Ethanol and lactic acid production from sugar and starch wastes by anaerobic acidification.

ABSTRACT: Anaerobic conversion of carbohydrates can generate various end-products. Besides physical parameters such as pH and temperature, the types of carbohydrate being fermented influences the fermentation pattern. Under uncontrolled pH, microbial mixed cultures from activated sludge and anaerobic digester sludge anaerobically produced ethanol from glucose while producing lactic acid from starch conversion. This trend was not only observed in batch trials. Also, continuous chemostat operation of anaerobic digester sludge resulted in the reproducible predominance of ethanol fermentation from glucose solution and lactic acid production from starch. Different feeding regimes and substrate availability (shock load versus continuous feeding) in glucose fermentation under non-controlled pH did not affect the ethanol production as the major end product. Shifts in feed composition from glucose to starch and vice versa result in an immediate change of fermentation end products formation.

Value Proposition of Untapped Wet Wastes: Carboxylic Acid Production through Anaerobic Digestion.

Project description:Although traditional anaerobic digestion (AD) process to produce methane-rich biogas from wet waste is deep-rooted, high carbon footprint and its low value as compared with other renewable sources demand advanced strategies to avoid its production. An emerging conversion pathway to arrest methanogenesis for producing value-added fuels and chemicals instead of biogas is sought as a sustainable alternative. This research provides a comprehensive analysis on current technology development, process challenges, applications, and economics for producing high-value short-chain carboxylic acids from AD of wet wastes. We show that (1) the theoretical energy yields of acids equal or exceed biogas, and (2) the cost of these acids is competitive with those produced from chemical markets, making this economically viable for mass production. With global abundance of wet waste feedstocks, this process of short-chain acid production provides a promising alternative to conventional biogas production technology, while achieving waste management and carbon mitigation goals.

Simultaneous reuse and treatment of sugar-sweetened beverage wastes for citric acid production.

Project description:This study aimed to evaluate the feasibility of using sugar-sweetened beverages (SSB) for citric acid (CA) production and its impact on chemical oxygen demand (COD) of SSB. Five types of SSB were used as a carbon source for CA production by A. niger, and the COD of each SSB was measured before and after the bioprocess. Results showed that all tested SSB were suitable for CA production, with maximum yields ranging from 13.01 to 56.62 g L- 1. The COD was reduced from 53 to 75.64%, indicating that the bioprocess effectively treated SSB wastes. The use of SSB as a substrate for CA production provides an alternative to traditional feedstocks, such as sugarcane and beet molasses. The low-cost and high availability of SSB makes it an attractive option for CA production. Moreover, the study demonstrated the potential of the bioprocess to simultaneously treat and reuse SSB wastes, reducing the environmental impact of the beverage industry.Supplementary informationThe online version contains supplementary material available at 10.1007/s13197-023-05761-9.

Granular activated carbon enhances volatile fatty acid production in the anaerobic fermentation of garden wastes.

Project description:Garden waste, one type of lignocellulosic biomass, holds significant potential for the production of volatile fatty acids (VFAs) through anaerobic fermentation. However, the hydrolysis efficiency of garden waste is limited by the inherent recalcitrance, which further influences VFA production. Granular activated carbon (GAC) could promote hydrolysis and acidogenesis efficiency during anaerobic fermentation. This study developed a strategy to use GAC to enhance the anaerobic fermentation of garden waste without any complex pretreatments and extra enzymes. The results showed that GAC addition could improve VFA production, especially acetate, and reach the maximum total VFA yield of 191.55 mg/g VSadded, which increased by 27.35% compared to the control group. The highest VFA/sCOD value of 70.01% was attained in the GAC-amended group, whereas the control group only reached 49.35%, indicating a better hydrolysis and acidogenesis capacity attributed to the addition of GAC. Microbial community results revealed that GAC addition promoted the enrichment of Caproiciproducens and Clostridium, which are crucial for anaerobic VFA production. In addition, only the GAC-amended group showed the presence of Sphaerochaeta and Oscillibacter genera, which are associated with electron transfer processes. Metagenomics analysis indicated that GAC addition improved the abundance of glycoside hydrolases (GHs) and key functional enzymes related to hydrolysis and acidogenesis. Furthermore, the assessment of major genera influencing functional genes in both groups indicated that Sphaerochaeta, Clostridium, and Caproicibacter were the primary contributors to upregulated genes. These findings underscored the significance of employing GAC to enhance the anaerobic fermentation of garden waste, offering a promising approach for sustainable biomass conversion and VFA production.

Lactic acid production from food waste at an anaerobic digestion biorefinery: effect of digestate recirculation and sucrose supplementation.

