Growth rate and locomotor performance tradeoff is not universal in birds.

ABSTRACT: Though a tradeoff between growth rate and locomotor performance has been proposed, empirical data on this relationship are still limited. Here we statistically analyze the associations of growth rate and flight ability in birds by assessing how growth rate is correlated with three wing parameters of birds: flight muscle ratio, wing aspect ratio, and wing loading. We find that fast-growing birds tended to have higher flight muscle ratios and higher wing loadings than slow-growing birds, which suggests that fast-growing birds may have better takeoff performance, but lower efficiency in maneuvering flight. Accordingly, our findings suggest that the relationship between growth rate and flight ability is more complex than a simple tradeoff. Since the hindlimbs also contribute greatly to the locomotion of birds, future investigations on the relationship between growth rate and hindlimb performance will provide more insights into the evolution of birds.

Tradeoff between lag time and growth rate drives the plasmid acquisition cost.

Project description:Conjugative plasmids drive genetic diversity and evolution in microbial populations. Despite their prevalence, plasmids can impose long-term fitness costs on their hosts, altering population structure, growth dynamics, and evolutionary outcomes. In addition to long-term fitness costs, acquiring a new plasmid introduces an immediate, short-term perturbation to the cell. However, due to the transient nature of this plasmid acquisition cost, a quantitative understanding of its physiological manifestations, overall magnitudes, and population-level implications, remains unclear. To address this, here we track growth of single colonies immediately following plasmid acquisition. We find that plasmid acquisition costs are primarily driven by changes in lag time, rather than growth rate, for nearly 60 conditions covering diverse plasmids, selection environments, and clinical strains/species. Surprisingly, for a costly plasmid, clones exhibiting longer lag times also achieve faster recovery growth rates, suggesting an evolutionary tradeoff. Modeling and experiments demonstrate that this tradeoff leads to counterintuitive ecological dynamics, whereby intermediate-cost plasmids outcompete both their low and high-cost counterparts. These results suggest that, unlike fitness costs, plasmid acquisition dynamics are not uniformly driven by minimizing growth disadvantages. Moreover, a lag/growth tradeoff has clear implications in predicting the ecological outcomes and intervention strategies of bacteria undergoing conjugation.

Adaptive evolution reveals a tradeoff between growth rate and oxidative stress during naphthoquinone-based aerobic respiration.

Project description:Evolution fine-tunes biological pathways to achieve a robust cellular physiology. Two and a half billion years ago, rapidly rising levels of oxygen as a byproduct of blooming cyanobacterial photosynthesis resulted in a redox upshift in microbial energetics. The appearance of higher-redox-potential respiratory quinone, ubiquinone (UQ), is believed to be an adaptive response to this environmental transition. However, the majority of bacterial species are still dependent on the ancient respiratory quinone, naphthoquinone (NQ). Gammaproteobacteria can biosynthesize both of these respiratory quinones, where UQ has been associated with aerobic lifestyle and NQ with anaerobic lifestyle. We engineered an obligate NQ-dependent γ-proteobacterium, Escherichia coli ΔubiC, and performed adaptive laboratory evolution to understand the selection against the use of NQ in an oxic environment and also the adaptation required to support the NQ-driven aerobic electron transport chain. A comparative systems-level analysis of pre- and postevolved NQ-dependent strains revealed a clear shift from fermentative to oxidative metabolism enabled by higher periplasmic superoxide defense. This metabolic shift was driven by the concerted activity of 3 transcriptional regulators (PdhR, RpoS, and Fur). Analysis of these findings using a genome-scale model suggested that resource allocation to reactive oxygen species (ROS) mitigation results in lower growth rates. These results provide a direct elucidation of a resource allocation tradeoff between growth rate and ROS mitigation costs associated with NQ usage under oxygen-replete condition.

Positive effects of fast growth on locomotor performance in pelagic fish juveniles.

Project description:Many laboratory experiments on aquatic vertebrates that inhabit closed water or coastal areas have highlighted negative effects of fast growth on swimming performance. Nonetheless, field studies on pelagic fishes have provided evidence of survival advantages of faster-growing individuals. To reconcile this contradiction, we examined the relationship between growth rate and swimming performance as a continuous function for juveniles of chub mackerel (Scomber japonicus) using 3D tracking analysis. For experiments, 20, 24, 27, and 30 days post-hatch individuals within the size range of 14.5-25.3 mm were used. We found that the growth-swimming (burst speed) relationship in chub mackerel was substantially positive and it was supported by morphological traits such as muscle area, which were also positively related with growth rate. This finding is consistent with field observations showing selective survival of fast-growing individuals of this species, reconciling the current contradiction between laboratory experiments and field observations. A dome-shaped quadratic curve described the relationship between growth rate and burst speed better than a linear or cubic function, suggesting that growth may trade-off with swimming performance, as reported in many previous studies, when it is extremely fast. These results, obtained from the rarely tested offshore species, strongly suggests the importance of experimental verification using animals that inhabit various types of habitats in understanding the principles underlying the evolution of growth-locomotor relationship.

