Project description:Prior research has shown that the perception of degraded speech is influenced by within sentence meaning and recruits one or more components of a frontal-temporal-parietal network. The goal of the current study is to examine whether the overall conceptual meaning of a sentence, made up of one set of words, influences the perception of a second acoustically degraded sentence, made up of a different set of words. Using functional magnetic resonance imaging (fMRI), we presented an acoustically clear sentence followed by an acoustically degraded sentence and manipulated the semantic relationship between them: Related in meaning (but consisting of different content words), Unrelated in meaning, or Same. Results showed that listeners' word recognition accuracy for the acoustically degraded sentences was significantly higher when the target sentence was preceded by a conceptually related compared to a conceptually unrelated sentence. Sensitivity to conceptual relationships was associated with enhanced activity in middle and inferior frontal, temporal, and parietal areas. In addition, the left middle frontal gyrus (LMFG), left inferior frontal gyrus (LIFG), and left middle temporal gyrus (LMTG) showed activity that correlated with individual performance on the Related condition. The superior temporal gyrus (STG) showed increased activation in the Same condition suggesting that it is sensitive to perceptual similarity rather than the integration of meaning between the sentence pairs. A fronto-temporo-parietal network appears to consolidate information sources across multiple levels of language (acoustic, lexical, syntactic, semantic) to build, and ultimately integrate conceptual information across sentences and facilitate the perception of a degraded speech signal. However, the nature of the sources of information that are available differentially recruit specific regions and modulate their activity within this network. Implications of these findings for the functional architecture of the network are considered.

Project description:In natural listening situations, speech perception is often impaired by degraded speech sounds arriving at the ear. Contextual speech information can improve the perception of degraded speech and modify neuronal responses elicited by degraded speech. However, most studies on context effects on neural responses to degraded speech confounded lexico-semantic and sublexical cues. Here, we used fMRI to investigate how prior sublexical speech (e.g. pseudowords cues) affects neural responses to degraded sublexical speech and hence its processing and recognition. Each trial consisted of three consecutively presented pseudowords, of which the first and third were identical and degraded. The second pseudoword was always presented in clear form and either matched or did not match the degraded pseudowords. Improved speech processing through sublexical processing was associated with BOLD activation increases in frontal, temporal, and parietal regions, including the primary auditory cortex (PAC), posterior superior temporal cortex, angular gyrus, supramarginal gyrus, middle temporal cortex, and somato-motor cortex. These brain regions are part of a speech processing network and are involved in lexico-semantic processing. To further investigate the adaptive changes in PAC, we conducted a bilateral region of interest analysis on PAC subregions. PAC ROIs showed bilaterally increased activation in the match condition compared with the mismatch condition. Our results show that the perception of unintelligible degraded speech is improved and the neuronal population response is enhanced after exposure to intact sublexical cues. Furthermore, our findings indicate that the processing of clear meaningless sublexical speech preceding degraded speech could enhance the activity in the brain regions that belong to the cortical speech processing network previously reported in studies investigating lexico-semantic speech.

Project description:Auditory verbal hallucinations (AVH) are a hallmark of psychotic experience. Various mechanisms including misattribution of inner speech and imbalance between bottom-up and top-down factors in auditory perception potentially due to aberrant connectivity between frontal and temporo-parietal areas have been suggested to underlie AVH. Experimental evidence for disturbed connectivity of networks sustaining auditory-verbal processing is, however, sparse. We compared functional resting-state connectivity in 49 psychotic patients with frequent AVH and 49 matched controls. The analysis was seeded from the left middle temporal gyrus (MTG), thalamus, angular gyrus (AG) and inferior frontal gyrus (IFG) as these regions are implicated in extracting meaning from impoverished speech-like sounds. Aberrant connectivity was found for all seeds. Decreased connectivity was observed between the left MTG and its right homotope, between the left AG and the surrounding inferior parietal cortex (IPC) and the left inferior temporal gyrus, between the left thalamus and the right cerebellum, as well as between the left IFG and left IPC, and dorsolateral and ventrolateral prefrontal cortex (DLPFC/VLPFC). Increased connectivity was observed between the left IFG and the supplementary motor area (SMA) and the left insula and between the left thalamus and the left fusiform gyrus/hippocampus. The predisposition to experience AVH might result from decoupling between the speech production system (IFG, insula and SMA) and the self-monitoring system (DLPFC, VLPFC, IPC) leading to misattribution of inner speech. Furthermore, decreased connectivity between nodes involved in speech processing (AG, MTG) and other regions implicated in auditory processing might reflect aberrant top-down influences in AVH.

