Project description:In type 1 diabetes, the replacement of the destroyed beta cells could restore physiological glucose regulation and eliminate the need for exogenous insulin. Immunoisolation of these foreign cellular transplants via biomaterial encapsulation is widely used to prevent graft rejection. While highly effective in blocking direct cell-to-cell contact, nonspecific inflammatory reactions to the implant lead to the overproduction of reactive oxygen species, which contribute to foreign body reaction and encapsulated cell loss. For antioxidant protection, cerium oxide nanoparticles (CONPs) are a self-renewable, ubiquitous, free radical scavenger currently explored in several biomedical applications. Herein, 2-12 alternating layers of CONP/alginate are assembled onto alginate microbeads containing beta cells using a layer-by-layer (LbL) technique. The resulting nanocomposite coatings demonstrate robust antioxidant activity. The degree of cytoprotection correlates with layer number, indicating tunable antioxidant protection. Coating of alginate beads with 12 layers of CONP/alginate provides complete protection to the entrapped beta cells from exposure to 100 × 10-6 m H2 O2 , with no significant changes in cellular metabolic activity, oxidant capacity, or insulin secretion dynamics, when compared to untreated controls. The flexibility of this LbL method, as well as its nanoscale profile, provides a versatile approach for imparting antioxidant protection to numerous biomedical implants, including beta cell transplantation.

Project description:The rational approach motivated the design of novel antimicrobial silver and silver–copper bimetallic nanoparticles contained within zeolitic imidazolate framework-8 supported on graphene oxide (GO), Ag@ZIF-8@GO, and AgCu@ZIF8@GO. In the resultant composites, ZIF-8 was able to prevent the stacking of GO sheets and also acted as a carrier for the nanoparticles within its cavities. GO, on the other hand, acted as an anchoring support enabling uniform dispersion of the nanocomposites, thus eliminating their aggregation. The morphological and physicochemical properties of the composites were determined through a variety of characterization techniques, for example, transmission electron microscopy, scanning electron microscopy, Fourier-transform infrared spectroscopy, p-X-ray diffraction (XRD), nitrogen sorption, and X-ray photoelectron spectroscopy (XPS). The energy-dispersive system and XPS supplied evidence of the presence of all expected components in the composites. The XRD provided proof of a crystalline, distorted ZIF-8 structure. Ag@ZIF8@GO and Ag–Cu@ZIF-8@GO composites were effective against both Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria as determined by the disc diffusion method. The role of silver nanoparticles (AgNPs) in the antibacterial activity of both Ag@ZIF8@GO and AgCu@ZIF8@GO was highlighted as crucial in the probable pathway in the antimicrobial activity of the composites.

Project description:In this work, we designed and fabricated a multifunctional nanocomposite system that consists of chitosan, raspberry-like silver nanoparticles, and graphene oxide. The room temperature atmospheric pressure microplasma (RT-APM) process provides a rapid, facile, and environmentally-friendly method for introducing silver nanoparticles into the composite system. Our composite can achieve a pH controlled single and/or dual drug release. Under pH 7.4 for methyl blue loaded on chitosan, the drug release profile features a burst release during the first 10 h, followed by a more stabilized release of 70-80% after 40-50 h. For fluorescein sodium loaded on graphene oxide, the drug release only reached 45% towards the end of 240 h. When the composite acted as a dual drug release system, the interaction of fluorescein sodium and methyl blue slowed down the methyl blue release rate. Under pH 4, both single and dual drug systems showed a much higher release rate. In addition, our composite system demonstrated strong antibacterial abilities against E. coli and S. aureus, as well as an excellent photothermal conversion effect under irradiation of near infrared lasers. The photothermal conversion efficiency can be controlled by the laser power. These unique functionalities of our nanocomposite point to its potential application in multiple areas, such as multimodal therapeutics in healthcare, water treatment, and anti-microbials, among others.

