ABSTRACT: Based on examination of herbarium specimens (including types) and living plants, as well as analysis of protologues and distributions, Parnassia tibetana, P. nubicola subsp. occidentalis, and P. nubicola var. nana are reduced to synonyms of P. nubicola.

Project description:P.brevistyla, P.delavayi and P.leptophylla. belong to Celastraceae, ParnassiaL.,Sect.NectarotrilobosDrude,Subsect.Xiphosandra (Franch.) Ku. Due to lack of material, the reliability of their taxonomic characteristics remained unknown and all three species have been retained. Following extensive field investigations, population sampling and examination of specimens, we determined that the previously used characteristics to distinguish these three species, i.e. the shape of basal leaves and the depth of staminode lobes, vary continuously within populations and should not be used to distinguish separate species. Therefore, P.brevistyla and P.leptophylla are hereby designated as synonyms of P.delavayi.

The complete chloroplast genome of Parnassia wightiana (Celastraceae).

Project description:The infrageneric relationships of the genus Parnassia were not clearly identified due to limited available genetic data. In this study, we presented the complete chloroplast genome of Parnassia wightiana for future phylogenetic study. The plastome of P. wightiana was 152,043 bp in length, with a large single-copy region (LSC) of 82,737 bp and a small single-copy region (SSC) of 19,030 bp, separated by a pair of inverted repeat regions (IRs) of 25,138 bp. It contained 134 genes, including 87 protein-coding genes, 37 tRNA genes, 8 rRNA genes, and 2 pseudogenes. The overall GC content was 37.1%, while the corresponding values in the LSC, SSC, and IR region were 35.0%, 30.8%, and 42.9%, respectively. The phylogenetic analysis supported the monophyly of Parnassia.

Four new synonyms and a new combination in Parnassia (Celastraceae).

Project description:Parnassia yunnanensis had been previously described based on mixed specimens containing materials partially belonging to Parnassia cacuminum, which makes the application of Parnassia yunnanensis ambiguous. Therefore, we lectotypified Parnassia yunnanensis and meanwhile synonymized Parnassia lanceolata var. oblongipetala under it. Parnassia yunnanensis var. longistipitata was found more similar to Parnassia cacuminum rather than Parnassia yunnanensis, thus a new combination, Parnassia cacuminum var. longistipitata comb. nov. was proposed. Furthermore, other three names (Parnassia vevusta, Parnassia degeensis and Parnassia kangdingensis) were reduced to synonyms of Parnassia cacuminum too.

Complete chloroplast genome sequence of Parnassia brevistyla (Celastraceae) and phylogenetic analysis with related species.

Project description:The taxonomic status of Parnassia has been widely discussed, which has been placed in more than five families, Parnassiaceae, Droseraceae, Saxifragaceae, and Celastraceae. Due to the lack of reliable genetic data, we sequenced and analyzed P. brevistyla chloroplast genome for future genetic study. The complete chloroplast genomes of Parnassia brevistyla was sequenced with NovaSeq 6000. The full length of P. brevistyla chloroplast genomes is 151,728 bp. A total of 114 unique genes, including 30 tRNA genes, four rRNA genes, and 80 protein-coding genes were found in the chloroplast genome. Using the whole chloroplast genome sequences alignment of 10 species from Celastraceae and Saxifragaceae, the phylogenetic relationship was built. The phylogenetic position of P. brevistyla was closely clustered with Celastraceae. The complete chloroplast genome of P. brevistyla provides utility information for further research of phylogenetic relationship and taxonomic status of Parnassia.

Taxonomic Identity of Carpinus dayongina Franchet (Betulaceae).

Project description:Carpinus polyneura and C. dayongina are recognised as separate species in Flora of China. In this study, the results of an examination of literature, morphological comparison and phenetic clustering of nuclear ITS sequences suggest that C. dayongina is conspecific with C. polyneura. Thus, we propose reducing C. dayongina to a synonym of C. polyneura.

Taxonomic note and description of new species of Fissocantharis Pic from China (Coleoptera, Cantharidae).

