Mechanisms of Plant Tolerance to RNA Viruses Induced by Plant-Growth-Promoting Microorganisms.

ABSTRACT: Plant viruses are globally responsible for the significant crop losses of economically important plants. All common approaches are not able to eradicate viral infection. Many non-conventional strategies are currently used to control viral infection, but unfortunately, they are not always effective. Therefore, it is necessary to search for efficient and eco-friendly measures to prevent viral diseases. Since the genomic material of 90% higher plant viruses consists of single-stranded RNA, the best way to target the viral genome is to use ribonucleases (RNase), which can be effective against any viral disease of plants. Here, we show the importance of the search for endophytes with protease and RNase activity combined with the capacity to prime antiviral plant defense responses for their protection against viruses. This review discusses the possible mechanisms used to suppress a viral attack as well as the use of local endophytic bacteria for antiviral control in crops.

Project description:Salinity is one of the most severe abiotic stress in the world. Also, the irrigated lands have been treated with second salinity. Canola is one of the most important industrial crops for oil production all over the world which is affected by salinity. Salt stress causes imbalanced ion hemostasis (Na+ and K+) and interrupted mineral absorption in canola. Also, salinity stress leads to oxidative stress (production and accumulation of reactive oxygen species (ROS). Accumulation of ROS is extremely dangerous and lethal for plants. As a consequence, canola production is reduced under salinity stress. So, a suitable approach should be found to deal with salinity stress and prevent the loss of production oilseed. Plant growth-promoting rhizobacteria (PGPR) can colonize on the plant root surface and alleviate the salt stress effect by providing minerals like nitrogen, phosphate, and potassium. Also, they alleviate salt stress by phytohormones like auxin (IAA), cytokinin (CK), and abscisic acid (ABA). This study focus on physiological parameters like leaf area (LA), root length (RL), shoot length (SL), chlorophyll fluorescence indexes (Fv/Fm and Fv/F0), relative water content (RWC), electrolyte leakage index (ELI), photosynthesis pigments (chlorophyll a, b, and carotenoids), Na+, and K+; and biochemical parameters like malondialdehyde (MDA) content, hydrogen peroxide content (H2O2), total protein content, proline, antioxidant capacity, and antioxidant enzyme activities in canola through the inoculation with Enterobacter sp. S16-3 and Pseudomonas sp. C16-2O. This study showed that LA, RL, SL, chlorophyll fluorescence indexes, RWC were significantly increased and ELI was significantly decreased in bacteria inoculated treatments. Also, MDA, H2O2 were decreased, and antioxidant capacity, proline, and antioxidant enzymes were increased due to inoculation with these bacteria. Besides, the amount of K+ as an index of salinity tolerance significantly increased, and leaf Na+ content was significantly decreased.

Project description:Plant growth regulators (PGRs) and plant growth promoting rhizobacteria (PGPRs) play an important role in mitigating abiotic stresses. However, little is known about the parallel changes in physiological processes coupled with metabolic changes induced by PGRs and PGPRs that help to cope with drought stress in chickpeas. The present investigation was carried out to study the integrative effects of PGRs and PGPRs on the physiological and metabolic changes, and their association with drought tolerance in two chickpea genotypes. Inoculated seeds of two chickpea genotypes, Punjab Noor-2009 (drought sensitive) and 93127 (drought tolerance), were planted in greenhouse condition at the University of Florida. Prior to sowing, seeds of two chickpea varieties were soaked for 3 h in 24 h old cultures of PGPRs (Bacillus subtilis, Bacillus thuringiensis, and Bacillus megaterium), whereas, some of the seeds were soaked in distilled water for the same period of time and were treated as control. Plant growth regulators, salicylic acid (SA) and putrescine (Put), were applied on 25 days old seedlings just prior to the induction of drought stress. Drought stress was imposed by withholding the supply of water on 25-day-old seedlings (at the three-leaf stage) and continued for the next 25 days until the soil water content reached 14%. Ultrahigh-performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS) analysis concomitant with physiological parameters were carried out in chickpea leaves at two-time points i.e. 14 and 25 d after imposition of drought stress. The results showed that both genotypes, treated with PGRs and PGPRs (consortium), performed significantly better under drought condition through enhanced leaf relative water content (RWC), greater biomass of shoot and root, higher Fv/FM ratio and higher accumulation of protein, sugar and phenolic compounds. The sensitive genotype was more responsive than tolerant one. The results revealed that the accumulation of succinate, leucine, disaccharide, saccharic acid and glyceric acid was consistently higher in both genotypes at both time points due to PGRs and PGPRs treatment. Significant accumulation of malonate, 5-oxo-L-proline, and trans-cinnamate occurred at both time points only in the tolerant genotype following the consortium treatment. Aminoacyl-tRNA, primary and secondary metabolite biosynthesis, amino acid metabolism or synthesis pathways, and energy cycle were significantly altered due to PGRs and PGPRs treatment. It is inferred that changes in different physiological and metabolic parameters induced by PGRs and PGPRs treatment could confer drought tolerance in chickpeas.

Project description:BackgroundSoil pollution by heavy metals increases the bioavailability of metals like hexavalent chromium (Cr (VI)), subsequently limiting plant growth and reducing the efficiency of phytoremediation. Plant growth-promoting rhizobacteria (PGPR) have substantial potential to enhance plant growth as well as plant tolerance to metal stress. The aim of this research was to investigate Cr (VI) phytoremediation enhancement by PGPR.ResultsThe results showed that the 27 rhizobacterial isolates studied were confirmed as Cr (VI)-resistant PGPR, by using classical biochemical tests (phosphate solubilization, nitrogen fixation, indole acetic acid, exopolysaccharides, hydrogen cyanide, siderophores, ammonia, cellulase, pectinase, and chitinase production) and showed variable levels of Cr (VI) resistance (300-600 mg/L). The best four selected Cr (VI)-resistant PGPR (NT15, NT19, NT20, and NT27) retained most of the PGP traits in the presence of 100-200 mg/L concentrations of Cr (VI). The inoculation of Medicago sativa with any of these four isolates improved the shoot and root dry weight. The NT27 isolate identified using 16S rDNA gene sequence analyses as a strain of Pseudomonas sp. was most effective in terms of plant growth promotion and stress level decrease. It increased shoot and root dry weights of M. sativa by 97.6 and 95.4%, respectively, in the presence of Cr (VI) when compared to non-inoculated control plants. It also greatly increased chlorophyll content and decreased the levels of stress markers, malondialdehyde, hydrogen peroxide, and proline. The results of the effect of Pseudomonas sp. on Cr content and bioaccumulation factor (BAF) of the shoots and roots of M. sativa plants showed the increase of plant biomass concomitantly with the increase of Cr root concentration in inoculated plants. This would lead to a higher potential of Cr (VI) phytostabilization.ConclusionsThis study demonstrates that the association M. sativa-Pseudomonas sp. may be an efficient biological system for the bioremediation of Cr (VI)-contaminated soils.
