Long-Wavelength Fluorescent Chemosensors for Hg²⁺ based on Pyrene.

ABSTRACT: A novel long-wavelength turn-on fluorescent chemosensor CS based on pyrene was synthesized to detect Hg²⁺. In the presence of other metal ions, CS could effectively recognize Hg²⁺ and produce the turn-on fluorescent emission at 607 nm. Also, the absorption spectrum exhibited red-shift. Meanwhile, the change of the solution color from yellow to orange was directly observed by the naked eye. The interaction between CS and Hg²⁺ was confirmed by the Job's plot, electrospray ionization mass spectrometry, scanning electron microscopy, and density functional theory calculations. It was found that the fluorescence of CS could be reversible when I^- was added into the solution of CS and Hg²⁺. CS illustrated high selectivity and good sensitivity for Hg²⁺ with the limit of detection of 36 nm. Moreover, CS could be utilized as test strips and silica gel plates to identify Hg²⁺.

Rhodamine-Based Fluorescent Probe for Highly Selective Determination of Hg2.

Project description:The determination of mercuric ions (Hg2+) in environmental and biological samples has attracted the attention of researchers lately. In the present work, a novel turn-on Hg2+ fluorescent probe utilizing a rhodamine derivative had been constructed and prepared. The probe could highly sensitively and selectively sense Hg2+. In the presence of excessive Hg2+, the probe displayed about 52-fold fluorescence enhancement in 50% H2O/CH3CH2OH (pH, 7.24). In the meantime, the colorless solution of the probe turned pink upon adding Hg2+. Upon adding mercuric ions, the probe interacted with Hg2+ and formed a 1:1 coordination complex, which had been the basis for recognizing Hg2+. The probe displayed reversible dual colorimetric and fluorescence sensing of Hg2+ because rhodamine's spirolactam ring opened upon adding Hg2+. The analytical performances of the probe for sensing Hg2+ were also studied. When the Hg2+ concentration was altered in the range of 8.0 × 10-8 to 1.0 × 10-5 mol L-1, the fluorescence intensity showed an excellent linear correlation with Hg2+ concentration. A detection limit of 3.0 × 10-8 mol L-1 had been achieved. Moreover, Hg2+ in the water environment and A549 cells could be successfully sensed by the proposed probe.

Pyrene-Based "Turn-Off" Probe with Broad Detection Range for Cu2+ , Pb2+ and Hg2+ Ions.

Project description:Detection of metals in different environments with high selectivity and specificity is one of the prerequisites of the fight against environmental pollution with these elements. Pyrenes are well suited for the fluorescence sensing in different media. The applied sensing principle typically relies on the formation of intra- and intermolecular excimers, which is however limiting the sensitivity range due to masking of e. g. quenching effects by the excimer emission. Herein we report a highly selective, structurally rigid chemical sensor based on the monomer fluorescence of pyrene moieties bearing triazole groups. This sensor can quantitatively detect Cu2+ , Pb2+ and Hg2+ in organic solvents over a broad concentrations range, even in the presence of ubiquitous ions such as Na+ , K+ , Ca2+ and Mg2+ . The strongly emissive sensor's fluorescence with a long lifetime of 165 ns is quenched by a 1 : 1 complex formation upon addition of metal ions in acetonitrile. Upon addition of a tenfold excess of the metal ion to the sensor, agglomerates with a diameter of about 3 nm are formed. Due to complex interactions in the system, conventional linear correlations are not observed for all concentrations. Therefore, a critical comparison between the conventional Job plot interpretation, the method of Benesi-Hildebrand, and a non-linear fit is presented. The reported system enables the specific and robust sensing of medically and environmentally relevant ions in the health-relevant nM range and could be used e. g. for the monitoring of the respective ions in waste streams.

A Highly Selective Fluorescent Probe for Hg2+ Based on a 1,8-Naphthalimide Derivative.

