ABSTRACT: The data provided in this paper are associated with the data in the «Binding of L-tryptophan and bovine serum albumin by novel gold nanoparticles capped with amphiphilic sulfonatomethylated calixresorcinarenes» paper (Shalaeva et al., 2019). Here, we represent i) the characterization data of calixresorcinarenes capped gold nanoparticles obtained by TEM and Vis- and IR spectroscopy; ii) the data giving the information about the interaction of modified AuNPs with L-tryptophan and bovine serum albumin by dynamic light scattering, spectrophotometry and fluorescence spectroscopy.

Formation and Stabilization of Gold Nanoparticles in Bovine Serum Albumin Solution.

Project description:The formation and growth of gold nanoparticles (AuNPs) were investigated in pH 7 buffer solution of bovine serum albumin (BSA) at room temperature. The processes were monitored by UV-Vis, circular dichroism, Raman and electron paramagnetic resonance (EPR) spectroscopies. TEM microscopy and dynamic light scattering (DLS) measurements were used to evidence changes in particle size during nanoparticle formation and growth. The formation of AuNPs at pH 7 in the absence of BSA was not observed, which proves that the albumin is involved in the first step of Au(III) reduction. Changes in the EPR spectral features of two spin probes, CAT16 and DIS3, with affinity for BSA and AuNPs, respectively, allowed us to monitor the particle growth and to demonstrate the protective role of BSA for AuNPs. The size of AuNPs formed in BSA solution increases slowly with time, resulting in nanoparticles of different morphologies, as revealed by TEM. Raman spectra of BSA indicate the interaction of albumin with AuNPs through sulfur-containing amino acid residues. This study shows that albumins act as both reducing agents and protective corona of AuNPs.

Elucidating the influence of gold nanoparticles on the binding of salvianolic acid B and rosmarinic acid to bovine serum albumin.

Project description:Salvianolic acid B and rosmarinic acid are two main water-soluble active ingredients from Salvia miltiorrhiza with important pharmacological activities and clinical applications. The interactions between salvianolic acid B (or rosmarinic acid) and bovine serum albumin (BSA) in the presence and absence of gold nanoparticles (Au NPs) with three different sizes were investigated by using biophysical methods for the first time. Experimental results proved that two components quenched the fluorescence of BSA mainly through a static mechanism irrespective of the absence or presence of Au NPs. The presence of Au NPs decreased the binding constants of salvianolic acid B with BSA from 27.82% to 10.08%, while Au NPs increased the affinities of rosmarinic acid for BSA from 0.4% to 14.32%. The conformational change of BSA in the presence of Au NPs (caused by a noncompetitive binding between Au NPs and drugs at different albumin sites) induced changeable affinity and binding distance between drugs and BSA compared with no Au NPs. The competitive experiments revealed that the site I (subdomain IIA) of BSA was the primary binding site for salvianolic acid B and rosmarinic acid. Additionally, two compounds may induce conformational and micro-environmental changes of BSA. The results would provide valuable binding information between salvianolic acid B (or rosmarinic acid) and BSA, and also indicated that the Au NPs could alter the interaction mechanism and binding capability of drugs to BSA, which might be beneficial to understanding the pharmacokinetics and biological activities of the two drugs.

Allosteric effects of gold nanoparticles on human serum albumin.

Project description:The ability of nanoparticles to alter protein structure and dynamics plays an important role in their medical and biological applications. We investigate allosteric effects of gold nanoparticles on human serum albumin protein using molecular simulations. The extent to which bound nanoparticles influence the structure and dynamics of residues distant from the binding site is analyzed. The root mean square deviation, root mean square fluctuation and variation in the secondary structure of individual residues on a human serum albumin protein are calculated for four protein-gold nanoparticle binding complexes. The complexes are identified in a brute-force search process using an implicit-solvent coarse-grained model for proteins and nanoparticles. They are then converted to atomic resolution and their structural and dynamic properties are investigated using explicit-solvent atomistic molecular dynamics simulations. The results show that even though the albumin protein remains in a folded structure, the presence of a gold nanoparticle can cause more than 50% of the residues to decrease their flexibility significantly, and approximately 10% of the residues to change their secondary structure. These affected residues are distributed on the whole protein, even on regions that are distant from the nanoparticle. We analyze the changes in structure and flexibility of amino acid residues on a variety of binding sites on albumin and confirm that nanoparticles could allosterically affect the ability of albumin to bind fatty acids, thyroxin and metals. Our simulations suggest that allosteric effects must be considered when designing and deploying nanoparticles in medical and biological applications that depend on protein-nanoparticle interactions.

