A new species of Phoebe (Lauraceae) from south-western China.

ABSTRACT: Here Phoebe hekouensis Bing Liu, W.Y. Jin, L.N. Zhao & Y. Yang from south-eastern Yunnan Province of China is described as new to science. This species is morphologically similar to P. megacalyx H.W. Li in the twigs being robust and brownish tomentose, the ovary densely pubescent and the tepals longer than 1 cm, but differs from the latter species by the leaves being broader, up to 18 cm (vs. 4.5-11.5 cm), the inflorescences shorter, ca. 10-15 cm long (vs. up to 23 cm), the ovary completely and densely pubescent (vs. pubescent only at the apical portion) and the stigma conspicuous (vs. inconspicuous). The new species also resembles P. macrocarpa C.Y. Wu, but differs from the latter by the tepals being longer, 9-13 mm long (vs. ca. 4 mm).

Complete chloroplast genome sequences of two endangered Phoebe (Lauraceae) species.

Project description:BACKGROUND:Phoebe (Lauraceae) comprises of evergreen trees or shrubs with approximately 100 species, distributed in tropical and subtropical Asia and Neotropical America. A total of 34 species and three varieties occur in China. Despite of economic and ecological value, only limited genomic resources are available for this genus. RESULTS:We sequenced the two complete chloroplast (cp) genomes of Phoebe chekiangensis and P. bournei using Illumina sequencing technology via a combined strategy of de novo and reference-guided assembly. We also performed comparative analyses with the cp genomes of P. sheareri and P. sheareri var. oineiensis previously reported. The chloroplast genomes of P. chekiangensis and P. bournei identically contain 112 genes consisting of 78 protein coding genes, 30 tRNA genes, and 4 rRNA genes, with the size of 152,849 and 152,853 bp, respectively. From the two chloroplast genomes, 131 SSRs were identified and 12 different SSRs located in five protein coding genes. The analysis showed the extremely conserved structure of chloroplast genomes with surprisingly little variations at the LSC/IR and SSC/IR boundaries. Moreover, the mean nucleotide diversity was found to be 0.162% for 77 regions, suggesting an extraordinarily low level of sequence divergence. Four highest divergent regions (trnH-psbA, rps14-trnT, petA-psbJ, ccsA-ndhD) with the percentage of nucleotide diversity higher than 0.50% were identified, which had potential use for species identification and phylogenetic studies. CONCLUSION:This study will facilitate our understanding of population genetics, phylogenetic relationship and plant evolution of Phoebe species.

Diaporthe species in south-western China.

Project description:Three strains of the genus Diaporthe were isolated from different plant hosts in south-western China. Phylogenetic analyses of the combined ITS, ?-tubulin, tef1 and calmoudulin dataset indicated that these strains represented three independent lineages in Diaporthe. Diaporthe millettiae sp. nov. clustered with D. hongkongensis and D. arecae, Diaporthe osmanthi sp. nov. grouped with D. arengae, D. pseudomangiferae and D. perseae and Diaporthe strain GUCC9146, isolated from Camellia sinensis, was grouped in the D. eres species complex with a close relationship to D. longicicola. These species are reported with taxonomic descriptions and illustrations.

Hoya pyrifolia (Apocynaceae), a new species from south-western Yunnan, China.

Project description:Hoya pyrifolia, a new species of Apocynaceae from Yunnan Province, China, is described and illustrated. Results from phylogenetic analyses, based on combined DNA fragments of the nuclear ribosomal external transcribed spacer (ETS), intergeneric transcribed spacer (ITS) and three plastid DNA fragments (matK, psbA-trnH and trnT-trnL), showed that the new species was nested within a clade, including Hoya species distributed in the subtropical foothills of the Himalayas and the Tibet-Sichuan Plateau. Morphologically, the new species can be distinguished from its close relatives by its pyriform and slightly pubescent leaves, as well as the 4-flowered inflorescences.

A new species and a new series of Elatostema (Urticaceae) from south-western China.

Project description:The new series ElatostemasectionWeddellia series Xingyiensia L.D. Duan & D.H. Yin (Urticaceae) is described. In addition, its new species Elatostemaxingyiense L.D. Duan & D.H. Yin, endemic to Guizhou Province, is also described and illustrated with photographs. The new series is morphologically similar to series Melanocarpa W.T. Wang and series Sublinearia W.T. Wang. The new species is most similar to E.melanocarpum, E.sublineare, E.obscurinerve, E.langicuspe and E.youyangense in morphology, but can be visibly distinguished by a combination of characters, including leaf vein, male inflorescences, female inflorescences and persistent tepals.

