Molecular Studies of Relationships and Identifications Among Insects of the Subfamily Panchaetothripinae (Thysanoptera, Thripidae).

Project description:The Panchaetothripinae comprises 42 genera and 146 species of leaf-feeding thrips, some of which are horticultural pests. We examined representatives of the 18 genera that include most of these pests. For species delimitation, we used DNA barcoding to produce171 sequences for 40 morphospecies. Most species were found to be monophyletic, although cryptic diversity was evident in 8 presumptive species. A multilocus molecular phylogenetic assessment was based on one mitochondrial (COI) and three nuclear loci (EF-1α, ITS2, and 28S) from 132 specimens (18 genera and 33 species), representing all genera and ~82% of species in China. Maximum likelihood (ML) and Bayesian inference (BI) confirmed monophyly of each genus with strong support. Monophyly of tribes Panchaetothripini and Monilothripini were refuted, but the well supported tribe Tryphactothripini was confirmed. Rhipiphorothrips was recovered as a sister to the remainder of the genera of Panchaetothripinae combined. Both analyses revealed two major clades. Clade A comprised the majority of the genera, including tribe Tryphactothripini. Clade B included only four genera of which two, Helionothrips and Caliothrips, are particularly species rich. The relationships of some genera remain unresolved.

The new species is characterised by the head entirely dark brown, antennal segments I-VIII almost uniformly yellowish brown and III-IV strongly vasiform, metascutellum without produced posterior margins, and male has no pore plate on the abdominal sternites.

Effects of Temperature on the Developmental and Reproductive Biology of North American Bean Thrips, Caliothrips fasciatus (Pergande) (Thysanoptera: Thripidae: Panchaetothripinae).

Project description:North American bean thrips, Caliothrips fasciatus, native to California U.S., has been detected inside the navels of navel oranges exported from California for more than 120 years. Despite this long history of accidental movement into new areas, this thrips has failed to establish populations outside of its native range. The cold accumulation hypothesis postulates that increasing levels of cold stress experienced by thrips overwintering inside navels is compounded when harvested fruit is shipped under cold storage conditions. Consequently, the fitness of surviving thrips is compromised, which greatly diminishes invasion potential. At the time this study was conducted, the effects of temperature on C. fasciatus fitness were unknown. To address this shortcoming, the effects of nine fluctuating temperatures that averaged 8, 10, 15, 20, 25, 30, 32, 35, and 37 °C over a 24 h period on the developmental and reproductive biology of C. fasciatus were evaluated. One linear and five nonlinear regression functions were fit to egg-to-adult development rate data for parent and offspring thrips to characterize thermal performance curves. Estimates of minimum, optimal, and maximum temperature thresholds for development were in the ranges of -4.37-6.52 °C (i.e., Tmin), 31.19-32.52 °C (i.e., Topt), and 35.07-37.98 °C (i.e., Tmax), respectively. Degree day accumulation to complete development, estimated from linear regression, ranged 370.37-384.61. Average development times for eggs, first and second instar larvae, propupae, pupae, and adult longevity, and mean lifetime fecundity of females were significantly affected by temperature. These biological responses to temperature may provide insight into how this abiotic variable affects the invasion potential of C. fasciatus.

A trichodorus (triplonchida: trichodoridae) nematode from thrips (thysanoptera: panchaetothripinae).

Project description:A thrips insect Caliothrips sp. (Thysanoptera: Panchaetothripinae) from persimmon fruit (Ebenaceae: Diospyros sp.) from an unknown origin, possibly Asia, was intercepted in a passenger bag in November 2012 at the Peace Arch Border Crossing from Canada to Blaine, WA, by a USDA-APHIS-PPQ port inspector. Nematodes were attached to the abdomen of the female insect and sent to us in saline. Seven nematodes (five females, two males) were measured and these and others were processed for permanent slides. An adult female and a female juvenile were prepared for PCR. Morphologically these nematodes belonged to the Trichodorus sparsus group, and the 28S rDNA D2-D3 sequence showed greatest similarity to Trichodorus paragiennensis (94%) and T. giennensis (93%), with greatest morphological similarity to the latter species. Among other morphological differences, the innermost uterus width is wider than in related species. Trichodorus spp. are normally found in soil, so this is the first population seen in the atypical habitat of an insect. Morphological and molecular characteristics of Trichodorus sp. are presented, but a putative new species name is not currently advisable because of relatively poor condition of specimens. Ecological associations are also discussed.

