Various pathways can repair DNA alkylation by chemotherapeutic agents such as temozolomide (TMZ). The enzyme O<sup>6</sup>-methylguanine methyltransferase (MGMT) removes O<sup>6</sup>-methylated DNA adducts, leading to the failure of chemotherapy in resistant glioblastomas.