Project description:Hg2+ has a significant hazardous impact on the environment and ecosystem. There is a great demand for new methods with high selectivity and sensitivity to determine mercury in life systems and environments. In this paper, a novel turn-on Hg2+ fluorescent probe has been reported with a naphthalimide group. The Hg2+ fluorescent probe was designed by the inspiration of the well-known specific Hg2+-triggered thioacetal deprotection reaction. A 1,2-dithioalkyl group was chosen as the specific recognition site of Hg2+. The probe showed weak fluorescence without Hg2+, and the color of the solution was light yellow. In the presence of Hg2+, the probe reacted specifically with the mercury ion to produce an aldehyde and emitted strong fluorescence, and the color of the solution also turned light green, thus realizing the monitoring of the mercury ion. The Hg2+ fluorescent probe showed outstanding sensitivity and selectivity toward Hg2+. Furthermore, the Hg2+ fluorescent probe could work in a wide pH range. The linear relationship between the fluorescence intensity at 510 nm and the concentration of Hg2+ was obtained in a range of Hg2+ concentration from 2.5 × 10-7 to 1.0 × 10-5 M. The detection limit was found to be 4.0 × 10-8 M for Hg2+. Furthermore, with little cell toxicity, the probe was successfully applied to the confocal image of Hg2+ in PC-12 cells.

Anthracene-Based Highly Selective and Sensitive Fluorescent "Turn-on" Chemodosimeter for Hg2.

Project description:Three π-extended anthracene-bearing thioacetals (1-3) have been synthesized, and their fluorescence "turn-on" responses to Hg2+ ions are studied. The chemodosimetric fluorescence-sensing behavior and their resulting hydrolysis via a desulfurization reaction mechanism leads to the formation of highly fluorescent respective aldehyde substitutions. Furthermore, this mechanism was supported by increase in the quantum yields of their resulting aldehydes and is correlated to their molecular substitution. The chemosensors 1-3 have exhibited to be promising receptors toward Hg2+ ions in the presence of other competitive metal ions. Moreover, the detection limits of 1-3 have been found to be in the nanomolar range (94, 59, and 235, respectively). Fluorescence microscopic imaging studies show that 1-2 have been found to be effective for fluorescence imaging in live cells. Moreover, compounds 1-3 act as potential candidates for the detection of Hg2+ in environmental and biological systems as well as real samples.

Long-wavelength fluorescent reporters for monitoring protein kinase activity.

Project description:In?vivo optical imaging must contend with the limitations imposed by the optical window of tissue (600-1000?nm). Although a wide array of fluorophores are available that are visualized in the red and near-IR region of the spectrum, with the exception of proteases, there are few long wavelength probes for enzymes. This situation poses a particular challenge for studying the intracellular biochemistry of erythrocytes, the high hemoglobin content of which optically obscures subcellular monitoring at wavelengths less than 600?nm. To address this, tunable fluorescent reporters for protein kinase activity were developed. The probing wavelength is preprogrammed by using readily available fluorophores, thereby enabling detection within the optical window of tissue, specifically in the far-red and near-IR region. These agents were used to monitor endogenous cAMP-dependent protein kinase activity in erythrocyte lysates and in intact erythrocytes when using a light-activatable reporter.

Synthesis and Fluorescent Property Study of Novel 1,8-Naphthalimide-Based Chemosensors.

Project description:A series of novel mono- and di-substituted N-n-butyl-1,8-naphthalimide derivatives were synthesized simultaneously via a three-step reaction. The single crystal structure of N-n-butyl-4-[N',N'-bis(2',4'-dichlorobenzoyl)ethylamino]-1,8-naphthalimide (3f) was determined. The UV-vis and fluorescence properties of compound 3f were investigated. The 3f showed highly selective and sensitive fluorescence changes response towards Pb2+. A titration of monomer with Pb2+ ion was performed. When Pb2+ ion concentration increased from 0 to 10 eq., the fluorescent intensity of 3f decreased from 199.97 to 48.21. The pH effect on 3f showed that it is stable in a wide range of pH. The results indicated that 3f might be a probe molecule for Pb2+.

