Where do infants go? A longstanding assumption is that infants primarily crawl or walk to reach destinations viewed while stationary. However, many bouts of spontaneous locomotion do not end at new people, places, or things. Study 1 showed that half of 10- and 13-month-old crawlers' (N = 29) bouts end at destinations-more than previously found with walkers. Project description:Locomotion empowers animals to move. Locomotor-initiating signals from the brain are funneled through descending neurons in the brainstem that act directly on spinal locomotor circuits. Little is known in mammals about which spinal circuits are targeted by the command and how this command is transformed into rhythmicity in the cord. Here we address these questions leveraging a mouse brainstem-spinal cord preparation from either sex that allows locating the locomotor command neurons with simultaneous Ca2+ imaging of spinal neurons. We show that a restricted brainstem area - encompassing the lateral paragigantocellular nucleus (LPGi) and caudal ventrolateral reticular nucleus (CVL) - contains glutamatergic neurons which directly initiate locomotion. Ca2+ imaging captures the direct LPGi/CVL locomotor initiating command in the spinal cord and visualizes spinal glutamatergic modules that execute the descending command and its transformation into rhythmic locomotor activity. Inhibitory spinal networks are recruited in a distinctly different pattern. Our study uncovers the principal logic of how spinal circuits implement the locomotor command using a distinct modular organization.

Project description:During walking, uneven surfaces impose new demands for controlling balance and forward progression at each step. It is unknown to what extent walking may be refined given an amount of stride-to-stride unpredictability at the distal level. Here, we explored the effects of an uneven terrain surface on whole-body locomotor dynamics immediately following exposure and after a familiarization period. Eleven young, unimpaired adults walked for 12 min on flat and uneven terrain treadmills. The whole-body center of mass excursion range (COMexc) and peak velocity (COMvel), step length and width were estimated. On first exposure to uneven terrain, we saw significant increases in medial-lateral COMexc and lateral COMvel, and in the variability of COMexc, COMvel and foot placement in both anterior-posterior and medial-lateral directions. Increases in step width and decreases in step length supported the immediate adoption of a cautious, restrictive solution on uneven terrain. After familiarization, step length increased and the variability of anterior-posterior COMvel and step length reduced, while step width and lateral COMvel reduced, alluding to a refinement of movement and a reduction of conservative strategies over time. However, the variability of medial-lateral COMexc and lateral COMvel increased, consistent with the release of previously constrained degrees of freedom. Despite this increase in variability, a strong relationship between step width and medial-lateral center of mass movement was maintained. Our results indicate that movement strategies of unimpaired adults when walking on uneven terrain can evolve over time with longer exposure to the surface.

Project description:Recent advances in neurogenetics have highlighted Drosophila melanogaster as an exciting model to study neural circuit dynamics and complex behavior. Automated tracking methods have facilitated the study of complex behaviors via high throughput behavioral screening. Here we describe a newly developed low-cost assay capable of real-time monitoring and quantifying Drosophila group activity. This platform offers reliable real-time quantification with open source software and a user-friendly interface for data acquisition and analysis. We demonstrate the utility of this platform by characterizing ethanol-induced locomotor activity in a dose-dependent manner as well as the effects of thermo and optogenetic manipulation of ellipsoid body neurons important for ethanol-induced locomotor activity. As expected, low doses of ethanol induced an initial startle and slow ramping of group activity, whereas high doses of ethanol induced sustained group activity followed by sedation. Advanced offline processing revealed discrete behavioral features characteristic of intoxication. Thermogenetic inactivation of ellipsoid body ring neurons reduced group activity whereas optogenetic activation increased activity. Together, these data establish the fly Group Activity Monitor (flyGrAM) platform as a robust means of obtaining an online read out of group activity in response to manipulations to the environment or neural activity, with an opportunity for more advanced post-processing offline.

