Complete mitochondrial genome of Sichuan's population of Aphis aurantii (Hemiptera: Aphididae).

ABSTRACT: The complete mitochondrial genome of the black citrus aphid from Sichuan Province of China, Aphis aurantii, was sequenced and analyzed. The mitochondrial genome was a double strand, circular molecule with 15,296 bp and an A + T content of 83.5%, comprising 13 PCGs, 22 tRNA genes, and two rRNA genes. Gene arrangement was conserved in the mitogenome of A. aurantii. A 631-bp long control region was found, with a high A + T content of 82.6%. All PCGs used standard ATN start codons and most PCGs ended with complete TAA stop codons. The phylogenetic analysis supported that A. aurantii was closely related to other five congeners of the genus Aphis.

Phylogenetic relationship and characterization of the complete mitochondrial genome of the black citrus aphid, Aphis aurantii (Hemiptera: Aphididae).

Project description:The black citrus aphid Aphis aurantii is a major pest of citrus and tea plants. In this study, we determined the complete mitochondrial genome (mitogenome) of A. aurantii. The mitogenome was 15,469 bp in length, consisting of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and a non-coding control region. Gene order of A. aurantii was highly conserved and identical to most other previously sequenced Aphididae. Initiation codons ATT, ATA, and ATG were identified in eight, three, and two PCGs, respectively, while stop codon TAA was found in 11 genes except for 2 genes (cox1 and nad4) which use incomplete stop codon T-. Phylogenetic analysis showed the topology (Pemphigidae + (Hormaphididae + (Greenideidae + Aphididae))) within Aphidoidea, and A. aurantii was more closely related to Aphis craccivora than to Aphis gossypii.

Characterization and phylogenetic analysis of the complete mitochondrial genome of Aphis spiraecola (Hemiptera: Aphididae).

Project description:Aphis spiraecola is an important pest of citrus and transmits a number of plant viruses. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of A. spiraecola. This mitogenome was 15,465 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Gene order was conserved and identical to that of Drosophila yakuba and to most other previously sequenced Aphididae. The whole mitogenome exhibited heavy AT nucleotide bias (81.8%). All 13 PCGs were initiated by the ATN (ATG, ATT and ATA) codon. Except for cox1 and nad4 which end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that A. spiraecola got together with the same genus species Aphis gossypii and Aphis craccivora with high support value, and Aphis had a close relationship with Schizaphis and Rhopalosiphum.

Project description:Aphid gosypii Glover, 1877 is a widely recognized economically important aphid species in the world. We have determined mitochondrial genome of A. gossypii collected in Korean peninsula. The circular mitogenome of A. gossypii is 15,872 bp including 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNAs, and a single control region of 627 bp. The base composition was AT-biased (83.8%). In comparison of Chinese A. gossypii mitochondrial genomes with that of Korean sample, 61 single nucleotide polymorphisms and 3 insertions and deletions were identified, presenting lower level of those of Nilaparvata lugens, Laodelphax striatellus, and Chilo suppresallis.

Project description:The complete mitochondrial genome of Aphis gossypii Glover cucumber biotype was sequenced using traditional PCR amplification coupled with Sanger sequencing. The genome is 15,870 bp long, with 83.7% AT content (MW048625). The genome encodes 37 typical mitochondrial genes, including 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNAs, a repeat region of 784 bp, and a control region of 627 bp. The base composition of the genome is A (45.4%), T (38.3%), C (10.5%), and G (5.8%). An analysis of two biotypes A. gossypii mitogenomes identified 77 single nucleotide polymorphisms and 1 insertion and deletion.

Project description:We have determined mitochondrial genome of A. gossypii isolated from Plantago asiatica in Korea. The circular mitogenome of A. gossypii is 16,045 bp including 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single control region of 798 bp. Its AT ratio is 83.8%. In comparison this mitogenome to Chinese and Korean A. gossypii mitogenomes, 66 single nucleotide polymorphisms (SNPs) and 176 insertions and deletions (INDELs) and 11 SNPs and 173 INDELs are identified, respectively, presenting similar level to those of Nilaparvata lugens, Laodelphax striatellus, and Spodoptera frugiperda and lower than that of Chilo suppresallis.