Project description:Despite a decade of engineering and process improvements, bacterial infection remains the primary threat to implanted medical devices. Zinc oxide nanoparticles (ZnO-NPs) have demonstrated antimicrobial properties. Their microbial selectivity, stability, ease of production, and low cost make them attractive alternatives to silver NPs or antimicrobial peptides. Here we sought to (1) determine the relative efficacy of ZnO-NPs on planktonic growth of medically relevant pathogens; (2) establish the role of bacterial surface chemistry on ZnO-NP effectiveness; (3) evaluate NP shape as a factor in the dose-response; and (4) evaluate layer-by-layer (LBL) ZnO-NP surface coatings on biofilm growth. ZnO-NPs inhibited bacterial growth in a shape-dependent manner not previously seen or predicted. Pyramid shaped particles were the most effective and contrary to previous work, larger particles were more effective than smaller particles. Differential susceptibility of pathogens may be related to their surface hydrophobicity. LBL ZnO-NO coatings reduced staphylococcal biofilm burden by >95%. From the Clinical Editor: The use of medical implants is widespread. However, bacterial colonization remains a major concern. In this article, the authors investigated the use of zinc oxide nanoparticles (ZnO-NPs) to prevent bacterial infection. They showed in their experiments that ZnO-NPs significantly inhibited bacterial growth. This work may present a new alternative in using ZnO-NPs in medical devices.

Project description:The combination of complex perception, defense, and camouflage mechanisms is a pivotal instinctive ability that equips organisms with survival advantages. The simulations of such fascinating multi-stimuli responsiveness, including thigmotropism, bioluminescence, color-changing ability, and so on, are of great significance for scientists to develop novel biomimetic smart materials. However, most biomimetic color-changing or luminescence materials can only realize a single stimulus-response, hence the design and fabrication of multi-stimuli responsive materials with synergistic color-changing are still on the way. Here, a bioinspired multi-stimuli responsive actuator with color- and morphing-change abilities is developed by taking advantage of the assembled cellulose nanocrystals-based cholesteric liquid crystal structure and its water/temperature response behaviors. The actuator exhibits superfast, reversible bi-directional humidity and near-infrared (NIR) light actuating ability (humidity: 9 s; NIR light: 16 s), accompanying with synergistic iridescent appearance which provides a visual cue for the movement of actuators. This work paves the way for biomimetic multi-stimuli responsive materials and will have a wide range of applications such as optical anti-counterfeiting devices, information storage materials, and smart soft robots.

Project description:Biomineralization of natural polymers in simulated body fluid (SBF) can significantly improve its biocompatibility, osteoconductivity, and osteoinductivity because of the hydroxyapatite (HAp) deposition. Nevertheless, the superficial HAp crystal deposition hamper the deep inorganic ions exchange in porous microgels, thus gradually leading to a nonuniform regeneration effect. Inspired by the pearl forming process, this article uses the microarray chips to fabricate the multi-layer mineralized graphene oxide (GO)-collagen (Col)-hydroxyapatite (HAp) microgel, denoted as MMGCH. These fabricated MMGCH microgels exhibit porous structure and uniform HAp distribution. Furthermore, the suitable microenvironment offered by microgel promotes the time-dependent proliferation and osteogenic differentiation of stem cells, which resulted in upregulated osteogenesis-related genes and proteins, such as alkaline phosphatase, osteocalcin, and collagen-1. Finally, the MMGCH microgels possess favorable bone regeneration capacities both in cranial bone defects and mandibular bone defects via providing a suitable microenvironment for host-derived cells to form new bone tissues. This work presents a biomimetic means aiming to achieve full-thickness and uniform HAp deposition in hydrogel for bone defect repair.

