistic description, thereby taking only reaction steps with weak-binding adsorbates into account. Herein, a new method, denoted as the electrochemical-step asymmetry index (ESAI), is presented, which approximates electrocatalytic activity by penalizing both too strong as well as too weak bonding of intermediate states in order to mimic the well-known Sabatier principle.•The electrochemical-step asymmetry index (ESAI) is a descriptor to approximate electrocatalytic activity based on the analysis of the free-energy changes for a given mechanistic description, exemplified by the oxygen evolution reaction (OER).•The concept of the ESAI is based on the assumption that the optimum free-energy landscape has an asymmetric shape because this may factor overpotential and kinetic effects in the analysis, and the ESAI penalizes both too strong as well as too weak bonding of intermediate states to render a thorough representation of the Sabatier principle feasible.•The ESAI is a conceptual development of the earlier proposed electrochemical-step symmetry index (ESSI), which relies on a symmetric distribution of the free-energy changes as thermodynamic optimum and which takes only weak-binding adsorbates into account.

Comparison of retinal nerve fiber layer thickness measurement bias and imprecision across three spectral-domain optical coherence tomography devices.

Project description:PurposeWe compared retinal nerve fiber layer (RNFL) bias and imprecision among three spectral-domain optical coherence tomographs (SD-OCT).MethodsA total of 152 eyes of 83 subjects (96 healthy and 56 glaucomatous eyes) underwent peripapillary RNFL imaging using at least 2 of the following 3 SD-OCT devices on the same day: Cirrus HD-OCT (optic nerve head [ONH]) cube 200 × 200 protocol), RTVue-100 (ONH protocol [12 radial lines and 13 concentric circles]), and 3D OCT-1000 (3D Scan 256 × 256 protocol). Calibration equations, bias and imprecision of RNFL measurements were calculated using structural equation models.ResultsThe calibration equations for healthy and glaucoma RNFL thickness measurements among the 3 devices were: Cirrus = 2.136 + 0.831*RTVue; Cirrus = -15.521 + 1.056*3D OCT-1000; RTVue = -21.257 + 1.271*3D OCT-1000. Using Cirrus bias as an arbitrary reference, RTVue bias was 1.20 (95% CI 1.09-1.32, P < 0.05) times larger and 3D OCT-1000 was 0.95 (0.87-1.03, P > 0.05) times smaller. Relative to 3D OCT-1000, the RTVue bias was 1.27 (1.13-1.42, P < 0.05). RTVue imprecision (healthy eyes 7.83, 95% CI 6.43-9.58; glaucoma cases 5.71, 4.19-7.64) was statistically significantly higher than both Cirrus (healthy eyes 3.23, 2.11-4.31; glaucoma cases 3.53, 0.69-5.24) and 3D OCT-1000 (healthy eyes 4.07, 3.11-5.35; glaucoma cases 5.33, 3.77-7.67) in healthy eyes. The imprecision also was significantly higher for RTVue measurements in healthy compared to glaucomatous eyes. None of the other comparisons was statistically significant.ConclusionsRTVue-100 showed higher imprecision (or higher measurement variability) than Cirrus HD-OCT and 3D OCT-1000 RNFL measurements. Three-dimensional cube scanning with post-hoc data sampling may be a factor reducing imprecision.

Reusable building blocks in biological systems.

Project description:One of the most widely recognized features of biological systems is their modularity. The modules that constitute biological systems are said to be redeployed and combined across several conditions, thus acting as building blocks. In this work, we analyse to what extent are these building blocks reusable as compared with those found in randomized versions of a system. We develop a notion of decompositions of systems into phenotypic building blocks, which allows them to overlap while maximizing the number of times a building block is reused across several conditions. Different biological systems present building blocks whose reusability ranges from single use (e.g. condition specific) to constitutive, although their average reusability is not always higher than random equivalents of the system. These decompositions reveal a distinct distribution of building block sizes in real biological systems. This distribution stems, in part, from the peculiar usage pattern of the elements of biological systems, and constitutes a new angle to study the evolution of modularity.

The impact of compact layer in biphasic scaffold on osteochondral tissue engineering.

Project description:The structure of an osteochondral biphasic scaffold is required to mimic native tissue, which owns a calcified layer associated with mechanical and separation function. The two phases of biphasic scaffold should possess efficient integration to provide chondrocytes and osteocytes with an independent living environment. In this study, a novel biphasic scaffold composed of a bony phase, chondral phase and compact layer was developed. The compact layer-free biphasic scaffold taken as control group was also fabricated. The purpose of current study was to evaluate the impact of the compact layer in the biphasic scaffold. Bony and chondral phases were seeded with autogeneic osteoblast- or chondrocyte-induced bone marrow stromal cells (BMSCs), respectively. The biphasic scaffolds-cells constructs were then implanted into osteochondral defects of rabbits' knees, and the regenerated osteochondral tissue was evaluated at 3 and 6 months after surgery. Anti-tensile and anti-shear properties of the compact layer-containing biphasic scaffold were significantly higher than those of the compact layer-free biphasic scaffold in vitro. Furthermore, in vivo studies revealed superior macroscopic scores, glycosaminoglycan (GAG) and collagen content, micro tomograph imaging results, and histological properties of regenerated tissue in the compact layer-containing biphasic scaffold compared to the control group. These results indicated that the compact layer could significantly enhance the biomechanical properties of biphasic scaffold in vitro and regeneration of osteochondral tissue in vivo, and thus represented a promising approach to osteochondral tissue engineering.

Conformal geometry of the retinal nerve fiber layer.

Project description:The nerve fiber layer of the human retina is made up of the retinal segments of ganglion cell axons. Its geometry can be described mathematically as a fibration of a 2D domain: a partition of a certain region into smooth curves. Here, we present a simple family of curves that closely models the observed geometry of the nerve fiber layer. For each retina, the pattern depends on 2 parameters, A and B: A computer program determines A and B for a given retina and the theory matches the retina with a standard deviation of approximately 6-8 degrees . These particular curves turn out to be the curves that would be generated if the growing ganglion cell axon tip moved down a gradient toward a source of diffusible neuroattractant at the disk and away from a weaker macular diffusible repellant. Thus, this model provides morphological evidence that diffusible substances provide positional information to the embryonic ganglion cell axons in finding their way to the optic nerve head.
