Ratiometric Polymer Probe for Detection of Peroxynitrite and the Application for Live-Cell Imaging.

ABSTRACT: Peroxynitrite (ONOO-) is one of the sources of oxidation stress involved in many biological signaling pathways. The role of ONOO- being a double-edged sword in biological systems drives the development of effective detection tools. In this work, a boronate-based polymeric fluorescent probe PB-PVA was synthesized and the probe performance was evaluated. The probe exhibits ratiometric sensing of ONOO- in a range of 0-6 µM. There is good linear relationship between the probe fluorescence intensity ratio and ONOO- concentration. The probe also displays moderate selectivity towards ONOO- over other ROS. Moreover, it is water-soluble and possesses good biocompatibility which aids the imaging of ONOO- in living cells. These properties could make the probe a promising tool in in vitro study related to ONOO-.

A Benzothiazole-Based Fluorescent Probe for Ratiometric Detection of Al3+ and Its Application in Water Samples and Cell Imaging.

Project description:An easily prepared benzothiazole-based probe (BHM) was prepared and characterized by general spectra, including 1H NMR, 13C NMR, HRMS, and single-crystal X-ray diffraction. Based on the synergistic mechanism of the inhabitation of intramolecular charge transfer (ICT), the BHM displayed high selectivity and sensitivity for Al3+ in DMF/H2O (v/v, 1/1) through an obvious blue-shift in the fluorescent spectrum and significant color change detected by the naked eye, respectively. The binding ratio of BHM with Al3+ was 1:1, as determined by the Job plot, and the binding details were investigated using FT-IR, 1H NMR titration, and ESI-MS analysis. Furthermore, the BHM was successfully applied in the detection of Al3+ in the Songhua River and on a test stripe. Fluorescence imaging experiments confirmed that the BHM could be used to monitor Al3+ in human stromal cells (HSC).

Application of a Colorimetric and Near-Infrared Fluorescent Probe in Peroxynitrite Detection and Imaging in Living Cells.

Project description:Peroxynitrite (ONOO-) plays a vital role in pathological and physiological processes, and an excessive amount of ONOO- causes various diseases. Developing a specific and sensitive method for the detection of ONOO- in biological systems is significant. Herein, we reported a novel colorimetric and near-infrared fluorescent probe (pyridin-4-ylmethyl (Z)-2-cyano-2-(3-((E)-4-hydroxystyryl)-5,5-dimethylcyclohex-2-en-1-ylidene)acetate diphenyl phosphinate group (AN-DP)) based on isophorone and phosphinate groups for ONOO- detection. The probe displayed excellent selectivity toward ONOO- compared with other relevant analytes. It showed a good linear relationship between the fluorescence intensity at 670 nm and ONOO- concentration (0-10 μM) with a low detection limit (53 nM). Importantly, the probe was a colorimetric and near-infrared fluorescent probe suitable for ONOO- detection. Furthermore, the probe could be used for imaging ONOO- in HepG2 cells.

Red-Emission Probe for Ratiometric Fluorescent Detection of Bisulfite and Its Application in Live Animals and Food Samples.

Project description:Key roles of bisulfite (HSO3 -) in food quality assurance and human health necessitate a reliable analytical method for rapid, sensitive, and selective detection of HSO3 -. Herein, a new red-emitting ratiometric fluorescence probe, BIQ, is reported for sensitive and selective detection of HSO3 - in food samples and live animals. Probe BIQ recognizes HSO3 - via a 1,4-nucleophilic addition reaction. As a result of this specific reaction, emission intensities at 625 and 475 nm are dramatically changed, allowing the detection of HSO3 - in a ratiometric fluorescence model in an aqueous solution. The obvious changes of solution color from pink to transparent and fluorescence color from rose-red to cyan allow the detection of HSO3 - by naked eyes. Furthermore, probe BIQ has fast response in color and fluorescence (<2 min), excellent selectivity, and a low detection limit (0.29 μM), which enables its application in HSO3 - detection in food samples and live organisms. The practical applications of probe BIQ are then demonstrated by the visualization of HSO3 - in live animals (zebrafish and nude mouse) as well as the determination of HSO3 - in white wine and sugar.

Highly Selective Fluorescent Probe for the Detection of Copper (II) and Its Application in Live Cell Imaging.

Project description:The development of fluorescent methods for the detection of metal ions is of great importance due to their diverse environmental and biological roles. Herein, a rhodamine 6G-based off-on fluorescent probe (L1) with a t-butyl pyrrole moiety as the recognition site was designed and synthesized. Photophysical studies show that L1 exhibits excellent sensitivity and selectivity towards Cu2+ to other metal ions in neutral acetonitrile aqueous media. Mechanism studies suggest that the recognition process may associate with a Cu2+ promoted hydrolysis reaction of L1. Furthermore, L1 has been successfully applied in fluorescence imaging of Cu2+ ion in living cells.

Live-Cell imaging and measurement of intracellular pH in filamentous fungi using a genetically encoded ratiometric probe.