Pedal to the metal: Cities power evolutionary divergence by accelerating metabolic rate and locomotor performance.

Project description:Metabolic rates of ectotherms are expected to increase with global trends of climatic warming. But the potential for rapid, compensatory evolution of lower metabolic rate in response to rising temperatures is only starting to be explored. Here, we explored rapid evolution of metabolic rate and locomotor performance in acorn-dwelling ants (Temnothorax curvispinosus) in response to urban heat island effects. We reared ant colonies within a laboratory common garden (25°C) to generate a laboratory-born cohort of workers and tested their acute plastic responses to temperature. Contrary to expectations, urban ants exhibited a higher metabolic rate compared with rural ants when tested at 25°C, suggesting a potentially maladaptive evolutionary response to urbanization. Urban and rural ants had similar metabolic rates when tested at 38°C, as a consequence of a diminished plastic response of the urban ants. Locomotor performance also evolved such that the running speed of urban ants was faster than rural ants under warmer test temperatures (32°C and 42°C) but slower under a cooler test temperature (22°C). The resulting specialist-generalist trade-off and higher thermal optimum for locomotor performance might compensate for evolved increases in metabolic rate by allowing workers to more quickly scout and retrieve resources.

Consequences of Fatherhood in the Biparental California Mouse (Peromyscus californicus): Locomotor Performance, Metabolic Rate, and Organ Masses.

Project description:Although effects of motherhood on mothers have been well documented in mammals, the effects of fatherhood on fathers are not well known. We evaluated effects of being a father on key metabolic and performance measures in the California mouse, Peromyscus californicus. California mice are genetically monogamous in the wild, and fathers show similar parental behavior to mothers, with the exception of lactation. To investigate the impact of fatherhood on fathers, focal males were paired with an intact female (breeding males), a tubally ligated female (nonbreeding males), or another male (virgins). Starting 3-5 d after the birth of each breeding pair's first litter, males were tested for locomotor performance (maximum sprint speed, treadmill endurance), basal metabolic rate (BMR), and maximum oxygen consumption ([Formula: see text]). At the end of the 11-d test period, mice were euthanized, hematocrit was determined, and organs were weighed. Speed, endurance, and [Formula: see text] were significantly repeatable between two replicate measurement days but did not differ among groups, nor did BMR. Breeding males had significantly larger hind limb muscles than did nonbreeding males, whereas virgin males had heavier subcutaneous fat pads than did nonbreeding and breeding males. Several correlations were observed at the level of individual variation (residuals from ANCOVA models), including positive correlations for endurance with [Formula: see text], [Formula: see text] with testes mass, and some of the digestion-related organs with each other. These results indicate that fatherhood may not have pronounced performance, metabolic, or morphological effects on fathers, at least under standard laboratory conditions and across a single breeding cycle.

Universal roles of hydrogen in electrochemical performance of graphene: high rate capacity and atomistic origins.

Project description:Atomic hydrogen exists ubiquitously in graphene materials made by chemical methods. Yet determining the effect of hydrogen on the electrochemical performance of graphene remains a significant challenge. Here we report the experimental observations of high rate capacity in hydrogen-treated 3-dimensional (3D) graphene nanofoam electrodes for lithium ion batteries. Structural and electronic characterization suggests that defect sites and hydrogen play synergistic roles in disrupting sp(2) graphene to facilitate fast lithium transport and reversible surface binding, as evidenced by the fast charge-transfer kinetics and increased capacitive contribution in hydrogen-treated 3D graphene. In concert with experiments, multiscale calculations reveal that defect complexes in graphene are prerequisite for low-temperature hydrogenation, and that the hydrogenation of defective or functionalized sites at strained domain boundaries plays a beneficial role in improving rate capacity by opening gaps to facilitate easier Li penetration. Additional reversible capacity is provided by enhanced lithium binding near hydrogen-terminated edge sites. These findings provide qualitative insights in helping the design of graphene-based materials for high-power electrodes.

Laboratory evolution reveals a two-dimensional rate-yield tradeoff in microbial metabolism.