In situ measurement of bovine serum albumin interaction with gold nanospheres.

Project description:We present in situ observations of adsorption of bovine serum albumin (BSA) on citrate-stabilized gold nanospheres. We implemented scattering correlation spectroscopy as a tool to quantify changes in the nanoparticle brownian motion resulting from BSA adsorption onto the nanoparticle surface. Protein binding was observed as an increase in the nanoparticle hydrodynamic radius. Our results indicate the formation of a protein monolayer at similar albumin concentrations as those found in human blood. Additionally, by monitoring the frequency and intensity of individual scattering events caused by single gold nanoparticles passing the observation volume, we found that BSA did not induce colloidal aggregation, a relevant result from the toxicological viewpoint. Moreover, to elucidate the thermodynamics of the gold nanoparticle-BSA association, we measured an adsorption isotherm which was best described by an anticooperative binding model. The number of binding sites based on this model was consistent with a BSA monolayer in its native state. In contrast, experiments using poly(ethylene glycol)-capped gold nanoparticles revealed no evidence for adsorption of BSA.

Mechanochemical Preparation, Characterization and Biological Activity of Stable CuS Nanosuspension Capped by Bovine Serum Albumin.

Project description:The biocompatible nanosuspension of CuS nanoparticles (NPs) using bovine serum albumin (BSA) as a capping agent was prepared using a two-stage mechanochemical approach. CuS NPs were firstly synthetized by a high-energy planetary ball milling in 15 min by milling elemental precursors. The stability of nanoparticles in the simulated body fluids was studied, revealing zero copper concentration in the leachates, except simulated lung fluid (SLF, 0.015%) and simulated gastric fluid (SGF, 0.078%). Albumin sorption on CuS NPs was studied in static and dynamic modes showing a higher kinetic rate for the dynamic mode. The equilibrium state of adsorption was reached after 90 min with an adsorption capacity of 86 mg/g compared to the static mode when the capacity 59 mg/g was reached after 2 h. Then, a wet stirred media milling in a solution of BSA was introduced to yield the CuS-BSA nanosuspension, being stable for more than 10 months, as confirmed by photon cross-correlation spectroscopy. The fluorescent properties of the nanosuspension were confirmed by photoluminescence spectroscopy, which also showed that tryptophan present in the BSA could be closer to the binding site of CuS than the tyrosine residue. The biological activity was determined by in vitro tests on selected cancer and non-tumor cell lines. The results have shown that the CuS-BSA nanosuspension inhibits the metabolic activity of the cells as well as decreases their viability upon photothermal ablation.

The Morphology Dependent Interaction between Silver Nanoparticles and Bovine Serum Albumin.

Project description:Biological applications of silver nanoparticles (AgNPs) depend on the covalently attached or adsorbed proteins. A series of biological effects of AgNPs within cells are determined by the size, shape, aspect ratio, surface charge, and modifiers. Herein, the morphology dependent interaction between AgNPs and protein was investigated. AgNPs with three different morphologies, such as silver nanospheres, silver nanorods, and silver nanotriangles, were employed to investigate the morphological effect on the interaction with a model protein: bovine serum albumin (BSA). The adsorptive interactions between BSA and the AgNPs were probed by UV-Vis spectroscopy, fluorescence spectroscopy, dynamic light scattering (DLS), Fourier transform infrared spectrometry (FTIR), transmission electron microscopy (TEM), and circular dichroism (CD) techniques. The results revealed that the particle size, shape, and dispersion of the three types of AgNPs markedly influence the interaction with BSA. Silver nanospheres and nanorods were capsulated by protein coronas, which led to slightly enlarged outer size. The silver nanotriangles evolved gradually into nanodisks in the presence of BSA. Fluorescence spectroscopy confirmed the static quenching the fluorescence emission of BSA by the three AgNPs. The FTIR and CD results suggested that the AgNPs with different morphologies had different effects on the secondary structure of BSA. The silver nanospheres and silver nanorods induced more pronounced structural changes than silver nanotriangles. These results suggest that the formation of a protein corona and the aggregation behaviors of AgNPs are markedly determined by their inherent morphologies.