Project description:Two new species of Fissocantharis Pic are described, Fissocantharisbifoveatus sp. n. (CHINA: Yunnan) and Fissocantharisacuticollis sp. n. (CHINA: Zhejiang, Fujian, Guangdong, Hunan). Fissocantharispieli (Pic, 1937) is redescribed and Fissocanthariskontumensis Wittmer, 1989 is provided with a supplementary description. Fissocantharisshanensis (Wittmer, 1997) is synonymized with Fissocanthariskontumensis. For the above four species, illustrations of male genitalia are provided, for the latter three also photos of female genitalia and abdominal sternites VIII, and for the new species photos of male habitus and antennae are presented. Additionally, the specific name of Fissocanthariswittmeri (Y. Yang et X. Yang, 2009), preoccupied by Fissocanthariswittmeri (Kazantsev, 2007), is replaced by Fissocanthariswalteri Y. Yang et X. Yang, nom. n. And Fissocanthariswittmeri (Kazantsev, 2007) is found to be a junior objective synonym of Fissocantharisdenominata (Wittmer, 1997).

A taxonomic note on the helicoid land snail genus Traumatophora (Eupulmonata, Camaenidae).

Project description:Traumatophoratriscalpta (Martens, 1875) is reported for the first time from the Tianmushan Mountains, Zhejiang Province, and its morpho-anatomy is described based on this new material. The genus Traumatophora is redefined on the basis of both shell and genital anatomy of its type species. The presence of the dart apparatus suggests this genus belongs to the subfamily Bradybaeninae rather than to the Camaeninae. This genus is distinguished from all other Chinese bradybaenine genera by the combination of the following key morphological characteristics: embryonic shell smooth, palatal teeth present, dart sac tiny with rounded proximal accessory sac that opens into a dart sac chamber, mucous glands well developed, entering an accessory sac through a papilla, epiphallic papilla absent, flagellum present. A comparison is also presented of Chinese bradybaenine genera with known terminal genitalia.

Research Note: Development and application of specific molecular identity cards for "Yufen 1" H line chickens.

Project description:The "Yufen 1" H line chicken (YF) has excellent characteristics including early sexual maturity and high egg production, and the conservation of its genetic diversity is the core of the breeding activity. To overcome misrepresented breeds and protect the integrity of the germplasm genetic resources, it is important to develop accurate and convenient methods to identify YF. In this study, whole genome resequencing was performed on the YF population, and bioinformatics analysis was conducted by combining the data from different breeds. Linkage disequilibrium (LD) analysis revealed that YF had the slowest LD-decay rate, suggesting strong natural and artificial selection in its history. Through selective sweep analysis, 1,126 selected regions in YF were identified, which contained 163,661 single nucleotide polymorphisms (SNPs). In particular, 5 specific SNPs (SNP1: Chr2:45509616, SNP2: Chr2:45510792, SNP3: Chr9:13788193, SNP4: Chr9:13795646, SNP5: Chr9:13798154) were found exclusively in the YF population. Subsequently, PCR amplification and Sanger sequencing confirmed the presence of these 5 SNPs in YF. Finally, 4 SNPs (SNP1, SNP2, SNP4, SNP5) were screened and verified using the Kompetitive Allele Specific PCR (KASP) typing technique. These SNPs can be used as specific molecular identity cards (IDs) for YF authentication. The present study is of great significance to ensure sustainable conservation and promotion of YF germplasm resources.

Linking metabolite production to taxonomic identity in environmental samples by (MA)LDI-FISH.

Project description:One of the greatest challenges in microbial ecology remains to link the metabolic activity of individual cells to their taxonomic identity and localization within environmental samples. Here we combined mass-spectrometric imaging (MSI) through (matrix-assisted) laser desorption ionization time-of-flight MSI ([MA]LDI-TOF/MSI) with fluorescence in situ hybridization (FISH) to monitor antibiotic production in the defensive symbiosis between beewolf wasps and 'Streptomyces philanthi' bacteria. Our results reveal similar distributions of the different symbiont-produced antibiotics across the surface of beewolf cocoons, which colocalize with the producing cell populations. Whereas FISH achieves single-cell resolution, MSI is currently limited to a step size of 20-50 μm in the combined approach because of the destructive effects of high laser intensities that are associated with tighter laser beam focus at higher lateral resolution. However, on the basis of the applicability of (MA)LDI-MSI to a broad range of small molecules, its combination with FISH provides a powerful tool for studying microbial interactions in situ, and further modifications of this technique could allow for linking metabolic profiling to gene expression.
