Project description:The aim of the experiment was a green synthesis of cobalt nanoparticles from the aqueous extract of Ziziphora clinopodioides Lam (CoNPs) and assessment of their cytotoxicity, antioxidant, antifungal, antibacterial, and cutaneous wound healing properties. The synthesized CoNPs were characterized using different techniques including UV-Vis., FT-IR spectroscopy, X-ray diffraction (XRD), energy dispersive X-ray spectrometry (EDS), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). According to the XRD analysis, 28.19 nm was measured for the crystal size of NPs. TEM and SEM images exhibited a uniform spherical morphology and average diameters of 29.08 nm for the biosynthesized nanoparticles. Agar diffusion tests were done to determine the antibacterial and antifungal characteristics. Minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and minimum fungicidal concentration (MFC) were specified by macro-broth dilution assay. CoNPs indicated higher antibacterial and antifungal effects than many standard antibiotics (p???0.01). Also, CoNPs prevented the growth of all bacteria at 2-4 mg/mL concentrations and removed them at 2-8 mg/mL concentrations (p???0.01). In the case of antifungal effects of CoNPs, they inhibited the growth of all fungi at 1-4 mg/mL concentrations and destroyed them at 2-16 mg/mL concentrations (p???0.01). The synthesized CoNPs had great cell viability dose-dependently and indicated this method was nontoxic. DPPH free radical scavenging test was done to assess the antioxidant potentials, which revealed similar antioxidant potentials for CoNPs and butylated hydroxytoluene. In vivo experiment, after creating the cutaneous wound, the rats were randomly divided into six groups: untreated control, treatment with Eucerin basal ointment, treatment with 3% tetracycline ointment, treatment with 0.2% Co(NO3)2 ointment, treatment with 0.2% Z. clinopodioides ointment, and treatment with 0.2% CoNPs ointment. These groups were treated for 10 days. For histopathological and biochemical analysis of the healing trend, a 3?×?3 cm section was prepared from all dermal thicknesses at day 10. Use of CoNPs ointment in the treatment groups substantially raised (p???0.01) the wound contracture, hydroxyl proline, hexosamine, hexuronic acid, fibrocyte, and fibrocytes/fibroblast rate and remarkably decreased (p???0.01) the wound area, total cells, neutrophil, and lymphocyte compared to other groups. In conclusion, CoNPs can be used as a medical supplement owing to their non-cytotoxic, antioxidant, antibacterial, antifungal, and cutaneous wound healing effects. Additionally, the novel nanoparticles (Co(NO3)2 and CoNPs) were good inhibitors of the ?-glycosidase, and cholinesterase enzymes.

Project description:Background:Becium grandiflorum has been used traditionally for treatment of different ailments including diabetes mellitus although it lacks scientific evidence. Thus, the present study was aimed at evaluating the antidiabetic effect of Becium grandiflorum in streptozotocin (STZ)?-induced diabetic mice. Methods:The antidiabetic activity of hydro-ethanolic (30:70) leaf extract of Becium grandiflorum was evaluated in ?STZ (45 mg/kg)?-induced diabetic and normal mice. Antihyperglycemic, hypoglycemic, oral glucose tolerance and body weight change effects of the extract were assessed after administering three doses of the extract (200, 400 and 600 mg/kg), glibenclamide 5 mg/kg (reference drug) and 2% ?Tween 80 (vehicle). One-way analysis of variance and Tukey's post hoc test were used for data analysis. Results:All doses of the extract (200 mg/kg (p<0.05), 400 mg/kg (p<0.05) and 600 mg/kg (p<0.01)) and glibenclamide 5 mg/kg (p<0.001) showed statistically significant blood glucose level reduction in normal mice as compared to Tween 80. The hydroalcoholic extract at a dose of 200 mg/kg (p<0.05), 400 mg/kg (p<0.01) and 600 mg/kg (p<0.001) showed better blood glucose tolerance after 60, 120 and 180?-minute treatment duration in normal mice as compared to negative control. In diabetic mice, Becium grandiflorum doses and the reference drug caused maximum reduction in blood glucose level at the end of the 15th day of treatment by 17.61%, 22.52%, 24.62% and 34.12%, respectively. The extract's doses and the standard drug showed significant (p<0.05) improvement in body weight while the diabetic control continued to lose their body weight. Conclusion:Thus, Becium grandiflorum exhibits antihyperglycemic activity in ?STZ?-induced diabetic mice, and shows improvement in oral glucose tolerance and body weight, which justifies the claimed use of the plant in ameliorating diabetes mellitus in Ethiopian folk medicine.