Project description:In this paper, three new species of eriophyoid mites in the family Eriophyidae associated with Phoebe hunanensis Hand.-Mazz. (Lauraceae), namely Gammaphytoptus striatilobus sp. n., Phyllocoptes setalsolenidion sp. n., and Dechela phoebe sp. n. are described and illustrated. All are vagrants causing no apparent damage to the same host plants.

Two new species of Microdochium from Indocalamus longiauritus in south-western China.

Project description:Microdochium species have often been reported as plant pathogens and saprophytes and are commonly isolated from some diseased plant hosts. The primary aim of the present study was to describe and illustrate two new Microdochium species isolated from the leaf spot of Indocalamus longiauritus in Yunnan Province, China, namely Microdochium yunnanense and M. indocalami, spp. nov., based on their morphology and multilocus phylogenetic analyses of the combined ITS, LSU, TUB2, and RPB2. DNA sequence data indicate that six strains represent three independent groups from related and similar species in Microdochium. Microdochium indocalami sp. nov. clustered with M. fisheri, M. lycopodinum, M. rhopalostylidis, and M. phragmitis. Microdochium yunnanense sp. nov. grouped with M. bolleyi. In addition, the strain SAUCC1017 is recorded as an unidentified species in Microdochium. Descriptions and illustrations of the new species in the genus and Microdochium sp. indet. are provided.

Big fruits with tiny tepals: An unusual new species of Lauraceae from southwestern China.

Project description:We collected an unusual new plant of Phoebe (Lauraceae) from southeastern Yunnan, China, which possesses more or less oblong leaves, paniculate inflorescences with strictly opposite lateral cymes, trimerous flowers with 4-locular stamens, and large fruits with tiny, equal, persistent tepals. Our molecular phylogenetic study based on nrITS, LEAFY and plastid matK sequences suggests that this species belongs to a clade of Phoebe including P. puwenensis, P. megacalyx, and P. macrocarpa. However, this species differs from the latter three species by subglabrous twigs, leaves and inflorescences (vs. pubescent twigs, leaves and inflorescences in the latter three species), larger fruits (5-8 cm long vs. 1-4 cm long in the latter three species), and smaller tepals (1-2.5 mm long vs. 5-15 mm long in the latter two species). As a result, Phoebe jinpingensis sp. nov. is described and illustrated here as new to science.

Morphological and molecular identification of Diaporthe species in south-western China, with description of eight new species.

Project description:Diaporthe species have often been reported as plant pathogens, endophytes and saprophytes, commonly isolated from a wide range of infected plant hosts. In the present study, twenty strains obtained from leaf spots of twelve host plants in Yunnan Province of China were isolated. Based on a combination of morphology, culture characteristics and multilocus sequence analysis of the rDNA internal transcribed spacer region (ITS), translation elongation factor 1-α (TEF), β-tubulin (TUB), calmodulin (CAL), and histone (HIS) genes, these strains were identified as eight new species: Diaporthe camelliae-sinensis, D. grandiflori, D. heliconiae, D. heterostemmatis, D. litchii, D. lutescens, D. melastomatis, D. pungensis and two previously described species, D. subclavata and D. tectonendophytica. This study showed high species diversity of Diaporthe in tropical rain forests and its hosts in south-western China.

Miocene diversification of a golden-thread nanmu tree species (Phoebe zhennan, Lauraceae) around the Sichuan Basin shaped by the East Asian monsoon.

Project description:Understanding the role of climate changes and geography as drivers of population divergence and speciation is a long-standing goal of evolutionary biology and can inform conservation. In this study, we used restriction site-associated DNA sequencing (RAD-seq) to evaluate genetic diversity, population structure, and infer demographic history of the endangered tree, Phoebe zhennan which is distributed around the Sichuan Basin. Genomic patterns revealed two distinct clusters, each largely confined to the West and East. Despite sympatry of the two genomic clusters at some sites, individuals show little or no evidence of genomic introgression. Demographic modeling supported an initial divergence time between the West and East lineages at ~15.08 Ma with further diversification within the West lineage at ~7.12 Ma. These times largely coincide with the two independent intensifications of the East Asian monsoon that were initiated during the middle (Langhian) and late Miocene (Messinian), respectively. These results suggest that the Miocene intensification phases of the East Asian monsoon played a pivotal role in shaping the current landscape-level patterns of genetic diversity within P. zhennan, as has been found for the interspecific divergence of other subtropical Chinese plants. Based on isolation-by-distance and species distribution modeling, we hypothesize that P. zhennan followed a ring diversification which was facilitated by the Sichuan Basin acting as barrier to gene flow. In situ and ex situ conservation management plans should consider the results obtained in this study to help secure the future of this beautiful and culturally significant endangered tree.