Project description:Voluntary limb modifications must be integrated with basic walking patterns during visually guided walking. In this study we tested whether voluntary gait modifications can become more automatic with practice. We challenged walking control by presenting visual stepping targets that instructed subjects to modify step length from one trial to the next. Our sequence learning paradigm is derived from the serial reaction-time (SRT) task that has been used in upper limb studies. Both random and ordered sequences of step lengths were used to measure sequence-specific and sequence-nonspecific learning during walking. In addition, we determined how age (i.e., healthy young adults vs. children) and biomechanical factors (i.e., walking speed) affected the rate and magnitude of locomotor sequence learning. The results showed that healthy young adults (age 24 ± 5 yr,n= 20) could learn a specific sequence of step lengths over 300 training steps. Younger children (age 6-10 yr,n= 8) had lower baseline performance, but their magnitude and rate of sequence learning were the same compared with those of older children (11-16 yr,n= 10) and healthy adults. In addition, learning capacity may be more limited at faster walking speeds. To our knowledge, this is the first study to demonstrate that spatial sequence learning can be integrated with a highly automatic task such as walking. These findings suggest that adults and children use implicit knowledge about the sequence to plan and execute leg movement during visually guided walking.

Project description:In previous phenotyping studies of mouse and rat exploratory behavior we developed a computational exploratory data analysis methodology including videotaping, tracking, preparatory methods for customized data analysis, a methodology for improving the replicability of results across laboratories, and algorithmic design for exposing the natural reference places (origins) used by animals during exploration. We then measured the animals' paths in reference to these origins, revealing robust, highly replicable modules termed excursions, which are performed from the origin into the environment and back to the origin. Origin-related exploration has been claimed to be phylogenetically conserved across the vertebrates. In the current study we use the same methodology to examine whether origin-related exploration has also been conserved in human pre-walking typically developing (TD) and a group of non-typically developing (NTD) infants in the presence of their stationary mother. The NTDs had been referred to a center for the early treatment of autism in infancy by pediatric neurologists and clinicians. The TDs established a reference place (origin) at mother's place and exhibited a modular partitioning of their path into excursions performed in reference to mother, visiting her often, and reaching closely. In contrast, the NTDs did not establish a distinct origin at the mother's place, or any other place, and did not partition the exploratory path into excursions. Once this difference is validated, the differences between the human infant groups may serve as an early referral tool for child development specialists. The absence of distinct modularity in human infants at risk of autism spectrum disorder can guide the search for animal models for this disorder in translational research.

Project description:Locomotor activity is used extensively as a behavioral output to study the underpinnings of circadian rhythms. Recent studies have required a populational approach for the study of circadian rhythmicity in Caenorhabditis elegans locomotion.We describe an imaging system for long-term automated recording and analysis of locomotion data of multiple free-crawling C. elegans animals on the surface of an agar plate. We devised image analysis tools for measuring specific features related to movement and shape to identify circadian patterns.We demonstrate the utility of our system by quantifying circadian locomotor rhythms in wild-type and mutant animals induced by temperature cycles. We show that 13 °C:18 °C (12:12h) cycles are sufficient to entrain locomotor activity of wild-type animals, which persist but are rapidly damped during 13 °C free-running conditions. Animals with mutations in tax-2, a cyclic nucleotide-gated (CNG) ion channel, significantly reduce locomotor activity during entrainment and free-running.Current methods for measuring circadian locomotor activity is generally restricted to recording individual swimming animals of C. elegans, which is a distinct form of locomotion from crawling behavior generally observed in the laboratory. Our system works well with up to 20 crawling adult animals, and allows for a detailed analysis of locomotor activity over long periods of time.Our population-based approach provides a powerful tool for quantification of circadian rhythmicity of C. elegans locomotion, and could allow for a screening system of candidate circadian genes in this model organism.

Project description:Flexibility and generativity are fundamental aspects of functional behavior that begin in infancy and improve with experience. How do infants learn to tailor their real-time solutions to variations in local conditions? On a nativist view, the developmental process begins with innate prescribed solutions, and experience elaborates on those solutions to suit variations in the body and the environment. On an emergentist view, infants begin by generating a variety of strategies indiscriminately, and experience teaches them to select solutions tailored to the current relations between their body and the environment. To disentangle these accounts, we observed coordination patterns in 11-month-old pre-walking infants with a range of cruising (moving sideways in an upright posture while holding onto a support) and crawling experience as they cruised over variable distances between two handrails they held for support. We identified infants' coordination patterns using a novel combination of computer-vision, machine-learning, and time-series analyses. As predicted by the emergentist view, the least experienced infants generated multiple coordination patterns inconsistently regardless of body size and handrail distance, whereas the most experienced infants tailored their coordination patterns to body-environment relations and switched solutions only when necessary. Moreover, the beneficial effects of experience were specific to cruising and not crawling, although both skills involve anti-phase coordination among the four limbs. Thus, findings support an emergentist view and suggest that everyday experience with the target skill may promote "learning to learn," where infants learn to assemble the appropriate solution for new problems on the fly.

