Novel Reassortant Avian Influenza A(H3N8) Virus Isolated from a Wild Bird in Jiangxi, China.

ABSTRACT: Here, we report the detection of a reassortant avian influenza A(H3N8) virus isolated from a wild bird in Poyang Lake, Jiangxi, China, in 2014. Phylogenetic analyses indicated that this virus is most likely derived from the Eurasian-origin H3Ny and HxN8 viruses and two strains endemic to China, namely, H5N1 and H5N6.

Genetic Characterization of a Natural Reassortant H3N8 Avian Influenza Virus Isolated from Domestic Geese in Guangxi, Southern China.

Project description:A H3N8 subtype of avian influenza virus, A/goose/Guangxi/020G/2009(H3N8) (GX020G), was isolated from the Guangxi Province of China in 2009. All eight gene segments of the GX020G strain were sequenced. Sequence analysis indicated that this H3N8 virus is a novel reassortant strain. The genome sequences provide useful information for understanding the epidemiology and molecular characteristics of the H3N8 subtype of influenza virus in southern China.

Detection of novel reassortant H9N2 avian influenza viruses in wild birds in Jiangxi Province, China.

Project description:The five avian influenza A/H9N2 viruses isolated from wild birds in Jiangxi, China in 2015 are novel reassortants which most likely evolved from multiple lineages. They shared a high similarity with isolates from poultry, suggesting a frequent contact and continuous viral circulation at the bird-poultry interface. Given the continuous reassortment of H9N2 viruses, it will of substantial importance to implement routine surveillance in wild birds to successfully control avian influenza viruses and better the early warning system of the emerging reassortants with pandemic potential.

Avian influenza H9N2 virus isolated from air samples in LPMs in Jiangxi, China.

Project description:BackgroundRecently, avian influenza virus has caused repeated worldwide outbreaks in humans. Live Poultry Markets (LPMs) play an important role in the circulation and reassortment of novel Avian Influenza Virus (AIVs). Aerosol transmission is one of the most important pathways for influenza virus to spread among poultry, from poultry to mammals, and among mammals.MethodsIn this study, air samples were collected from LPMs in Nanchang city between April 2014 and March 2015 to investigate possible aerosol transmission of AIVs. Air samples were detected for Flu A by Real-Time Reverse Transcription-Polymerase Chain Reaction (RRT-PCR). If samples were positive for Flu A, they were inoculated into 9- to 10-day-old specific-pathogen-free embryonated eggs. If the result was positive, the whole genome of the virus was sequenced by MiSeq. Phylogenetic trees of all 8 segments were constructed using MEGA 6.05 software.ResultsTo investigate the possible aerosol transmission of AIVs, 807 air samples were collected from LPMs in Nanchang city between April 2014 and March 2015. Based on RRT-PCR results, 275 samples (34.1%) were Flu A positive, and one virus was successfully isolated with embryonated eggs. The virus shared high nucleotide homology with H9N2 AIVs from South China.ConclusionsOur study provides further evidence that the air in LPMs can be contaminated by influenza viruses and their nucleic acids, and this should be considered when choosing and evaluating disinfection strategies in LPMs, such as regular air disinfection. Aerosolized viruses such as the H9N2 virus detected in this study can increase the risk of human infection when people are exposed in LPMs.

Characterization of an Avian Influenza Virus H9N2 Strain Isolated from a Wild Bird in Southern China.

Project description:We isolated an avian influenza virus H9N2 strain from a wild bird in the Guangxi Province of southern China in 2013 named A/turtledove/Guangxi/49B6/2013(H9N2) (GX49B6). We aimed to understand the genetic characters of the GX49B6 strain by analyzing the complete genome sequence. The results showed that our isolated strain has features of low pathogenic avian influenza viruses and viruses that infect humans. The discovery of the complete genome sequence of the GX49B6 strain may be helpful to further the understanding of the epidemiology and surveillance of avian influenza viruses in the field.

Adaptation of Two Wild Bird-Origin H3N8 Avian Influenza Viruses to Mammalian Hosts.

Project description:Wild birds play an important role in the emergence, evolution, and spread of zoonotic avian influenza viruses (AIVs). However, there are few studies on the cross-species transmission of the H3N8 AIV originating from wild birds. In this study, we investigated the transmissibility and pathogenicity of two H3N8 low pathogenic avian influenza viruses (LPAIVs) isolated from wild birds, GZA1 and XJ47, to mammals. The HA genes of both strains belonged to Eurasian isolates, while the other genes were derived from a variety of other subtypes of AIVs. Both strains can infect specific-pathogen-free (SPF) chickens, BALB/c mice, and guinea pigs. The XJ47 strain spread horizontally in SPF chickens and guinea pigs. The GZA1 strain did not spread horizontally but caused higher weight loss and mild lung inflammation in mice. P12-GZA1- and P12-XJ47-adapted strains obtained after 12 passages in the lung of mice showed enhanced pathogenicity in mice, which led to obvious clinical symptoms, lung inflammation, and 100% death. Both adapted strains have the reported mutation T97I in the PA, and the reported mutation D701N in PB2 has been found in the P12-GZA1-adapted strain. This study provides an important scientific basis for the continuous monitoring of wild AIVs and the mechanism underlying AIV cross-species transmission.

Complete Genome Sequence of a Novel Reassortant H6N8 Avian Influenza Virus Isolated from Wild Waterfowl in Poyang Lake, China.

Project description:Here, we report the complete genome sequence of an H6N8 avian influenza virus (AIV) isolated from wild waterfowl in Poyang Lake, China, in 2016. Phylogenetic analysis showed that it was a novel reassortant AIV between domestic ducks and wild waterfowl. The finding of this study is helpful for our understanding of the ecology and the evolutionary characteristics of H6 subtypes of AIV in birds.

Novel Reassortant Avian Influenza A(H5N6) Virus, China, 2021.

Project description:Although reports of human infection with influenza A(H5N6) increased in 2021, reports of similar H5N6 virus infection in poultry are few. We detected 10 avian influenza A(H5N6) clade 2.3.4.4b viruses in poultry from 4 provinces in China. The viruses showed strong immune-escape capacity and complex genetic reassortment, suggesting further transmission risk.

Thus, the genome information of the H3N2 isolate will help to investigate further the epidemiology of H3 subtype AIVs in China.
