Synthesis of an Alleged Byproduct Precursor in Iodixanol Preparation.

ABSTRACT: N ¹,N ³-Bis(2,3-dihydroxypropyl)-2,4,6-triiodo-5-(N-(oxiran-2-ylmethyl)acetamido)isophthalamide (1), the alleged precursor of several minor byproducts formed when the X-ray contrast agent iodixanol is synthesized from 5-acetamido-N ¹,N ³-bis(2,3-dihydroxypropyl)-2,4,6-triiodoisophthalamide (2), has been successfully prepared with an overall yield of 25%. Epoxide 1 enabled the confirmation of its presence in the reaction mixture during the preparation of iodixanol when amide 2 was used as the starting material.

Preparation of Crystalline LaFeO3 Nanoparticles at Low Calcination Temperature: Precursor and Synthesis Parameter Effects.

Project description:Substantial effort has been devoted to fabricating nanocrystalline lanthanum ferrite (LaFeO3), and calcination is the crucial process of crystallization in both high-temperature strategies and wet chemical methods. Lowering the calcination temperature gives the ability to resist the growth and agglomeration of nanoparticles, therefore contributing to preserve their unique nanostructures and properties. In this work, we prepared crystalline LaFeO3 nanoparticles with a calcination process at 500 °C, lower than the calcination temperature required in most wet chemistry methods. Correspondingly, the experimental conditions, including stoichiometric ratios, pH values, precipitants, complexant regent, and the calcination temperatures, were investigated. We found that the crystalline LaFeO3 was formed with crystalline NaFeO2 after calcination at 500 °C. Furthermore, the structure of FeO6 octahedra that formed in coprecipitation was associated with the process of crystallization, which was predominantly determined by calcination temperature. Moreover, an illusion of pure-phase LaFeO3 was observed when investigated by X-ray diffraction spectroscopy, which involves amorphous sodium ferrite or potassium ferrite, respectively. These findings can help prepare nanostructured perovskite oxides at low calcination temperatures.

Total Synthesis of the Alleged Structure of Crenarchaeol Enables Structure Revision*.

Project description:Crenarchaeol is a glycerol dialkyl glycerol tetraether lipid produced exclusively in Archaea of the phylum Thaumarchaeota. This membrane-spanning lipid is undoubtedly the structurally most sophisticated of all known archaeal lipids and an iconic molecule in organic geochemistry. The 66-membered macrocycle possesses a unique chemical structure featuring 22 mostly remote stereocenters, and a cyclohexane ring connected by a single bond to a cyclopentane ring. Herein we report the first total synthesis of the proposed structure of crenarchaeol. Comparison with natural crenarchaeol allowed us to propose a revised structure of crenarchaeol, wherein one of the 22 stereocenters is inverted.

One-pot Solvothermal Synthesis of Well-ordered Layered Sodium Aluminoalcoholate Complex: A Useful Precursor for the Preparation of Porous Al2O3 Particles.

Project description:One-pot solvothermal synthesis of a robust tetranuclear sodium hexakis(glycolato)tris(methanolato)aluminate complex Na3[Al4(OCH3)3(OCH2CH2O)6] via a modified yet rigorous base-catalyzed transesterification mechanism is presented here. Single crystal X-ray diffraction (SCXRD) studies indicate that this unique Al complex contains three penta-coordinate Al3+ ions, each bound to two bidentate ethylene glycolate chelators and one monodentate methanolate ligand. The remaining fourth Al3+ ion is octahedrally coordinated to one oxygen atom from each of the six surrounding glycolate chelators, effectively stitching the three penta-coordinate Al moieties together into a novel tetranuclear Al complex. This aluminate complex is periodically self-assembled into well-ordered layers normal to the [110] axis with the intra-/inter-layer bindings involving extensive ionic bonds from the three charge-counterbalancing Na+ cations rather than the more typical hydrogen bonding interactions as a result of the fewer free hydroxyl groups present in its structure. It can also serve as a valuable precursor toward the facile synthesis of high-surface-area alumina powders using a very efficient rapid pyrolysis technique.

Radiopaque scaffolds based on electrospun iodixanol/polycaprolactone fibrous composites.

Project description:Grafts based on biodegradable polymer scaffolds are increasingly used in tissue-engineering applications as they facilitate natural tissue regeneration. However, monitoring the position and integrity of these scaffolds over time is challenging due to radiolucency. In this study, we used an electrospinning method to fabricate biodegradable scaffolds based on polycaprolactone (PCL) and iodixanol, a clinical contrast agent. Scaffolds were implanted subcutaneously into C57BL/6 mice and monitored in vivo using longitudinal X-ray imaging and micro-computed tomography (CT). The addition of iodixanol altered the physicochemical properties of the PCL scaffold; notably, as the iodixanol concentration increased, the fiber diameter decreased. Radiopacity was achieved with corresponding signal enhancement as iodine concentration increased while exhibiting a steady time-dependent decrease of 0.96% per day in vivo. The electrospun scaffolds had similar performance with tissue culture-treated polystyrene in supporting the attachment, viability, and proliferation of human mesenchymal stem cells. Furthermore, implanted PCL-I scaffolds had more intense acute inflammatory infiltrate and thicker layers of maturing fibrous tissue. In conclusion, we developed radiopaque, biodegradable, biocompatible scaffolds whose position and integrity can be monitored noninvasively. The successful development of other imaging enhancers may further expand the use of biodegradable scaffolds in tissue engineering applications.

