A new species of Mountain Dragon (Reptilia: Agamidae: Diploderma) from the D. dymondi complex in southern Sichuan Province, China.

ABSTRACT: Mountain Dragons of the genus Diploderma Hallowell, 1861 were recently resurrected from the paraphyletic genus Japalura sensu lato (Wang et al., 2019a). Despite the recent split, Diploderma still represents one of the most diverse groups of agamid lizard from Asia, including 25 species recognized currently, with most species found in China ((Wang et al., 2019a, 2019b). Although increasing attention has been paid to cryptic diversity within the genus in Southwest China during the past decade, most studies have focused on a single species complex, D. flaviceps, only (Manthey et al., 2012; Wang et al., 2015, 2016, 2017, 2019a), with few studies on other congeners that also have widespread distributions. One such example is D. dymondi (Boulenger, 1906).

Discovery of Japalura chapaensis Bourret, 1937 (Reptilia: Squamata: Agamidae) from Southeast Yunnan Province, China.

Project description:Due to a paucity of surveys in northern Indochina and lack of international collaborations among neighboring countries, recognized distributional ranges for many amphibian and reptile species end at the political borders for some countries, despite seemingly continuous suitable habitat spanning the region. Combining both morphological and genetic data, we report the first discovery of Japalura chapaensis, a rare agamid lizard believed previously to be endemic to northern Vietnam only, along the border region of southeastern Yunnan Province, China. To facilitate future research on the genus Japalura sensu lato in Indochina, we provide detailed descriptions of additional specimens of this rare species, including the first description of coloration in life and an expanded diagnosis, and discuss the species boundary of J. chapaensis with respect to its congeners.

Characterization of the complete mitochondrial genome and phylogenetic status of a recently described species of Mountain Dragon, Diploderma vela (Reptilia: Squamata: Agamidae), from the upper Lantsang valley in west China.

Project description:The mountain dragon, Diploderma vela, is an endemic and protected valley lizard that inhabits the upper Lantsang Valley in West China. In this study, we sequenced the complete mitochondrial genome of a male individuals of D. vela using next-generation sequencing methodologies. The complete mitogenome is 16,432 bp in length and contains one noncoding control regions, 13 protein-coding, 22 transfer RNA and two ribosomal RNA genes. The mitogenome content and structure of D. vela was consistent with the previously published representatives of the family. A Bayesian phylogenetic analysis using the complete mitochondrial genomes of Agamidae fully resolved D. vela in the Draconinae, a result consistent with previous investigations. This study provides bioinformatic data for better understanding the evolution and the phylogenetic history of the mountain dragon.

Blood cells and hematological parameters of the mountain dragon, Diploderma micangshanensis (Squamata: Lacertilia: Agamidae).

Project description:Hematological characteristics reflect the health status of animals and their physiological adaptation to the environment. However, few studies focused on the species of Diploderma. In this study, the blood cells and the hematological parameters of Diploderma micangshanensis, a species endemic to China, were examined based on 48 healthy adult (32 males and 16 females). The blood cells and hematological parameters of D. micangshanensis were similar to those of other lizard species. Although the values of erythrocyte morphometric characters and hematological parameters varied between males and females, the differences were only significant in the case of the hematocrit and erythrocyte size, which may allow for higher oxygen availability in males. Hemoglobin, mean corpuscular hemoglobin concentration, and mean corpuscle volume were strongly affected by the snout-vent length and/or body mass, which reflect the physiological adaptation to the oxygen requirement of different individuals. This is the first report of hematological data from a species of Diploderma, and the results will provide data for research on the adaptive evolution and health assessment in this species and other congeners.

A new species of the toothed toad Oreolalax (Anura, Megophryidae) from Sichuan Province, China.

Project description:The toad genus Oreolalax is widely distributed in southwest China and northern Vietnam. A new species of the genus is described from Sichuan Province, China. Phylogenetic analyses based on the mitochondrial 12S rRNA and 16S rRNA gene sequences supported the new species as an independent clade clustered into the clade also containing O. nanjiangensis and O. chuanbeiensis. The new species can be distinguished from its congeners by a combination of the following characters: body size moderate (SVL 51.2-64.2 mm in males); head broad; tympanum hidden; interorbital region with dark triangular pattern; belly with marbling; male lacking spines on lip margin; spiny patches on chest small with thick sparse spines in male; nuptial spines thick and sparse; tibio-tarsal articulation reaching beyond nostril when leg stretched forward; toe webbing at base.

A new species of Scutellaria (Scutellarioideae, Lamiaceae) from Sichuan Province in southwest China.