A chelating tetrazine-based ligand was employed to isolate an unprecedented tetranuclear Hg4 complex exhibiting HgI/HgII mixed valence, which also contains metal-metal bonds. Single-crystal X-ray diffraction, X-ray photoelectron spectroscopy, solid-state nuclear magnetic resonance, and theoretical approaches (density functional theory) were employed to shed some light on the structure and self-assembly of this discrete molecule.

Long Wavelength TCF-Based Fluorescent Probe for the Detection of Alkaline Phosphatase in Live Cells.

Project description:A long wavelength TCF-based fluorescent probe (TCF-ALP) was developed for the detection of alkaline phosphatase (ALP). ALP-mediated hydrolysis of the phosphate group of TCF-ALP resulted in a significant fluorescence "turn on" (58-fold), which was accompanied by a colorimetric response from yellow to purple. TCF-ALP was cell-permeable, which allowed it to be used to image ALP in HeLa cells. Upon addition of bone morphogenic protein 2, TCF-ALP proved capable of imaging endogenously stimulated ALP in myogenic murine C2C12 cells. Overall, TCF-ALP offers promise as an effective fluorescent/colorimetric probe for evaluating phosphatase activity in clinical assays or live cell systems.

Novel Benzothiazole-Based Highly Selective Ratiometric Fluorescent Turn-On Sensors for Zn2+ and Colorimetric Chemosensors for Zn2+, Cu2+, and Ni2+ Ions.

Project description:Metal ions play a very important role in environmental as well as biological fields. The detection of specific metal ions at a minute level caught much attention, and hence, several probes are available in the literature. Even though benzothiazole-based molecules have a special place in the medicinal field, only very few chemosensors are reported based on this moiety. The current work describes the design and synthesis of the benzothiazole-based chemosensor for a highly selective and sensitive detection of biologically important metal ions such as Zn2+, Cu2+, and Ni2+. The sensing studies of compound-1 showed a ratiometric as well as colorimetric response toward Zn2+, Cu2+, and Ni2+ ions and color changes from colorless to yellow and is found to be insensitive toward various metal ions (Cd2+, Cr3+, Mn2+, Pb2+, Ba2+, Al3+, Ca2+, Fe2+, Fe3+, Mg2+, K+, and Na+). Further, compound-1 exhibited ratiometric as well as turn-on-enhanced fluorescence response toward Zn2+ ions and turn off response for Cu2+ and Ni2+ ions. The Job plots revealed that the binding stoichiometry of compound-1 and metal ions is 2:1. The detection limits were found to be 0.25 ppm for Zn2+, while it was 0.30 ppm and 0.34 ppm for Ni2+ and Cu2+, respectively. In addition, density functional theory results strongly support the colorimetric response of metals, and the reversibility studies suggested that compound-1 can be used as a powerful chemosensor for the detection of Zn2+, Cu2+, and Ni2+ ions. The bioimaging data illustrated that compound-1 is a very effective ratiometric sensor for Zn2+ ions in live cells.

Rational design of fluorescent probe for Hg2+ by changing the chemical bond type.

Project description:Two kinds of fluorescent probes DFBT and DFABT, and their corresponding water-soluble compounds WDFBT and WDFABT, based on the trimers containing a benzo[2,1,3]thiadiazole moiety and two fluorene moieties are synthesized. Their luminescent behavior towards Hg2+ ions and other various metal ions in organic and water solutions are studied in detail via absorption and emission spectroscopy. All these probes show a selective "on-off-type" fluorescent response to Hg2+ ions in solution over other metal ions with a maximum detection limit of 10-7 M. Importantly, the probe type can be changed from irreversible to reversible by altering the bridge mode between the functional units from C[triple bond, length as m-dash]C triple bond to C-C single bond. Their detection mechanisms towards Hg2+ are studied in detail via mass spectrometry and Job plots, which are attributed to irreversible chemical reaction for DFABT and WDFABT and a reversible coordination reaction for DFBT and WDFBT respectively. Our research results about this kind of organic fluorescent probe provide valuable information to the future design of practical Hg2+ fluorescent probes.