Project description:Sweet potato (Ipomoea batatas, Lam.) is an important root vegetable in developing countries. After its domestication in Neotropical America, human migration led to the distribution of the sweet potato plant throughout the world. Both leaf and storage root are high in compounds of nutritional value. Yet, the storage roots are of particular value due to their significant content of provitamin A (?-carotene). The breeding effort for elite sweet potato lines led to the reduction of genetic diversity and the potential to improve other traits. The focus of the present study was to assess the metabolic diversity of 27 sweet potato cultivars including landraces and improved varieties. A metabolite profiling approach was optimised for sweet potato leaf and storage root tissue and 130 metabolites identified with three different analysis platforms. The data highlighted a lack of correlation between storage root phenotype and leaf metabolism. Furthermore, the metabolic diversity of storage roots was based on the secondary metabolism, including phenylpropanoids and carotenoids. Three cultivars of three different flesh colouration (yellow, orange and purple) showed a significant difference of the primary metabolism. This data demonstrates the value of metabolite profiling to breeding programs as a means of identifying differences in phenotypes/chemotypes and characterising parental material for future pre-breeding resources.

Project description:Oligogalacturonide fragments that activate defensive genes in plant leaves heretofore have been thought to be generated only by pathogen-derived pectin-degrading enzymes, because polygalacturonase (PG) activity has not been reported in leaves. Here, we report that mRNAs encoding a PG catalytic subunit protein and its regulatory (beta-subunit) protein are expressed in tomato leaves in response to wounding, systemin, and oligosaccharide elicitors. Synthesis of the two subunits in response to wounding is systemic and is accompanied by an increase in PG activity in extracts from both wounded and unwounded leaves. The finding that PG subunit mRNAs and PG enzyme activity are induced by wounding indicates that herbivore attacks can produce endogenous oligogalacturonide elicitors that may be involved in the local and systemic activation of defense responses against both herbivores and pathogens.

Project description:For many years, Aegle marmelos (A. marmelos) has been used medicinally and as a dietary supplement. Despite this, there are minimal research data on A. marmelos phytochemical properties and pharmacological effects. This study aimed to explore the phytoconstituents, cytotoxicity, glucose uptake, and antioxidant and antidiabetic potential of an alcoholic extract of A. marmelos leaf. The cytotoxicity of A. marmelos in HepG2 cells was tested in vitro, and the results revealed that it has strong cytocompatibility and cytoprotective properties. The extract's antioxidant activities were investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) methods. Antioxidant potential was shown to be quite impressive. The enzymes α-amylase and α-glycosidase were found to be substantially inhibited by A. marmelos, with IC50 values of 46.21 and 42.07 mg/mL, respectively. In HepG2 cells, A. marmelos significantly reduced ROS levels that were elevated due to high glucose and enhanced glucose consumption (p < 0.05). These activities might be due to the enrichment of bioactive phytoconstituents analyzed chromatographically using GC/MS and HPLC. The findings of this study show that A. marmelos could be an effective restorative therapy for diabetes and related diseases.

Project description:LapA encodes an acidic leucine aminopeptidase that is wound-inducible and found only in tomato (Solanum lycopersicum) and a small subset of the Solanaceae. LapA is expressed in developing flowers, fruit and in foliage after wounding, insect feeding (Manduca sexta), Pseudomonas syringae pv. tomato infection, salinity, and water-deficit stress. Using Agrobacterium tumefaciens, UC82b plants were transformed with a chimeric gene containing the CaMV 35S promoter and LapA cDNA in the sense orientation. Kanamycin-resistant plants were screened by RNA and protein blots to identify plants that did not accumulate and LAP-A protein upon wounding; these plants were designated LapA-silenced (LapA-SI) lines. The LapA-SI plants have an impaired wound response and do not accumulate wound response transcripts (proteinase inhibitors and polyphenol oxidase) to wild-type levels. LAP-A is the first aminopeptidase to be shown to have a regulatory role in plants. LapA-SI and UC82b plants were germinated on filter paper for 7 days before being planted in soil. Plants were grown in a growth chamber, (16 hour days at 28°C, 8 hour nights at 24°C, 50% humidity) for three weeks. Plants were wounded by crushing each leaflet on each of two lower leaves with pliers. Wounded leaves were collected at 0, 1 and 8 hr (local wounded tissue). Two upper, unwounded leaves were collected from the same plants at the same times (systemically wounded tissue). Five to seven plants were pooled for each time point. The experiment was repeated three times. RNA was extracted using a hot phenol/LiCl protocol and purified and DNase treated using the SV Total RNA Isolation Kit from Promega.