Project description:Scutellaria wuana, a new species discovered from a xeric valley in Muli County of Sichuan Province in southwest China, is described and illustrated. Morphologically, the new species shares similarities with S. mairei, but can be readily distinguished by a suite of morphological characters including a white-pubescent erect stem, conspicuous leaf petioles, and a yellow corolla with a trapeziform lower-middle lip lobe. The habitat and distribution of S. wuana are also distinctive. The position of the new species within Scutellaria is examined in a phylogenetic context using the nuclear ribosomal internal and external transcribed spacers. Additionally, we examine leaf epidermal and pollen grain micromorphology of the new species and putative relatives.

Influenza D Virus in Animal Species in Guangdong Province, Southern China.

Project description:Molecular tests revealed influenza D viruses of D/OK lineage widely circulating in farmed animal species in Guangdong Province, southern China. In particular, we found high levels of influenza D virus infection in goats and pigs. We also detected viral RNA in serum specimens and feces of animals with certain severe diseases.

Complete mitochondrial genome of Laudakia sacra (Reptilia: Agamidae).

Project description:The complete mitochondrial genome (mitogenome) sequence of Laudakia sacra was determined by using a PCR-based method. The total length of mitogenome is 16,555?bp, and contains 13 typical vertebrate protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes and 2 control regions. Only ND6 gene and 8 tRNA genes on the L-strand other than are encoded on the H-strand. The phylogenetic tree of L. sacra and 13 other species were built. The DNA data present here will facilitate future taxonomic work of the genus Laudakia.

Molecular detection of Porcine astrovirus in Sichuan Province, China.

Project description:Porcine astrovirus (PoAstV) is widely distributed worldwide, and is highly prevalent among piglets with or without diarrhea, existing as at least five distinct lineages (PoAstV1-PoAstV5) within the genus Mamastrovirus. However, our knowledge of the diversity and epidemiology of PoAstV in China is limited.In this study, fecal samples from 21/120 (17.5 %) domestic pigs, including 18/100 (18 %) diarrheic and 3/20 (15 %) healthy pigs, and from 1/9 (11.1 %) healthy wild boars tested in Sichuan Province were positive for PoAstV on reverse transcription-PCR. Of the 22 positive samples, 13.6 % were positive for PoAstV only, whereas 40.9 % also contained Porcine epidemic diarrhea virus (PEDV), 22.7 % also contained porcine group A rotavirus (PRoVA), and 22.7 % also contained PEDV and PRoVA. A phylogenetic analysis of the RdRp gene revealed genetic heterogeneity among the PoAstV sequences and two lineages were detected in this study, with PoAstV-2 predominant. PoAstV-5 was detected in wild boars for the first time.PoAstV infections exist in Sichuan Province regardless of the disease status in the pig population, either alone or in combination with other enteric viruses, and may be associated with diarrhea.

Spatial and temporal patterns of amphibian species richness on Tianping Mountain, Hunan Province, China.

Project description:Exploring species richness patterns across space and time can help in understanding species distribution and in formulating conservation strategies. Among taxa, amphibians are of utmost importance as they are highly sensitive to environmental changes due to their unique life histories (Zhong et al., 2018). Here, we investigated the spatial and temporal patterns of amphibian species richness on Tianping Mountain in China. Specifically, we established 10 transects at low to high elevations, and sampled amphibians in April, June, August, and October 2017. Our results demonstrated that amphibian species composition and richness varied significantly at both spatial and temporal scales and were associated with gradients of environmental change in microhabitats on Tianping Mountain.

Viral communities associated with porcine respiratory disease complex in intensive commercial farms in Sichuan province, China.

Project description:Porcine respiratory disease complex (PRDC), a common piglet disease, causes substantive economic losses in pig farming. To investigate the viral diversity associated with PRDC, the viral communities in serum and nasal swabs from 26 PRDC-affected piglets were investigated using metagenomics. By deep sequencing and de novo assembly, 17 viruses were identified in two pooled libraries (16 viruses from serum, nine from nasal swabs). Porcine circovirus (PCV)-2, porcine reproductive and respiratory syndrome virus (PRRSV) and pseudorabies virus, all commonly associated with PRDC, were identified in the two pooled samples by metagenomics, but most viruses comprised small linear and circular DNAs (e.g. parvoviruses, bocaviruses and circoviruses). PCR was used to compare the detection rates of each virus in the serum samples from 36 PRDC-affected piglets versus 38 location-matched clinically healthy controls. The average virus category per sample was 6.81 for the PRDC-affected piglets and 4.09 for the controls. Single or co-infections with PCV-2 or PRRSV had very high detection rates in the PRDC-affected piglets. Interestingly, porcine parvovirus (PPV)-2, PPV-3, PPV-6 and torque teno sus virus 1a were significantly associated with PRDC. These results illustrate the complexity of viral communities in the PRDC-affected piglets and highlight the candidate viruses associated with it.