Project description:Growth rate and yield are fundamental features of microbial growth. However, we lack a mechanistic and quantitative understanding of the rate-yield relationship. Studies pairing computational predictions with experiments have shown the importance of maintenance energy and proteome allocation in explaining rate-yield tradeoffs and overflow metabolism. Recently, adaptive evolution experiments of Escherichia coli reveal a phenotypic diversity beyond what has been explained using simple models of growth rate versus yield. Here, we identify a two-dimensional rate-yield tradeoff in adapted E. coli strains where the dimensions are (A) a tradeoff between growth rate and yield and (B) a tradeoff between substrate (glucose) uptake rate and growth yield. We employ a multi-scale modeling approach, combining a previously reported coarse-grained small-scale proteome allocation model with a fine-grained genome-scale model of metabolism and gene expression (ME-model), to develop a quantitative description of the full rate-yield relationship for E. coli K-12 MG1655. The multi-scale analysis resolves the complexity of ME-model which hindered its practical use in proteome complexity analysis, and provides a mechanistic explanation of the two-dimensional tradeoff. Further, the analysis identifies modifications to the P/O ratio and the flux allocation between glycolysis and pentose phosphate pathway (PPP) as potential mechanisms that enable the tradeoff between glucose uptake rate and growth yield. Thus, the rate-yield tradeoffs that govern microbial adaptation to new environments are more complex than previously reported, and they can be understood in mechanistic detail using a multi-scale modeling approach.

Which muscles compromise human locomotor performance with age?

Project description:Ageing leads to a progressive decline in human locomotor performance. However, it is not known whether this decline results from reduced joint moment and power generation of all lower limb muscle groups or just some of them. To further our understanding of age-related locomotor decline, we compare the amounts of joint moments and powers generated by lower limb muscles during walking (self-selected), running (4 m s(-1)) and sprinting (maximal speed) among young, middle-aged and old adults. We find that age-related deficit in ankle plantarflexor moment and power generation becomes more severe as locomotion change from walking to running to sprinting. As a result, old adults generate more power at the knee and hip extensors than their younger counterparts when walking and running at the same speed. During maximal sprinting, young adults with faster top speeds demonstrate greater moments and powers from the ankle and hip joints, but interestingly, not from the knee joint when compared with the middle-aged and old adults. These findings indicate that propulsive deficit of ankle contributes most to the age-related locomotor decline. In addition, reduced muscular output from the hip rather than from knee limits the sprinting performance in older age.

Universal mechanisms of sound production and control in birds and mammals.

Project description:As animals vocalize, their vocal organ transforms motor commands into vocalizations for social communication. In birds, the physical mechanisms by which vocalizations are produced and controlled remain unresolved because of the extreme difficulty in obtaining in vivo measurements. Here, we introduce an ex vivo preparation of the avian vocal organ that allows simultaneous high-speed imaging, muscle stimulation and kinematic and acoustic analyses to reveal the mechanisms of vocal production in birds across a wide range of taxa. Remarkably, we show that all species tested employ the myoelastic-aerodynamic (MEAD) mechanism, the same mechanism used to produce human speech. Furthermore, we show substantial redundancy in the control of key vocal parameters ex vivo, suggesting that in vivo vocalizations may also not be specified by unique motor commands. We propose that such motor redundancy can aid vocal learning and is common to MEAD sound production across birds and mammals, including humans.

Predation changes the shape of thermal performance curves for population growth rate.

Project description:Ectotherms generally demonstrate nonlinear changes in performance (e.g., movement speed, individual growth, population growth) as a function of temperature that are characterized by thermal performance curves (TPC). Predation risk elicits phenotypic and behavioral changes that likewise impact performance measures. We tested whether exposure to predation Orthocyclops modestus impacts the maximum population growth rate (rmax) TPC of the protist Paramecium aurelia. We fit predator and non-predator exposed P. aurelia population growth rates to a function previously shown to best describe Paramecium population growth rate TPC's (Lactin-2) and compared subsequent parameter estimates between curves. For Paramecium exposed to predation risk, maximum population growth increased more rapidly as temperatures rose and decreased more rapidly as temperatures fell compared to the initial temperature. The area under each TPC curve remained approximately the same, consistent with the idea of a trade-off in performance across temperatures. Our results indicate TPCs are flexible given variation in food web context and that trophic interactions may play an important role in shaping TPCs. Furthermore, this and other studies illustrate the need for a mechanistic model of TPCs with parameters tied to biologically meaningful properties.
