Tumor suppressor genes in the <i>CDKN2A/B</i> locus (<i>p15INK4b</i>, <i>p16INK4a</i>, and <i>p14ARF</i>) function as biological barriers to transformation and are the most frequently silenced or deleted genes in human cancers.