Bovine serum albumin nanoparticles with fluorogenic near-IR-emitting squaraine dyes.

Project description:Two squaraine (SQ) dyes, N-propanesulfonate-benzothiazolium squaraine (SQ-1) and N-propanesulfonate-benzoindolium squaraine (SQ-2), were synthesized with sulfonate groups to increase water solubility. Both dyes are almost nonfluorescent in aqueous solution with fluorescent quantum yields of 0.03, but exhibited fluorescence enhancement after noncovalently binding with bovine serum albumin (BSA). Upon addition of BSA, the fluorescence intensity increased by ca. a factor of 10, along with a 10-fold extension in the fluorescence lifetime. SQ-1 and SQ-2 interacted with BSA efficiently and appeared to show a preference for binding at site II, which involves combinational effects of electrostatic and hydrophobic interactions. The fluorogenic squaraine dyes were then used to label BSA, forming BSA-based nanoparticles (NPs) through noncovalent binding. The resulting BSA-SQ NPs exhibited enhanced near-IR fluorescence and reduced aggregation of the squaraine moiety. The BSA-SQ NPs were used for cell incubation and bioimaging studies. Confocal fluorescent images were obtained for HCT 116 cells incubated with the BSA-SQ NPs and LysoSensor Green, demonstrating the utility of the NP probes for intracellular imaging. This strategy ovecomes the generally low fluorescence emission of SQ dyes in water and aggregation-reduced fluorescence, providing a versatile strategy for sensing and imaging in biological environments.

Effect of Gold Nanoparticles on the Conformation of Bovine Serum Albumin: Insights from CD Spectroscopic Analysis and Molecular Dynamics Simulations.

Project description:With the development of nanotechnology, there is growing interest in using nanoparticles (NPs) for biomedical applications, such as diagnostics, drug delivery, imaging, and nanomedicine. The protein's structural stability plays a pivotal role in its functionality, and any alteration in this structure can have significant implications, including disease progression. Herein, we performed a combined experimental and computational study of the effect of gold NPs with a diameter of 5 nm (5 nm Au-NPs) on the structural stability of bovine serum albumin (BSA) protein in the absence and presence of NaCl salt. Circular dichroism spectroscopy showed a loss in the secondary structure of BSA due to the synergistic effect of Au-NPs and NaCl, and Thioflavin T fluorescence assays showed suppressed β-sheet formation in the presence of Au-NPs in PBS, emphasizing the intricate interplay between NPs and physiological conditions. Additionally, molecular dynamics (MD) simulations revealed that 5 nm Au-NP induced changes in the secondary structure of the BSA monomer in the presence of NaCl, highlighting the initial binding mechanism between BSA and Au-NP. Furthermore, MD simulations explored the effect of smaller Au-NP (3 nm) and nanocluster (Au-NC with the size of 1 nm) on the binding sites of the BSA monomer. Although the formation of stable BSA-Au conjugates was revealed in the presence of NPs of different sizes, no specific protein binding sites were observed. Moreover, due to its small size, 1 nm Au-NC decreased helical content and hydrogen bonds in the BSA monomer, promoting protein unfolding more significantly. In summary, this combined experimental and computational study provides comprehensive insights into the interactions among Au nanosized substances, BSA, and physiological conditions that are essential for developing tailored nanomaterials with enhanced biocompatibility and efficacy.

Binding Constants of Substituted Benzoic Acids with Bovine Serum Albumin.

Project description:Experimental data on the affinity of various substances to albumin are essential for the development of empirical models to predict plasma binding of drug candidates. Binding of 24 substituted benzoic acid anions to bovine serum albumin was studied using spectrofluorimetric titration. The equilibrium constants of binding at 298 K were determined according to 1:1 complex formation model. The relationships between the ligand structure and albumin affinity are analyzed. The binding constant values for m- and p-monosubstituted acids show a good correlation with the Hammett constants of substituents. Two- and three-parameter quantitative structure-activity relationship (QSAR) models with theoretical molecular descriptors are able to satisfactorily describe the obtained values for the whole set of acids. It is shown that the electron-density distribution in the aromatic ring exerts crucial influence on the albumin affinity.
