Bulky 2,6-disubstituted aryl siloxanes and a disilanamine.

ABSTRACT: The crystal structures of 5-bromo-1,3-di-tert-butyl-2-[(tri-methyl-sil-yl)-oxy]benzene, C17H29BrOSi, (I), 1,3-di-tert-butyl-2-[(tri-methyl-sil-yl)-oxy]benzene, C17H30OSi, (II), and N-(2,6-diiso-propyl-phen-yl)-1,1,1-trimethyl-N-(tri-methyl-sil-yl)silanamine, C18H35NSi2, (III), are reported. Compound (I) crystallizes in space group P21/c with Z' = 1, (II) in Pnma with Z' = 0.5 and (III) in Cmcm with Z' = 0.25. Consequently, the mol-ecules of (II) are constrained by m and those of (III) by m2m site symmetries. Despite this, both (I) and (II) are distorted towards mild boat conformations, as is typical of 2,6-di-tert-butyl-substituted phenyl compounds, reflecting the high local steric pressure of the flanking alkyl groups. Compound (III) by contrast is planar and symmetric, and this lack of distortion is compatible with the lower steric pressure of the flanking 2,6-diisopropyl substituents.

Project description:A concise, modular, asymmetric synthesis of cis-2,6-disubstituted piperazines from readily available amino acid precursors is described. The key step in the synthesis is a Pd-catalyzed carboamination of a N1-aryl-N2-allyl-1,2-diamine with an aryl bromide. The products are obtained in 14-20:1 dr, with >97% ee, and the key cyclizations are the first examples of six-membered ring formation via Pd-catalyzed carboamination reactions of unsaturated amines with aryl halides.

Project description:New substituted [30]trithiadodecaazahexaphyrines (hemihexaphyrazines) were synthesized by a crossover condensation of 2,5-diamino-1,3,4-thiadiazole with 4-chloro-5-(2,6-diisopropylphenoxy)- or 4,5-bis-(2,6-diisopropylphenoxy)phthalonitriles. The compounds were characterized by 1H-, 13C-NMR, including COSY, HMBC, and HSQC spectroscopy, MALDI TOF spectrometry, elemental analysis, IR and UV-Vis absorbance and fluorescence techniques.

Synthesis of 2,6-trans-disubstituted 5,6-dihydropyrans from (Z)-1,5-syn-endiols.

Project description:Certain (Z)-1,5-syn-diols 2 may be converted into 2,6-trans-5,6-dihydropyrans by using phosphonium salt 4 or phosphorane 5 as dehydrating agents. A more general four step procedure converts the (Z)-1,5-syn-endiols into enantiomeric dihydropyrans ent-3 via regioselective silylation of the allylic alcohol unit followed by mesylate formation and base-promoted nucleophilic displacement.

Reactivity Modes of Cp*M-Type Half-Sandwich Dichalcogenolate Complexes with 2,6-Disubstituted Aryl Azides: The Effects of the Metal Center, Chalcogen, and Ligand Moiety on Product Formation.

Project description:Cp*M-type half-sandwich dichalcogenolate complexes bearing either carborane or benzene moieties show diverse reactivity patterns toward two selected 2,6-disubstituted aryl azides under thermal or photolytic conditions. The chalcogen (S and Se) has little effect on the formation of final products. However, the effects of both the metal center and the ligand moiety of the metal precursor on the reactions were significant. Compared to iridium precursor Cp*IrS2C2B10H10 (1a), rhodium and cobalt analogues (1b: Cp*RhS2C2B10H10, 1c: Cp*CoS2C2B10H10) demonstrated no reactivity toward aryl azides. The reaction of Cp*IrSe2C2B10H10 (1d) with 2,6-Me2C6H3N3 led to the clean formation of complex 2 with C(sp3)-H activation of one methyl group of the Cp* ligand and loss of N2 along with the rearrangement of the benzene ring of the original azide ligand, whereas the treatment of Cp*IrS2C6H4 (1e) with 2,6-Me2C6H3N3 under the same reaction conditions gave a 16-electron half-sandwich complex 5 featuring C-N coupling on one methyl group from the Cp* ligand. When 2-Me-6-NO2C6H3N3 was employed, the same reaction patterns for forming products (3 and 6) with the nitro group migrating to the para-position versus the original aryl azide were observed. In addition, the reaction with metal precursor 1d generated another product 4 featuring the exchange of nitro and azido groups, while the reaction with 1e afforded another complex 7 with the formation of the N-NO2 moiety. All new complexes were characterized by spectroscopy methods, and single-crystal X-ray analyses were performed for complexes 2 and 5-7. Furthermore, radical mechanisms for the formation of complexes 2-7 were proposed.

