Ottelia fengshanensis, a new bisexual species of Ottelia (Hydrocharitaceae) from southwestern China.

ABSTRACT: Ottelia fengshanensis, a new species (Hydrocharitaceae) from southwest China is here described and illustrated. Comparing its morphological features to putative close relatives O. guanyangensis, it has 3-4 flowers (vs. 2-5) each spathe, hexagonal-cylindric fruit, white styles (vs. yellow), green leaves (vs. dark green) and fruit tiny winged (vs. winged obviously). Molecular phylogenetic investigation of four DNA sequences (ITS, rbcL, trnK5' intron and trnS-trnG) and the Poisson Tree Processes model for species delimitation (PTP) analysis, further resolves O. fengshanensis as a new species that is close to O. guanyangensis with distinct support.

Characterization of the complete chloroplast genome of Ottelia alismoides (Hydrocharitaceae), a vulnerable submerged macrophyte in China.

Project description:Ottelia alismoides (Linn.) Pers. (Hydrocharitaceae) is an endangered submerged macrophyte in China. In his study, we assembled complete chloroplast (cp) genome of O. alismoides based on the Illumina reads. The cp genome of O. alismoides was 157,885 bp in length, including a large single copy (LSC) region of 87,707 bp and small single copy (SSC) region of 20,234 bp, separated by two inverted repeat (IR) regions of 24,972 bp each. The cp genome encoded 115 genes including 70 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. The GC content of cp genome of O. alismoides is 36.6%. A maximum likelihood phylogenetic analysis revealed that O. alismoides is closely related to O. acuminate var. songmingensis.

Development and characterization of EST-SSR markers for Ottelia acuminata var. jingxiensis (Hydrocharitaceae).

Project description:Simple sequence repeat (SSR) markers were derived from transcriptomic data for Ottelia acuminata (Hydrocharitaceae), a species comprising five endemic and highly endangered varieties in China. Sixteen novel SSR markers were developed for O. acuminata var. jingxiensis. One to eight alleles per locus were found, with a mean of 2.896. The observed and expected heterozygosity ranged from 0.000 to 1.000 and 0.000 to 0.793, respectively. Interestingly, in cross-varietal amplification, 13 out of the 16 loci were successfully amplified in O. acuminata var. acuminata, and 12 amplified in each of the other three varieties of O. acuminata. These newly developed SSR markers will facilitate further study of genetic variation and provide important genetic data needed for appropriate conservation of natural populations of all varieties of O. acuminata.

Dysosmavillosa (Berberidaceae), a new species from Guizhou, Southwestern China.

Project description:A new species, Dysosmavillosa Z.W.Wang & H.C.Xi, is described and illustrated based on collections from the Yueliang Mountains in Congjiang County of Guizhou, Southwestern China. It is morphologically similar to D.difformis (Hemsl. & E.H.Wilson) T.H.Wang ex T.S.Ying, but can be easily distinguished from D.difformis by its inflorescences bearing a greater number of flowers (5-13 vs. 2-5), white-villous stems, petioles, and abaxial leaf blade, and stigma dark purple-red. In addition, we also compare this new species to the three species of Podophyllum (i.e., P.glaucescens J.M.H.Shaw, P.hemsleyi J.M.H.Shaw & Stearn, and P.trilobulum J.M.H.Shaw) which are insufficiently known and listed as putative members of Dysosma in Flora of China. Dysosmavillosa can also be easily distinguished from P.glaucescens (7-flowered; to 40 cm tall) and P.hemsleyi (4-flowered; to 40 cm tall) by its inflorescences bearing a greater number of flowers, relatively smaller stature (9-23 cm tall), stems, petioles and abaxial leaf blade densely white-villose. Although the stems and petioles of P.trilobulum also possess fine short hairs, it can be easily distinguished from D.villosa by its trilobulate leaves, inflorescence with fewer flowers (2-5), and the position of inflorescence (inserted at or above midpoint on petiole of upper leaf).

Four New Species of Hemileccinum (Xerocomoideae, Boletaceae) from Southwestern China.

Project description:The genus Hemileccinum belongs to the subfamily Xerocomoideae of the family Boletaceae. In this study, phylogenetic inferences of Hemileccinum based on sequences of a single-locus (ITS) and a multi-locus (nrLSU, tef1-α, rpb1, rpb2) were conducted. Four new species, namely H. abidum, H. brevisporum, H. ferrugineipes and H. parvum were delimited and proposed based on morphological and molecular evidence. Descriptions and line-drawings of them were presented, as well as their comparisons to allied taxa. Our study shed new light on the recognition of the genus. The pileipellis of the species in this genus should mostly be regarded as (sub)epithelium to hyphoepithelium, because the pileipellis of most studied species here is composed of short inflated cells in the inner layer (subpellis) and filamentous hyphae in outer layer (suprapellis). The basidiospores of the studied species, including the type species, H. impolitum, have a warty surface.

Veronicastrum wulingense (Plantaginaceae), a new species from Southwestern Hubei, China.