Project description:Superparamagnetic Fe3O4 nanoparticles (NPs) based nanomaterials have been exploited in various biotechnology fields including biomolecule separation. However, slow accumulation of Fe3O4 NPs by magnets may limit broad applications of Fe3O4 NP-based nanomaterials. In this study, we report fabrication of Fe3O4 NPs double-layered silica nanoparticles (DL MNPs) with a silica core and highly packed Fe3O4 NPs layers. The DL MNPs had a superparamagnetic property and efficient accumulation kinetics under an external magnetic field. Moreover, the magnetic field-exposed DL MNPs show quantitative accumulation, whereas Fe3O4 NPs single-layered silica nanoparticles (SL MNPs) and silica-coated Fe3O4 NPs produced a saturated plateau under full recovery of the NPs. DL MNPs are promising nanomaterials with great potential to separate and analyze biomolecules.

Project description:Multi-color luminescence materials are important in the illumination, solid-state three-dimensional display, information storage, biological labelling and anticounterfeiting fields. Herein, we designed a novel core-shell structure upconversion nanoparticle (UCNP) material (NaYF4) with lanthanide ion doping to achieve multi-color luminescence under a single NIR excitation laser. Different from the typical single-sensitizer materials, the core-shell structure utilizes Nd3+, Yb3+, Tm3+ and Er3+ ions to obtain tuning of the color and brightness. The doping of Nd3+ ions enhances the weak color (red) light source to maintain the light color balance. Benefiting from the color adjustment of the sensitizers and the change of the core-shell coating, bright-white emission and flexible color emission from red to green, cyan and blue can be achieved via the diverse doped rare earth ions in a single UCNP under continuous-wave laser excitation (980 nm). Simultaneously, the emission color of the UCNPs can change with the intensity of the excitation light source and the wavelength. The bright-white emission can be used for lighting displays, and the flexible full-color emission can be applied in the anticounterfeiting and information storage fields.

Project description:A facile strategy to prepare GO-based nanocomposites with both gold nanoparticles (AuNPs) and ferrocene (Fc) moieties was developed. The surface of GO was modified with PFcMAss homopolymer by surface-initiated atom transfer radical polymerization of a new methacrylate monomer of 2-((2-(methacryloyloxy)ethyl)disulfanyl)ethyl ferrocene-carboxylate (FcMAss), consisting of disulfide as an anchoring group for stabilizing AuNPs and Fc group as an additional functionality. AuNPs with an average diameter of about 4.1 nm were formed in situ on the surface of PFcMAss-decorated GO (GO-PFcMAss) via Brust-Schiffrin method to give GO-PFcMAss-AuNPs multifunctional nanocomposites bearing GO, AuNPs and Fc groups. The obtained nanocomposites were characterized by X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), transmission electron microscopy (TEM) and atomic force microscopy (AFM). Since disulfide-containing polymers, rather than the commonly used thiol-containing compounds, were employed as ligands to stabilize AuNPs, much more stabilizing groups were attached onto the surface of GO, and thus more AuNPs were able to be introduced onto the surface of GO. Besides, polymeric chains on the surface of GO endowed GO-PFcMAss-AuNPs nanocomposites with excellent colloidal stability, and the usage of a disulfide group provides possibility to efficiently incorporate additional functionalities by easily modifying structure of disulfide-based monomer.

Project description:Multiple drug-resistance bacteria (MDRB) infection is one of the top three threats to human health according to the World Health Organization (WHO). Due to the large penetration depth and reduced photodamage, two-photon imaging is an highly promising technique for clinical MDRB diagnostics. Since most commercially available water-soluble organic dyes have low two-photon absorption cross-section and rapid photobleaching tendency, their applications in two-photon imaging is highly limited. Driven by the need, in this article we report extremely high two-photon absorption from aptamer conjugated graphene oxide (σ₂PA = 50800 GM) which can be used for highly efficient two-photon fluorescent probe for MDRB imaging. Reported experimental data show that two-photon photoluminescence imaging color, as well as luminescence peak position can be tuned from deep blue to red, just by varying the excitation wavelength without changing its chemical composition and size. We have demonstrated that graphene oxide (GO) based two-photon fluorescence probe is capable of imaging of multiple antibiotics resistance MRSA in the first and second biological transparency windows using 760-1120 nm wavelength range.