Project description:This paper describes a general kernel regression approach to predict experimental conditions from activity patterns acquired with functional magnetic resonance image (fMRI). The standard approach is to use classifiers that predict conditions from activity patterns. Our approach involves training different regression machines for each experimental condition, so that a predicted temporal profile is computed for each condition. A decision function is then used to classify the responses from the testing volumes into the corresponding category, by comparing the predicted temporal profile elicited by each event, against a canonical hemodynamic response function. This approach utilizes the temporal information in the fMRI signal and maintains more training samples in order to improve the classification accuracy over an existing strategy. This paper also introduces efficient techniques of temporal compaction, which operate directly on kernel matrices for kernel classification algorithms such as the support vector machine (SVM). Temporal compacting can convert the kernel computed from each fMRI volume directly into the kernel computed from beta-maps, average of volumes or spatial-temporal kernel. The proposed method was applied to three different datasets. The first one is a block-design experiment with three conditions of image stimuli. The method outperformed the SVM classifiers of three different types of temporal compaction in single-subject leave-one-block-out cross-validation. Our method achieved 100% classification accuracy for six of the subjects and an average of 94% accuracy across all 16 subjects, exceeding the best SVM classification result, which was 83% accuracy (p=0.008). The second dataset is also a block-design experiment with two conditions of visual attention (left or right). Our method yielded 96% accuracy and SVM yielded 92% (p=0.005). The third dataset is from a fast event-related experiment with two categories of visual objects. Our method achieved 77% accuracy, compared with 72% using SVM (p=0.0006).

Project description:Binocular rivalry is a phenomenon where the simultaneous presentation of two different stimuli to the two eyes leads to alternating perception of the two stimuli. The temporary dominance of one stimulus over the other is influenced by several factors. Here, we studied the influence of reward on binocular rivalry dynamics and its neural representation in visual cortex. Orthogonal rotating grating stimuli were shown continuously, while monetary reward was given during the conscious perception of one stimulus but not the other. Periods of perceptual dominance were assessed both through participants' subjective report and objectively using functional magnetic resonance imaging and multi-voxel pattern analysis. Results did not confirm previous evidence for an effect of reward on perceptual dominance durations. Exploratory post-hoc analyses indicated that knowledge regarding both the reward contingency and the subjective nature of perceptual alternations may have interfered with potential reward effects on perceptual phase durations, suggesting a moderating role of meta-cognitive awareness in reward-based perceptual inference. Future studies of top-down influences on bistable perception should carefully consider the methodological challenges related to meta-cognitive awareness.

Project description:Three-dimensional interactive virtual environments (VEs) are a powerful tool for brain-imaging based cognitive neuroscience that are presently under-utilized. This paper presents machine-learning based methods for identifying brain states induced by realistic VEs with improved accuracy as well as the capability for mapping their spatial topography on the neocortex. VEs provide the ability to study the brain under conditions closer to the environment in which humans evolved, and thus to probe deeper into the complexities of human cognition. As a test case, we designed a stimulus to reflect a military combat situation in the Middle East, motivated by the potential of using real-time functional magnetic resonance imaging (fMRI) in the treatment of post-traumatic stress disorder. Each subject experienced moving through the virtual town where they encountered 1-6 animated combatants at different locations, while fMRI data was collected. To analyze the data from what is, compared to most studies, more complex and less controlled stimuli, we employed statistical machine learning in the form of Multi-Voxel Pattern Analysis (MVPA) with special attention given to artificial Neural Networks (NN). Extensions to NN that exploit the block structure of the stimulus were developed to improve the accuracy of the classification, achieving performances from 58 to 93% (chance was 16.7%) with six subjects. This demonstrates that MVPA can decode a complex cognitive state, viewing a number of characters, in a dynamic virtual environment. To better understand the source of this information in the brain, a novel form of sensitivity analysis was developed to use NN to quantify the degree to which each voxel contributed to classification. Compared with maps produced by general linear models and the searchlight approach, these sensitivity maps revealed a more diverse pattern of information relevant to the classification of cognitive state.