Project description:BackgroundThe genus Veronicastrum Heist. ex Fabr. are mainly distributed in East Asia, and only Veronicastrum virginicum (L.) Farw. is disjunctively distributed in eastern North America. The south area of China (extending to Taiwan Island) is the richest in Veronicastrum species. It is of medicinal importance in China as traditional herbs used to treat ascites diseases that caused by schistosomiasis. During field investigation of plant resources in Pingbaying National Forest Park, Southwestern Hubei, China, an unknown flowering population of Veronicastrum was discovered from thick humus layers adjacent to rocks under broad-leaved forests by walkways. They were collected and morphological characters assesed for further taxonomic treatment. Molecular analysis was also conducted to ascertain its phylogenetic position in the genus Veronicastrum.ResultsThis species is similar to Veronicastrum liukiuense (Ohwi) T.Yamaz. from the Ryukyu Islands, but can be distinctly differed by its axillary inflorescences (versus terminal on short leafy branches), pedicels up to 2.5 mm (versus sessile), corollas purple to purple-red (versus white tinged with pale purple) and florescence June to July (versus September to October). Also, phylogenetic studies showed the species was an independent clade in the genus Veronicastrum based on the maximum likelihood (ML) analyses using two different matrix sequences of concatenated molecular markers. The plastid genome of this new species is also reported in this study for the first time.ConclusionThe morphological and molecular evidences support the recognition of Veronicastrum wulingense as a new species.

The new species is morphologically similar to O. japonica, but differs from the latter by having terete stems which are densely hirtellous, usually persistent ciliate stipules with well-developed colleters inside the base of the stipule, shorter corolla tubes and shorter stamens and styles.

New Species of Talaromyces (Trichocomaceae, Eurotiales) from Southwestern China.

Project description:Species of Talaromyces are cosmopolitan and ubiquitous, and some are of industrial and medicinal importance. Species of Talaromyces have been successively reported in China. During our examinations of samples collected from southwestern China, two new species belonging to Talaromyces sect. Talaromyces were further discovered based on phylogenetic analyses and morphological comparisons. Talaromyces ginkgonis sp. nov., isolated from a partially colonized fruit of Ginkgo biloba, differs from closely-related fungi in the combination of conidia ellipsoidal, smooth and 3.5-4 × 2-3 μm, no growth on CYA at 37 °C and sequence divergences; T. shilinensis sp. nov. is distinguished from its related allies in the combination of smooth conidia, colonies 10-11 mm diameter on CYA at 25 °C and sequence differences. Detailed descriptions and illustrations of the new taxa are given.

Deciphering the Taxonomic Delimitation of Ottelia acuminata (Hydrocharitaceae) Using Complete Plastomes as Super-Barcodes.

Project description:Accurate species delimitation and identification, which is a challenging task in traditional morphology-based taxonomy, is crucial to species conservation. Ottelia acuminata (Hydrocharitaceae) is a severely threatened submerged macrophyte endemic to southwestern China. The taxonomy of O. acuminata, which has long been in dispute, remains unresolved, impeding effective conservation and management practices. Here, we aim to address the long-standing issues concerning species boundary and intraspecific subdivision of O. acuminata using complete plastome sequences as super-barcodes. The taxonomic delimitation of O. acuminata was explored using phylogenetic inference and two independent sequence-based species delimitation schemes: automatic barcode gap discovery (ABGD) and multi-rate Poisson tree processes (mPTP). The reciprocally reinforcing results support the reduction of the closely related congeneric species, O. balansae and O. guanyangensis, as two conspecific varieties of O. acuminata. Within the newly defined O. acuminata, accurate varietal identification can be achieved using plastome super-barcodes. These findings will help inform future decisions regarding conservation, management and restoration of O. acuminata. This case study suggests that the use of plastome super-barcodes can provide a solution for species delimitation and identification in taxonomically difficult plant taxa, thus providing great potential to lessen the challenges of inventorying biodiversity, as well as biologically monitoring and assessing threatened species.

Spiradiclisdetianensis (Rubiaceae, Ophiorrhizeae), a new species from southwestern Guangxi, China.

Project description:A new species of Rubiaceae, Spiradiclisdetianensis is described from a limestone karst area of southwestern China. This new species is morphologically similar to S.cordata and S.spathulata. All of them have rosetted habit and long peduncles, but it differs from the former by the cuneate leaf bases (vs. basally cordate) and much longer corolla tubes (1.8-2.2 cm long vs. ca. 5 mm long), and from the latter mainly by its tubular-funnel shaped corolla (vs. slenderly salver shaped), 4.5-6.8 (vs. 1.5-2) mm in diam, inside throat and corolla densely puberulent (vs. glabrous except a ring of long hairs at the middle). It also resembles to S.tubiflora, but differs clearly by its subrosulate habit (vs. procumbent to creeping), longer leaf blades (7.0-10.5 cm vs. 0.5-2.5 cm) and longer corolla tubes (18-22 mm vs. 14-16 mm). At same time, color photos, illustrations, detailed descriptions and conservation status of the new species are provided.
