Causes and Consequences of Snake Venom Variation.

ABSTRACT: Snake venoms are mixtures of toxins that vary extensively between and within snake species. This variability has serious consequences for the management of the world's 1.8 million annual snakebite victims. Advances in 'omic' technologies have empowered toxinologists to comprehensively characterize snake venom compositions, unravel the molecular mechanisms that underpin venom variation, and elucidate the ensuing functional consequences. In this review, we describe how such mechanistic processes have resulted in suites of toxin isoforms that cause diverse pathologies in human snakebite victims and we detail how variation in venom composition can result in treatment failure. Finally, we outline current therapeutic approaches designed to circumvent venom variation and deliver next-generation treatments for the world's most lethal neglected tropical disease.

Project description:Elucidating the molecular mechanisms underlying snake venom variability provides important clues for understanding how the biological functions of this powerful toxic arsenal evolve. We analyzed in detail individual transcripts and venom protein isoforms produced by five specimens of a venomous snake (Bothrops atrox) from two nearby but genetically distinct populations from the Brazilian Amazon rainforest which show functional similarities in venom properties. Individual variation was observed among the venoms of these specimens, but the overall abundance of each general toxin family was conserved both in transcript and in venom protein levels. However, when expression of independent paralogues was analyzed, remarkable differences were observed within and among each toxin group, both between individuals and between populations. Transcripts for functionally essential venom proteins (“core function” proteins) were highly expressed in all specimens and showed similar transcription/translation rates. In contrast, other paralogues (“adaptive” proteins) showed lower expression levels and the toxins they coded for varied among different individuals. These results provide support for the inferences that (a) expression and translational differences play a greater role in defining adaptive variation in venom phenotypes than does sequence variation in protein coding genes and (b) convergent adaptive venom phenotypes can be generated through different molecular mechanisms. Significance: Analysis of individual transcripts and venom protein isoforms produced by specimens of a venomous snake (Bothrops atrox), from the Brazilian Amazon rainforest, revealed that transcriptional and translational mechanisms contribute to venom phenotypic variation. Our finding of evidence for high expression of toxin proteins with conserved function supports the hypothesis that the venom phenotype consists of two kinds of proteins: conserved “core function” proteins that provide essential functional activities with broader relevance and less conserved “adaptive” proteins that vary in expression and may permit customization of protein function. These observations allowed us to suggest that genetic mechanisms controlling venom variability are not restricted to selection of gene copies or mutations in structural genes but also to selection of the mechanisms controlling gene expression, contributing to the plasticity of this important phenotype for venomous snakes.

Project description:Animal movement comes in a variety of 'types' including small foraging movements, larger one-way dispersive movements, seasonally-predictable round-trip migratory movements, and erratic nomadic movements. Although most individuals move at some point throughout their lives, movement patterns can vary widely across individuals within the same species: differing within an individual over time (intra-individual), among individuals in the same population (inter-individual), or among populations (inter-population). Yet, studies of movement (theoretical and empirical alike) more often focus on understanding 'typical' movement patterns than understanding variation in movement. Here, I synthesize current knowledge of movement variation (drawing parallels across species and movement types), describing the causes (what factors contribute to individual variation), patterns (what movement variation looks like), consequences (why variation matters), maintenance (why variation persists), implications (for management and conservation), and finally gaps (what pieces we are currently missing). By synthesizing across scales of variation, I span across work on plasticity, personality, and geographic variation. Individual movement can be driven by factors that act at the individual, population, community and ecosystem level and have ramifications at each of these levels. Generally the consequences of movement are less well understood than the causes, in part because the effects of movement variation are often nested, with variation manifesting at the population level, which in turn affects communities and ecosystems. Understanding both cause and consequence is particularly important for predicting when variation begets variation in a positive feedback loop, versus when a negative feedback causes variation to be dampened successively. Finally, maintaining standing variation in movement may be important for facilitating species' ability to respond to future environmental change.