E. coli K5 fermentation and the preparation of heparosan, a bioengineered heparin precursor.

Project description:Heparosan is an acidic polysaccharide natural product, which serves as the critical precursor in heparin biosynthesis and in the chemoenzymatic synthesis of bioengineered heparin. Heparosan is also the capsular polysaccharide of Escherichia coli K5 strain. The current study was focused on the examination of the fermentation of E. coli K5 with the goal of producing heparosan in high yield and volumetric productivity. The structure and molecular weight properties of this bacterial heparosan were determined using polyacrylamide gel electrophoresis (PAGE) and Fourier transform mass spectrometry. Fermentation of E. coli K5 in a defined medium using exponential fed-batch glucose addition with oxygen enrichment afforded heparosan at 15?g/L having a number average molecular weight of 58,000?Da and a weight average molecular weight of 84,000?Da.

Superelectrophiles in Synthesis: Preparation of Aromatic Imides.

Project description:Aromatic carboxylic acids are found to undergo reactions with isocyanates, wherein triflic acid promotes the formation of aromatic imide products in fair to good yields. It is proposed that the carboxylic acid group directs the isocyanate electrophile to the ortho-position. This is thought to occur by the formation of a temporary carbamic acid anhydride group, which cleaves upon ortho-functionalization. A series of imide products are synthesized, and the synthesis of a potential selective inhibitor of tyrosyl DNA phosphodiesterase II is performed.

Glutamine: precursor or nitrogen donor for citrulline synthesis?

Project description:Although glutamine is considered the main precursor for citrulline synthesis, the current literature does not differentiate between the contribution of glutamine carbon skeleton vs. nonspecific nitrogen (i.e., ammonia) and carbon derived from glutamine oxidation. To elucidate the role of glutamine and nonspecific nitrogen in the synthesis of citrulline, l-[2-(15)N]- and l-[5-(15)N]glutamine and (15)N-ammonium acetate were infused intragastrically in mice. The amino group of glutamine labeled the three nitrogen groups of citrulline almost equally. The amido group and ammonium acetate labeled the ureido and amino groups of citrulline, but not the delta-nitrogen. D(5)-glutamine also infused in this arm of the study, which traces the carbon skeleton of glutamine, was utilized poorly, accounting for only 0.2-0.4% of the circulating citrulline. Dietary glutamine nitrogen (both N groups) incorporation was 25-fold higher than the incorporation of its carbon skeleton into citrulline. To investigate the relative contributions of the carbon skeleton and nonspecific carbon of glutamine, arginine, and proline to citrulline synthesis, U-(13)C(n) tracers of these amino acids were infused intragastrically. Dietary arginine was the main precursor for citrulline synthesis, accounting for approximately 40% of the circulating citrulline. Proline contribution was minor (3.4%), and glutamine was negligible (0.4%). However, the glutamine tracer resulted in a higher enrichment in the ureido group, indicating incorporation of nonspecific carbon from glutamine oxidation into carbamylphosphate used for citrulline synthesis. In conclusion, dietary glutamine is a poor carbon skeleton precursor for the synthesis of citrulline, although it contributes both nonspecific nitrogen and carbon to citrulline synthesis.

Isolation of senescent cells by iodixanol (OptiPrep) density gradient-based separation.

Project description:ObjectivesChemotherapeutic drugs induce senescence in cancer cells but, unlike replicative senescence or oncogene-induced senescence, do so rather inefficiently and depending on DNA damage. A thorough understanding of the biology of chemotherapy-induced senescent cells requires their isolation from a mixed population of adjacent senescent and non-senescent cancer cells.Materials and methodsWe have developed and optimized a rapid iodixanol (OptiPrep)-based gradient centrifugation system to identify, isolate and characterize doxorubicin (DXR)-induced senescent hepatocellular carcinoma (HCC) cells (HepG2 and Huh-7) in vitro.ResultsAfter cellular exposure to DXR, we used iodixanol gradient-based centrifugation to isolate and re-plate cells on collagen-coated flasks, despite their low or null proliferative capacity. The isolated cell populations were enriched for DXR-induced senescent HCC cells, as confirmed by proliferation arrest assay, and ?-galactosidase and DNA damage-dependent ?H2A.X staining.ConclusionsAnalysing pure cultures of chemotherapy-induced senescent versus non-responsive cancer cells will increase our knowledge on chemotherapeutic mechanisms of action, and help refine current therapeutic strategies.

Ultrafast shock synthesis of nanocarbon from a liquid precursor.

Project description:Carbon nanoallotropes are important nanomaterials with unusual properties and promising applications. High pressure synthesis has the potential to open new avenues for controlling and designing their physical and chemical characteristics for a broad range of uses but it remains little understood due to persistent conceptual and experimental challenges, in addition to fundamental physics and chemistry questions that are still unresolved after many decades. Here we demonstrate sub-nanosecond nanocarbon synthesis through the application of laser-induced shock-waves to a prototypical organic carbon-rich liquid precursor-liquid carbon monoxide. Overlapping large-scale molecular dynamics simulations capture the atomistic details of the nanoparticles' formation and evolution in a reactive environment and identify classical evaporation-condensation as the mechanism governing their growth on these time scales.