Project description:A novel, genetically encoded, ratiometric pH probe (RaVC) was constructed to image and measure intracellular pH in living hyphae of Aspergillus niger. RaVC is a chimeric protein based on the pH-sensitive probe pHluorin, which was partially codon optimized for expression in Aspergillus. Intracellular pH imaging and measurement was performed by simultaneous, dual-excitation confocal ratio imaging. The mean cytoplasmic pH measured was 7.4 to 7.7 based on calibrating RaVC in situ within nigericin-treated hyphae. Pronounced, longitudinal cytoplasmic pH gradients were not observed in the apical 20 microm of actively growing hyphae at the periphery of 18-h-old colonies. The cytoplasmic pH remained unchanged after prolonged growth in buffered medium with pH values between 2.5 or 9.5. Sudden changes in external pH significantly changed cytoplasmic pH by <1.3 pH units, but it returned to its original value within 20 min following treatment. The weak acid and antifungal food preservative sorbic acid caused prolonged, concentration-dependent intracellular acidification. The inhibition of ATPases with N-ethylmaleimide, dicychlohexylcarbodimide, or sodium azide caused the cytoplasmic pH to decrease by <1 pH unit. Treatment with the protonophore carbonyl cyanide m-chlorophenylhydrazone or cyanide p-(trifluoromethoxy) phenylhydrazone reduced the cytoplasmic pH by <1 pH unit. In older hyphae from 32-h-old cultures, RaVC became sequestered within large vacuoles, which were shown to have pH values between 6.2 and 6.5. Overall, our study demonstrates that RaVC is an excellent probe for visualizing and quantifying intracellular pH in living fungal hyphae.

A ratiometric Raman probe for live-cell imaging of hydrogen sulfide in mitochondria by stimulated Raman scattering.

Project description:Stimulated Raman Scattering (SRS) coupled with alkyne tags has been an emerging imaging technique to visualize small-molecule species with high sensitivity and specificity. Here we describe the development of a ratiometric Raman probe for visualizing hydrogen sulfide (H2S) species in living cells as the first alkyne-based sensor for SRS microscopy. This probe uses an azide unit as a selective reactive site, and it targets mitochondria with high specificity. The SRS ratiometric images show a strong response to H2S level changes in living cells.

Quantitative imaging of glutathione in live cells using a reversible reaction-based ratiometric fluorescent probe.

Project description:Glutathione (GSH) plays an important role in maintaining redox homeostasis inside cells. Currently, there are no methods available to quantitatively assess the GSH concentration in live cells. Live cell fluorescence imaging revolutionized the field of cell biology and has become an indispensable tool in current biological studies. In order to minimize the disturbance to the biological system in live cell imaging, the probe concentration needs to be significantly lower than the analyte concentration. Because of this, any irreversible reaction-based GSH probe can only provide qualitative results within a short reaction time and will exhibit maximum response regardless of the GSH concentration if the reaction is completed. A reversible reaction-based probe with an appropriate equilibrium constant allows measurement of an analyte at much higher concentrations and, thus, is a prerequisite for GSH quantification inside cells. In this contribution, we report the first fluorescent probe-ThiolQuant Green (TQ Green)-for quantitative imaging of GSH in live cells. Due to the reversible nature of the reaction between the probe and GSH, we are able to quantify mM concentrations of GSH with TQ Green concentrations as low as 20 nM. Furthermore, the GSH concentrations measured using TQ Green in 3T3-L1, HeLa, HepG2, PANC-1, and PANC-28 cells are reproducible and well correlated with the values obtained from cell lysates. TQ Green imaging can also resolve the changes in GSH concentration in PANC-1 cells upon diethylmaleate (DEM) treatment. In addition, TQ Green can be conveniently applied in fluorescence activated cell sorting (FACS) to measure GSH level changes. Through this study, we not only demonstrate the importance of reaction reversibility in designing quantitative reaction-based fluorescent probes but also provide a practical tool to facilitate redox biology studies.

A rationally designed rhodamine-based fluorescent probe for molecular imaging of peroxynitrite in live cells and tissues.

Project description:Herein, we present the rational design, synthesis, characterization, and biological applications of a new rhodamine-based fluorescent probe, HKYellow, for the detection and molecular imaging of peroxynitrite, an important highly reactive oxidant involved in a variety of physiological and pathological processes. HKYellow was rationally designed on the basis of the efficient fluorescence quenching effect of the N-phenyl group to the rhodamine core and a peroxynitrite-triggered N-dearylation reaction to achieve a sensitive and selective fluorescence turn-on response toward peroxynitrite in chemical and biological levels. This probe has been thoroughly evaluated for the robust imaging of peroxynitrite in live cells and tissues. By utilizing HKYellow, we provide the first visual evidence that peroxynitrite is generated in mouse liver tissues under an acute alcohol binge or ischemic-reperfusion condition. This probe should be a powerful molecular imaging tool for interrogating the complex biological roles of peroxynitrite under various biological settings.

A Novel Ratiometric Fluorescent Probe for Mercury (II) ions and Application in Bio-imaging.

Project description:Since the accumulation of mercury (II) ions in the environment and ecosystem causes serious problems to environment and disease, the recognition of Hg2+ ions and its bio-imaging is of high importance. In sight of the advantages of fluorescence probes, a new probe (PMH) was facilely synthesized by incorporating phenylimidazole fluorophore and 3-methyl-2- benzothiazolinone hydrazone hydrochloride monohydrate. The PMH probe exhibited a ratiometric response for Hg2+ ions with fluorescence intensity increasing at 520 nm and decreasing at 445 nm simultaneously. The PMH probe interacted with Hg2+ ions in seconds with high optical stability and showed good selectivity over other metal ions. In addition, the probe has excellent biocompatibility and imaging performance in cells and zebrafish.
