High-pressure synthesis and crystal structure of SrGa4As4.

ABSTRACT: Strontium tetra-gallate(II,III) tetra-arsenide, SrGa4As4, was synthesized in a Walker-type multianvil apparatus under high-pressure/high-temperature conditions of 8?GPa and 1573?K. The com-pound crystallizes in a new structure type (P3221, Z = 3) as a three-dimensional (3D) framework of corner-sharing SrAs8 quadratic anti-prisms with strontium situated on a twofold rotation axis (Wyckoff position 3b). This arrangement is surrounded by a 3D framework which can be described as alternately stacked layers of either condensed GaIIIAs4 tetra-hedra or honeycomb-like layers built up from distorted ethane-like GaII 2As6 units com-prising Ga-Ga bonds.

Crystal structure of SrGeO3 in the high-pressure perovskite-type phase.

Project description:Single crystals of the SrGeO3 (strontium germanium trioxide) high-pressure phase have been synthesized successfully at 6?GPa and 1223?K. The compound crystallizes with the ideal cubic perovskite-type structure (space group Pm-3m), which consists of a network of corner-linked regular GeO6 octa-hedra (point-group symmetry m-3m), with the larger Sr atoms located at the centers of cavities in the form of SrO12 cubocta-hedra (point-group symmetry m-3m) in the network. The degrees of covalencies included in the Sr-O and the Ge-O bonds calculated from bond valences are 20.4 and 48.9%, respectively. Thus, the Ge-O bond of the GeO6 octa-hedron in the SrGeO3 perovskite has a strong covalency, comparable to those of the Si-O bonds of the SiO4 tetra-hedra in silicates with about 50% covalency. The thermal vibrations of the O atoms in the title compound are remarkably suppressed in the directions of the Ge-O bonds. This anisotropy ranks among the largest observed in stoichiometric cubic perovskites.

Combined crystal structure prediction and high-pressure crystallization in rational pharmaceutical polymorph screening.

Project description:Organic molecules, such as pharmaceuticals, agro-chemicals and pigments, frequently form several crystal polymorphs with different physicochemical properties. Finding polymorphs has long been a purely experimental game of trial-and-error. Here we utilize in silico polymorph screening in combination with rationally planned crystallization experiments to study the polymorphism of the pharmaceutical compound Dalcetrapib, with 10 torsional degrees of freedom one of the most flexible molecules ever studied computationally. The experimental crystal polymorphs are found at the bottom of the calculated lattice energy landscape, and two predicted structures are identified as candidates for a missing, thermodynamically more stable polymorph. Pressure-dependent stability calculations suggested high pressure as a means to bring these polymorphs into existence. Subsequently, one of them could indeed be crystallized in the 0.02 to 0.50 GPa pressure range and was found to be metastable at ambient pressure, effectively derisking the appearance of a more stable polymorph during late-stage development of Dalcetrapib.

High-resolution X-ray crystal structure of bovine H-protein using the high-pressure cryocooling method.

Project description:Recently, many technical improvements in macromolecular X-ray crystallography have increased the number of structures deposited in the Protein Data Bank and improved the resolution limit of protein structures. Almost all high-resolution structures have been determined using a synchrotron radiation source in conjunction with cryocooling techniques, which are required in order to minimize radiation damage. However, optimization of cryoprotectant conditions is a time-consuming and difficult step. To overcome this problem, the high-pressure cryocooling method was developed (Kim et al., 2005) and successfully applied to many protein-structure analyses. In this report, using the high-pressure cryocooling method, the X-ray crystal structure of bovine H-protein was determined at 0.86 Å resolution. Structural comparisons between high- and ambient-pressure cryocooled crystals at ultra-high resolution illustrate the versatility of this technique. This is the first ultra-high-resolution X-ray structure obtained using the high-pressure cryocooling method.

The first crystal structure of a macromolecular assembly under high pressure: CpMV at 330 MPa.

Project description:The structure of cubic Cowpea mosaic virus crystals, compressed at 330 MPa in a diamond anvil cell, was refined at 2.8 A from data collected using ultrashort-wavelength (0.331 A) synchrotron radiation. With respect to the structure at atmospheric pressure, order is increased with lower Debye Waller factors and a larger number of ordered water molecules. Hydrogen-bond lengths are on average shorter and the cavity volume is strongly reduced. A tentative mechanistic explanation is given for the coexistence of disordered and ordered cubic crystals in crystallization drops and for the disorder-order transition observed in disordered crystals submitted to high pressure. Based on such explanation, it can be concluded that pressure would in general improve, albeit to a variable extent, the order in macromolecular crystals.

High-pressure protein crystal structure analysis of Escherichia coli dihydrofolate reductase complexed with folate and NADP.