Project description:Post-stroke walking impairment is a significant cause of chronic disability worldwide and often leads to loss of life roles for survivors and their caregivers. Walking impairment is traditionally classified into mild (>0.8 m/s), moderate (0.41-0.8 m/s), and severe (≤0.4 m/s), and those categorized as "severe" are more likely to be homebound and at greater risk of falls, fractures, and rehospitalization. In addition, there are minimal effective walking rehabilitation strategies currently available for this subgroup. Backward locomotor treadmill training (BLTT) is a novel and promising training approach that has been demonstrated to be safe and feasible across all levels of impairment; however, its benefits across baseline walking impairment levels (severe (≤0.4 m/s) vs. mild-moderate (>0.4 m/s)) have not been examined. Thirty-nine adults (>6 months post-stroke) underwent 6 days of BLTT (3×/week) over 2 weeks. Baseline and PRE to POST changes were measured during treadmill training and overground walking. Individuals with baseline severe walking impairment were at a more significant functional disadvantage across all spatiotemporal walking measures at baseline and demonstrated fewer overall gains post-training. However, contrary to our working hypothesis, both groups experienced comparable increases in cadence, bilateral percent single support times, and step lengths. BLTT is well tolerated and beneficial across all walking impairment levels, and baseline walking speed (≤0.4 m/s) should serve as a covariate in the design of future walking rehabilitation trials.

Project description:Unintentional injuries are among the ten leading causes of death in older adults; falls cause 60% of these deaths. Despite their effectiveness to improve balance and reduce the risk of falls, balance training programs have several drawbacks in practice, such as lack of engaging elements, boring exercises, and the effort and cost of travelling, ultimately resulting in low adherence. Exergames, that is, digital games controlled by body movements, have been proposed as an alternative to improve balance. One of the main challenges for exergames is to automatically quantify balance during game-play in order to adapt the game difficulty according to the skills of the player. Here we perform a multidimensional exploratory data analysis, using visualization techniques, to find useful measures for quantifying balance in real-time. First, we visualize exergaming data, derived from 400 force plate recordings of 40 participants from 20 to 79 years and 10 trials per participant, as heat maps and violin plots to get quick insight into the nature of the data. Second, we extract known and new features from the data, such as instantaneous speed, measures of dispersion, turbulence measures derived from speed, and curvature values. Finally, we analyze and visualize these features using several visualizations such as a heat map, overlapping violin plots, a parallel coordinate plot, a projection of the two first principal components, and a scatter plot matrix. Our visualizations and findings suggest that heat maps and violin plots can provide quick insight and directions for further data exploration. The most promising measures to quantify balance in real-time are speed, curvature and a turbulence measure, because these measures show age-related changes in balance performance. The next step is to apply the present techniques to data of whole body movements as recorded by devices such as Kinect.

Project description:The function of the brain is unlikely to be understood without an accurate description of its output, yet the nature of movement elements and their organization remains an open problem. Here, movement elements are identified from dynamics of walking in flies, using unbiased criteria. On one time scale, dynamics of walking are consistent over hundreds of milliseconds, allowing elementary features to be defined. Over longer periods, walking is well described by a stochastic process composed of these elementary features, and a generative model of this process reproduces individual behavior sequences accurately over seconds or longer. Within elementary features, velocities diverge, suggesting that dynamical stability of movement elements is a weak behavioral constraint. Rather, long-term instability can be limited by the finite memory between these elementary features. This structure suggests how complex dynamics may arise in biological systems from elements whose combination need not be tuned for dynamic stability.