Project description:The speech we hear every day is typically "degraded" by competing sounds and the idiosyncratic vocal characteristics of individual speakers. While the comprehension of "degraded" speech is normally automatic, it depends on dynamic and adaptive processing across distributed neural networks. This presents the brain with an immense computational challenge, making degraded speech processing vulnerable to a range of brain disorders. Therefore, it is likely to be a sensitive marker of neural circuit dysfunction and an index of retained neural plasticity. Considering experimental methods for studying degraded speech and factors that affect its processing in healthy individuals, we review the evidence for altered degraded speech processing in major neurodegenerative diseases, traumatic brain injury and stroke. We develop a predictive coding framework for understanding deficits of degraded speech processing in these disorders, focussing on the "language-led dementias"-the primary progressive aphasias. We conclude by considering prospects for using degraded speech as a probe of language network pathophysiology, a diagnostic tool and a target for therapeutic intervention.

Project description:Previous studies investigating the anterior cingulate cortex (ACC) have relied on a number of tasks which involved cognitive control and attentional demands. In this fMRI study, we tested the model that ACC functions as an attentional network in the processing of language. We employed a paradigm that requires the processing of concurrent linguistic information predicting that the cognitive costs imposed by competing trials would engender the activation of ACC. Subjects were confronted with sentences where the semantic content conflicted with the prosodic intonation (CONF condition) randomly interspaced with sentences which conveyed coherent discourse components (NOCONF condition). We observed the activation of the rostral ACC and the middle frontal gyrus when the NOCONF condition was subtracted from the CONF condition. Our findings provide evidence for the involvement of the rostral ACC in the processing of complex competing linguistic stimuli, supporting theories that claim its relevance as a part of the cortical attentional circuit. The processing of emotional prosody involved a bilateral network encompassing the superior and medial temporal cortices. This evidence confirms previous research investigating the neuronal network that supports the processing of emotional information.

Project description:The present study aimed to investigate the effects of degraded speech perception and binaural unmasking using functional near-infrared spectroscopy (fNIRS). Normal hearing listeners were tested when attending to unprocessed or vocoded speech, presented to the left ear at two speech-to-noise ratios (SNRs). Additionally, by comparing monaural versus diotic masker noise, we measured binaural unmasking. Our primary research question was whether the prefrontal cortex and temporal cortex responded differently to varying listening configurations. Our a priori regions of interest (ROIs) were located at the left dorsolateral prefrontal cortex (DLPFC) and auditory cortex (AC). The left DLPFC has been reported to be involved in attentional processes when listening to degraded speech and in spatial hearing processing, while the AC has been reported to be sensitive to speech intelligibility. Comparisons of cortical activity between these two ROIs revealed significantly different fNIRS response patterns. Further, we showed a significant and positive correlation between self-reported task difficulty levels and fNIRS responses in the DLPFC, with a negative but non-significant correlation for the left AC, suggesting that the two ROIs played different roles in effortful speech perception. Our secondary question was whether activity within three sub-regions of the lateral PFC (LPFC) including the DLPFC was differentially affected by varying speech-noise configurations. We found significant effects of spectral degradation and SNR, and significant differences in fNIRS response amplitudes between the three regions, but no significant interaction between ROI and speech type, or between ROI and SNR. When attending to speech with monaural and diotic noises, participants reported the latter conditions being easier; however, no significant main effect of masker condition on cortical activity was observed. For cortical responses in the LPFC, a significant interaction between SNR and masker condition was observed. These findings suggest that binaural unmasking affects cortical activity through improving speech reception threshold in noise, rather than by reducing effort exerted.