Project description:A high-pressure crystallographic study was conducted on Escherichia coli dihydrofolate reductase (ecDHFR) complexed with folate and NADP+ in crystal forms containing both the open and closed conformations of the M20 loop under high-pressure conditions of up to 800?MPa. At pressures between 270 and 500?MPa the crystal form containing the open conformation exhibited a phase transition from P21 to C2. Several structural changes in ecDHFR were observed at high pressure that were also accompanied by structural changes in the NADP+ cofactor and the hydration structure. In the crystal form with the closed conformation the M20 loop moved as the pressure changed, with accompanying conformational changes around the active site, including NADP+ and folate. These movements were consistent with the suggested hypothesis that movement of the M20 loop was necessary for ecDHFR to catalyze the reaction. In the crystal form with the open conformation the nicotinamide ring of the NADP+ cofactor undergoes a large flip as an intermediate step in the reaction, despite being in a crystalline state. Furthermore, observation of the water molecules between Arg57 and folate elucidated an early step in the substrate-binding pathway. These results demonstrate the possibility of using high-pressure protein crystallography as a method to capture high-energy substates or transient structures related to the protein reaction cycle.

RbZnFe(PO4)2: synthesis and crystal structure.

Project description:A new iron phosphate, rubidium zinc iron(III) phosphate, RbZnFe(PO4)2, has been synthesized as single crystals by the flux method. This compound is isostructural to the previously reported KCoAl(PO4)2 [Chen et al. (1997 ?). Acta Cryst. C53,1754-1756]. Its structure consists of a three-dimensional framework built up from corner-sharing PO4 and (Zn,Fe)O4 tetra-hedra. This mode of linkage forms channels parallel to the [100], [010] and [001] directions in which the Rb(+) ions are located.

Synthesis and Crystal Structure of Gold Nanobelts.

Project description:Gold nanobelts were synthesized by the reduction of tetrachloroauric acid with ascorbic acid in the presence of the surfactants cetyltrimethylammonium bromide and sodium dodecylsulfate. The resulting structures have rectangular cross sectional dimensions that are tens of nanometers and lengths that are tens to hundreds of micrometers. We find that the nanobelt yield and resulting structures are very sensitive to temperature which is likely due to the transition of the surfactant solution from wormlike micelles to spherical micelles. The nanobelt crystal structure contains a mixture of face centered cubic and hexagonally close packed lattice phases that can be isolated and examined individually due to the unique nanobelt size and shape.

A Sr2+-metal-organic framework with high chemical stability: synthesis, crystal structure and photoluminescence property.

Project description:Metal-organic frameworks (MOFs) are typically built by assembly of metal centres and organic linkers, and have emerged as promising crystalline materials in a variety of fields. However, the stability of MOFs is a key limitation for their practical applications. Herein, we report a novel Sr 2+: -MOF [Sr4(Tdada)2(H2O)3(DMF)2] (denoted as NKU- 105: , NKU?=?Nankai University; H4Tdada?=?5,5'-((thiophene-2,5-dicar bonyl)bis(azanediyl))diisophthalic acid; DMF?=?N,N-dimethylformamide) featuring an open square channel of about 6?Å along the c-axis. Notably, NKU- 105: exhibits much outstanding chemical stability against common organic solvents, boiling water, acids and bases, relative to most MOF materials. Furthermore, NKU- 105: is an environment-friendly luminescent material with a bright cyan emission.This article is part of the themed issue 'Coordination polymers and metal-organic frameworks: materials by design'.

Synthesis and crystal structure of NaMgFe(MoO4)3.

Project description:The iron molybdate NaMgFe(MoO4)3 {sodium magnesium iron(III) tris-[molybdate(VI)]} has been synthesized by the flux method. This compound is isostructural with ?-NaFe2(MoO4)3 and crystallizes in the triclinic space group P-1. Its structure is built up from [Mg,Fe]2O10 units of edge-sharing [Mg,Fe]O6 octa-hedra which are linked to each other through the common corners of [MoO4] tetra-hedra. The resulting anionic three-dimensional framework leads to the formation of channels along the [101] direction in which the Na(+) cations are located.

Synthesis and crystal structure of ABW-type SrFe1.40V0.60O4.

Project description:Single crystals of SrFe1.40V0.60O4, strontium tetra-oxidodi[ferrate(III)/vanad-ate(III)], have been obtained as a side product in the course of sinter experiments aimed at the synthesis of double perovskites in the system SrO-Fe2O3-V2O5. The crystal structure can be characterized by layers of six-membered rings of TO4-tetra-hedra (T: FeIII, VIII) perpendicular to [100]. Stacking of the layers along [100] results in a three-dimensional framework enclosing tunnel-like cavities in which SrII cations are incorporated for charge compensation. The sequence of directedness of up (U) and down (D) pointing vertices of neighboring tetra-hedra in a single six-membered ring is UUUDDD. The topology of the tetra-hedral framework belongs to the zeolite-type ABW.