Project description:Low lactic acid (LA) yields from direct food waste (FW) fermentation restrict this production pathway. However, nitrogen and other nutrients within FW digestate, in combination with sucrose supplementation, may enhance LA production and improve feasibility of fermentation. Therefore, this work aimed to improve LA fermentation from FWs by supplementing nitrogen (0-400 mgN·L-1) as NH4Cl or digestate and dosing sucrose (0-150 g·L-1) as a low-cost carbohydrate. Overall, NH4Cl and digestate led to similar improvements in the rate of LA formation (0.03 ± 0.02 and 0.04 ± 0.02 h-1 for NH4Cl and digestate, respectively), but NH4Cl also improved the final concentration, though effects varied between treatments (5.2 ± 4.6 g·L-1). While digestate altered the community composition and increased diversity, sucrose minimised community diversion from LA, promoted Lactobacillus growth at all dosages, and enhanced the final LA concentration from 25 to 30 g·L-1 to 59-68 g·L-1, depending on nitrogen dosage and source. Overall, the results highlighted the value of digestate as a nutrient source and sucrose as both community controller and means to enhance the LA concentration in future LA biorefinery concepts.

Lignocellulosic ethanol production by starch-base industrial yeast under PEG detoxification.

Project description:Cellulosic ethanol production from lignocellulosic biomass offers a sustainable solution for transition from fossil based fuels to renewable alternatives. However, a few long-standing technical challenges remain to be addressed in the development of an economically viable fermentation process from lignocellulose. Such challenges include the needs to improve yeast tolerance to toxic inhibitory compounds and to achieve high fermentation efficiency with minimum detoxification steps after a simple biomass pretreatment. Here we report an in-situ detoxification strategy by PEG exo-protection of an industrial dry yeast (starch-base). The exo-protected yeast cells displayed remarkably boosted vitality with high tolerance to toxic inhibitory compounds, and with largely improved ethanol productivity from crude hydrolysate derived from a pretreated lignocellulose. The PEG chemical exo-protection makes the industrial S. cerevisiae yeast directly applicable for the production of cellulosic ethanol with substantially improved productivity and yield, without of the need to use genetically modified microorganisms.

Simultaneous Saccharification and Fermentation of Sugar Beet Pulp with Mixed Bacterial Cultures for Lactic Acid and Propylene Glycol Production.

Project description:Research into fermentative production of lactic acid from agricultural by-products has recently concentrated on the direct conversion of biomass, whereby pure sugars are replaced with inexpensive feedstock in the process of lactic acid production. In our studies, for the first time, the source of carbon used is sugar beet pulp, generated as a by-product of industrial sugar production. In this paper, we focus on the simultaneous saccharification of lignocellulosic biomass and fermentation of lactic acid, using mixed cultures with complementary assimilation profiles. Lactic acid is one of the primary platform chemicals, and can be used to synthesize a wide variety of useful products, including green propylene glycol. A series of controlled batch fermentations was conducted under various conditions, including pretreatment with enzymatic hydrolysis. Inoculation was performed in two sequential stages, to avoid carbon catabolite repression. Biologically-synthesized lactic acid was catalytically reduced to propylene glycol over 5% Ru/C. The highest lactic acid yield was obtained with mixed cultures. The yield of propylene glycol from the biological lactic acid was similar to that obtained with a water solution of pure lactic acid. Our results show that simultaneous saccharification and fermentation enables generation of lactic acid, suitable for further chemical transformations, from agricultural residues.

Maize Milling By-Products: From Food Wastes to Functional Ingredients Through Lactic Acid Bacteria Fermentation.

Project description:Although recognized as important sources of functional compounds, milling by-products are often removed from the cereal kernel prior milling process. Indeed, the high presence of fiber in bran and the co-presence of lipids and lipase in germ are often considered as downsides for breadmaking. In this work, Lactobacillus plantarum T6B10 and Weissella confusa BAN8 were used as selected starters to ferment maize milling by-products mixtures made with heat-treated or raw germ and bran. The effects on the biochemical and nutritional features as well as the stability of the milling by-products were investigated. Lactic acid bacteria metabolisms improved the free amino acids and peptides concentrations and the antioxidant activity and caused a relevant phytic acid degradation. Moreover, fermentation allowed a marked decrease of the lipase activity, stabilizing the matrix by preventing oxidative processes. The use of fermented by-products as ingredients improved the nutritional, textural and sensory properties of wheat bread. Fortified breads (containing 25% of fermented by-products) were characterized by a concentration in dietary fiber and proteins of ca. 11 and 13% of dry matter, respectively. Compared to the use of the unfermented ones, the addition of pre-fermented by-products to bread caused a significant increase in protein digestibility (up to 60%), and a relevant decrease of the starch hydrolysis index (ca. 13%). According to the results, this study demonstrates the potential of fermentation to convert maize bran and germ, commonly considered food wastes, into nutritive improvers, meeting nutritional and sensory requests of modern consumers.
