Further pathogenicity testing of Verticillium nonalfalfae, a biocontrol agent against the invasive Tree of Heaven (Ailanthus altissima), on non-target tree species in Europe

Project description:Verticillium nonalfalfae is an effective biological control agent against the highly invasive Tree of Heaven (Ailanthus altissima) in Europe, and previous research on ten economically and ecologically important tree species occurring in Austria have so far not revealed undesired non-target effects. In this study, another nine tree species including five native, two non-native as well as two invasive alien tree species were tested for susceptibility to the particular strain of V. nonalfalfae (Vert56) used for biological control of A. altissima. Stem inoculations on potted seedlings revealed that this strain of V. nonalfalfae is generally host-adapted to A. altissima. It induced chlorosis, necrosis and wilting already within two weeks post inoculation on A. altissima and resulted in almost completely defoliated or dead seedlings at the end of the vegetation period. Apart from two species (Quercus rubra and Sorbus aucuparia), that suffered from other abiotic/biotic agents, no mortality was observed on all other tree species tested; however, symptoms caused by other abiotic factors were also found on Prunus avium and Ulmus glabra. All tested tree species exhibited vascular discolorations and the fungus could be re-isolated at varying frequencies (6—100%) from inoculated seedlings of all non-target tree species, although five of these species exhibited no external symptoms. Results confirmed high susceptibility (S) of A. altissima to V. nonalfalfae, whereas Acer platanoides, Castanea sativa, Q. rubra, S. aucuparia and U. glabra were considered as tolerant (T), and A. negundo, P. avium, P. serotina and Q. petraea were rated as possible resistant (PR) due to the low rates of re-isolation. Complete mitochondrial genome of the Verticillium-wilt causing plant pathogen Verticillium nonalfalfae.

Project description:Verticillium nonalfalfae is a fungal plant pathogen that causes wilt disease by colonizing the vascular tissues of host plants. The disease induced by hop isolates of V. nonalfalfae manifests in two different forms, ranging from mild symptoms to complete plant dieback, caused by mild and lethal pathotypes, respectively. Pathogenicity variations between the causal strains have been attributed to differences in genomic sequences and perhaps also to differences in their mitochondrial genomes. We used data from our recent Illumina NGS-based project of genome sequencing V. nonalfalfae to study the mitochondrial genomes of its different strains. The aim of the research was to prepare a V. nonalfalfae reference mitochondrial genome and to determine its phylogenetic placement in the fungal kingdom. The resulting 26,139 bp circular DNA molecule contains a full complement of the 14 "standard" fungal mitochondrial protein-coding genes of the electron transport chain and ATP synthase subunits, together with a small rRNA subunit, a large rRNA subunit, which contains ribosomal protein S3 encoded within a type IA-intron and 26 tRNAs. Phylogenetic analysis of this mitochondrial genome placed it in the Verticillium spp. lineage in the Glomerellales group, which is also supported by previous phylogenetic studies based on nuclear markers. The clustering with the closely related Verticillium dahliae mitochondrial genome showed a very conserved synteny and a high sequence similarity. Two distinguishing mitochondrial genome features were also found-a potential long non-coding RNA (orf414) contained only in the Verticillium spp. of the fungal kingdom, and a specific fragment length polymorphism observed only in V. dahliae and V. nubilum of all the Verticillium spp., thus showing potential as a species specific biomarker.

A New Canthinone-Type Alkaloid Isolated from Ailanthus altissima Swingle.

Project description:The present investigation of the chemical constituents of the stem barks of Ailanthus altissima has resulted in the isolation of six canthinone-type alkaloids, including a new compound, (R)-5-(1-hydroxyethyl)-canthine-6-one (1), and five known compounds (2-6). Moreover, four phenyl propanoids (7-10), two lignans (11 and 12), two triterpenoids (13 and 14) and a fatty acid (15) having previously known chemical structures were isolated during the same course of this study. The structure of the new compound was elucidated by physical (m.p., [?]D) and spectroscopic data (¹H-NMR, (13)C-NMR, 2D NMR, and HR-DART-MS) interpretation and its absolute configuration was determined by electronic circular dichroism (ECD) data and quantum chemical calculations. The inflammatory activities of the isolates were screened on lipopolysaccharide (LPS)-induced nitric oxide (NO), a proinflammatory mediator, in RAW 264.7 cells. Among these isolated compounds, six compounds exhibited significant inhibition of NO production, with IC50 values in the range of 5.92 ± 0.9 to 15.09 ± 1.8 ?M.