Ligand-controlled cobalt-catalyzed regiodivergent hydroboration of aryl,alkyl-disubstituted internal allenes.

Project description:We report a stereoselective regiodivergent hydroboration of aryl,alkyl-disubstituted internal allenes with pinacolborane (HBpin) in the presence of cobalt catalysts generated from bench-stable Co(acac)2 and bisphosphine ligands. An interesting correlation between the regioselectivity of this hydroboration and the bite angles of bisphosphine ligands was identified. When hydroboration was conducted with cobalt catalysts containing bisphosphines with medium bite angles (e.g. 98° for dppb and 96° for dppf), HBpin was selectively added to the alkyl-substituted double bond. However, HBpin was selectively added to the aryl-substituted double bond when the reactions were conducted with cobalt catalysts containing bisphosphines with large bite angles (e.g. 111° for xantphos and 114° for Nixantphos). A range of internal allenes underwent these Co-catalyzed hydroboration reactions in a regiodivergent manner to yield the corresponding (Z)-alkenylboronates in high isolated yields and with high regioselectivity. These reactions show good functional group compatibility and can be readily scaled up to gram scales without using a dry box. In addition, the comparison of regioselectivity between the Co-catalyzed hydrosilylation and hydroboration reactions of the same allene substrate suggests that this Co-catalyzed regiodivergent hydroboration of allenes proceeds through a Co-Bpin intermediate. Deuterium-labeling experiments suggest that the Co-Bpin intermediates react with allenes to form allylcobalt species which then react with HBpin to release (Z)-alkenylboronate products.

Lateral Metallation and Redistribution Reactions of Sodium Ferrates Containing Bulky 2,6-Diisopropyl-N-(trimethylsilyl)anilide Ligands.

Project description:Alkali-metal ferrates containing amide groups have emerged as regioselective bases capable of promoting Fe-H exchanges of aromatic substrates. Advancing this area of heterobimetallic chemistry, a new series of sodium ferrates is introduced incorporating the bulky arylsilyl amido ligand N(SiMe3 )(Dipp) (Dipp=2,6-iPr2 -C6 H3 ). Influenced by the large steric demands imposed by this amide, transamination of [NaFe(HMDS)3 ] (HMDS=N(SiMe3 )2 ) with an excess of HN(SiMe3 )(Dipp) led to the isolation of heteroleptic [Na(HMDS)2 Fe{N(SiMe3 )Dipp}]∞ (1) resulting from the exchange of just one HMDS group. An alternative co-complexation approach, combining the homometallic metal amides [NaN(SiMe3 )Dipp] and [Fe{N(SiMe3 )Dipp}2 ] induces lateral metallation of one Me arm from the SiMe3 group in the iron amide furnishing tetrameric [NaFe{N(SiCH2 Me2 )Dipp}{N(SiMe3 )Dipp}]4 (2). Reactivity studies support that this deprotonation is driven by the steric incompatibility of the single metal amides rather than the basic capability of the sodium reagent. Displaying synergistic reactivity, heteroleptic sodium ferrate 1 can selectively promote ferration of pentafluorobenzene using one of its HMDS arms to give heterotrileptic [Na{N(SiMe3 )Dipp}(HMDS)Fe(C6 F5 )]∞ (4). Attempts to deprotonate less activated pyridine led to the isolation of NaHMDS and heteroleptic Fe(II) amide [(py)Fe{N(SiMe3 )Dipp}(HMDS)] (5), resulting from an alternative redistribution process which is favoured by the Lewis donor ability of this substrate.

Discovery of a Novel 2,6-Disubstituted Glucosamine Series of Potent and Selective Hexokinase 2 Inhibitors.

Project description:A novel series of potent and selective hexokinase 2 (HK2) inhibitors, 2,6-disubstituted glucosamines, has been identified based on HTS hits, exemplified by compound 1. Inhibitor-bound crystal structures revealed that the HK2 enzyme could adopt an "induced-fit" conformation. The SAR study led to the identification of potent HK2 inhibitors, such as compound 34 with greater than 100-fold selectivity over HK1. Compound 25 inhibits in situ glycolysis in a UM-UC-3 bladder tumor cell line via (13)CNMR measurement of [3-(13)C]lactate produced from [1,6-(13)C2]glucose added to the cell culture.