Siamothrips balteus, a new species of Scirtothrips genus-group from China (Thysanoptera, Thripidae).

Project description:The third species of the genus Siamothrips Okajima, Siamothrips balteussp. n., is described from China. The new species is characterised by the abdominal tergite II uniformly brown, III-VII with a brown area medially but pale on lateral thirds, tergite VIII smooth medially, tergite X with 3-4 rows of microtrichia medially, and abdominal sternite VII with one pair of discal setae laterally. A key to the three species has been constructed and is presented here.

Reproductive Behavior of Echinothrips americanus (Thysanoptera: Thripidae).

Project description:Most Thysanoptera possess a haplo-diploid reproductive mode and reproduce via arrhenotoky. Females can mature eggs successively throughout almost their entire life, but in most terebrantian thrips spermiogenesis is complete by adult male eclosion, and testes contain only mature spermatids. In parasitoid wasps this phenomenon of preadult spermiogenesis is described as prospermatogeny. It is unclear if prospermatogeny and this predetermined sperm quantity have implications for mating strategy and fitness. In this study, we give a detailed description of mating behavior of the thripine species Echinothrips americanus, which largely corresponds with the only available data of another species of this family, Frankliniella occidentalis (Thysanoptera: Thripidae). With investigations using light microscopy, we describe for the first time the chronological sequence of internal processes during copulation. The release of male accessory gland material followed subsequently by spermatozoa indicates production of a female-determined type 1 spermatophore. Despite prospermatogeny, males are able to inseminate 10 females with an equal amount of spermatozoa. It is only the quantity of glandular material that decreases with the number of previous copulations. Based on these new findings, the reproductive strategy of this species is discussed.

Identification and Virulence Characterization of Two Akanthomyces attenuatus Isolates Against Megalurothrips usitatus (Thysanoptera: Thripidae).

Project description:Megalurothrips usitatus (Bagnall) is one of the most harmful pests of leguminous plants. In order to expand our knowledge on the infection of M. usitatus by entomopathogenic fungi, two newly identified isolates of Akanthomyces attenuatus (Zare & Gams) were tested for their pathogenicity against M. usitatus. Both isolates of A. attenuatus (SCAUDCL-38 and SCAUDCL-56) were isolated from soil and were identified by morphological and molecular analyses. The adult females of M. usitatus were treated with five different concentrations (1 × 104, 1 × 105, 1 × 106, 1 × 107, and 1 × 108 conidia/mL) of the isolates. Our results revealed 76.25% and 57.5% mortality of M. usitatus after five days of treatment with 1 × 108 conidia/mL of SCAUDCL-38 and SCAUDCL-56, respectively. The median lethal concentrations (LC50) of SCAUDCL-38 and SCAUDCL-56 calculated through linear regression analysis after five days of fungal treatment of M. usitatus were 1.9 × 106 and 1.5 × 107 conidia/mL, respectively, whereas the median lethal time (LT50) observed for 1 × 108 conidia/mL of SCAUDCL-38 and SCAUDCL-56 were 3.52 days and 4.9 days, respectively. A. attenuatus isolates SCAUDCL-38 and SCAUDCL-56 are highly pathogenic strains of M. usitatus. These findings offer valuable information on the development and commercialization of alternative control measures against M. usitatus.

This is characterised by considerable intra-population variation in the number and size of brown markings on the abdominal tergites. Project description:South East Asia pest thrips species, Thrips parvispinus (Karny), is a serious pest on a number of agricultural and horticultural crops in a number of plant families. Based on an integrated approach of morphology and DNA barcoding, invasion of this serious pest is reported first time from India on papaya plantations. Molecular data have corroborated with the morphological identification. Haplotyping data suggested that the Indonesia may be a probable source of invasion of this pest to India.