Project description:We have determined the mitochondrial genome of A. gossypii isolated from Leonurus japonicus in Korea. The circular mitogenome of A. gossypii is 16,044 bp, including 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single control region of 797 bp. AT ratio is 83.8%. 88 SNPs and 4 INDELs (175 bp) were identified against mitogenome of A. gossypii isolated from cotton species. Especially, largest INDEL (170 bp) was in the control region. Phylogenetic trees show that four A. gossypii mitogenomes were clustered in one clade.

Population Genetic Structure of Aphis gossypii Glover (Hemiptera: Aphididae) in Korea.

Project description:Aphis gossypii Glover (Hemiptera: Aphididae) is a serious polyphagous agricultural pest worldwide. In the present study, we used eight microsatellite markers to investigate the genetic structure and diversity of A. gossypii populations in Korea. Samples were collected from 37 locations in Korea (18 populations in 2016, 14 populations in 2017, and five populations in 2018) from pepper plants. A. gossypii had low to moderate genetic diversity, and expected heterozygosity (HE) ranged from 0.354 to 0.719. A Mantel test of isolation by distance indicated no relationship between genetic structure and geographic distance among all populations (r2 = 0.0004, p = 0.370), suggesting high gene flow among populations in Korea. Populations of A. gossypii in Korea were divided into two distinct genetic clusters (ΔK = 2). In 2016 and 2017, the genetic clusters changed into opposite genetic structures within one year mostly in northwest and southeast parts of Korea. Possible relevance of study results was discussed. Chemical control, cyclical parthenogenesis, and immigrants from the exterior might have resulted in this low genetic diversity and opposite genetic clusters.

The complete mitochondrial genome of Brevicoryne brassicae (Hemiptera: Aphididae).

Project description:The mitochondrial genome of Brevicoryne brassicae (Hemiptera: Aphididae) was determined by Illumina paired-end sequencing. The genome size is 15,927 bp with 45.19% A, 39.03% T, 9.92% C and 5.86% G. It is encoded with 13 protein-coding genes, 22 transfer RNAs and two ribosome RNAs. The phylogenetic tree showed that B. brassicae clustered within clade of Aphididae, had a closer relationship with genus Diuraphis than the rest genera. This study provides important information for future study on the evolution, genetic and molecular biology of Brevicoryne.

The Complete Mitochondrial Genome of Mindarus keteleerifoliae (Insecta: Hemiptera: Aphididae) and Comparison with Other Aphididae Insects.

Project description:The mitogenome of Mindarus keteleerifoliae Zhang (Hemiptera: Aphididae) is a 15,199 bp circular molecule. The gene order and orientation of M. keteleerifoliae is similarly arranged to that of the ancestral insect of other aphid mitogenomes, and, a tRNA isomerism event maybe identified in the mitogenome of M. keteleerifoliae. The tRNA-Trp gene is coded in the J-strand and the same sequence in the N-strand codes for the tRNA-Ser gene. A similar phenomenon was also found in the mitogenome of Eriosoma lanigerum. However, whether tRNA isomers in aphids exist requires further study. Phylogenetic analyses, using all available protein-coding genes, support Mindarinae as the basal position of Aphididae. Two tribes of Aphidinae were recovered with high statistical significance. Characteristics of the M. keteleerifoliae mitogenome revealed distinct mitogenome structures and provided abundant phylogenetic signals, thus advancing our understanding of insect mitogenomic architecture and evolution. But, because only eight complete aphid mitogenomes, including M. keteleerifoliae, were published, future studies with larger taxon sampling sizes are necessary.

Complete mitochondrial genome of Mollitrichosiphum tenuicorpus (Okajima, 1908) (Hemiptera: Aphididae: Greenideinae).

Project description:Here we report the complete mitochondrial genome of the aphid species Mollitrichosiphum tenuicorpus. The M. tenuicorpus mitogenome is 15,727 bp in length and comprising 37 genes typically present in insect mitogenomes, a control region, and a unique repeat region. All protein-coding genes (PCGs) terminate with TAA or TAG except for cox1, which is terminated with T-. The secondary structure of trnS (AGN) loses the dihydrouridine (DHU) arm, but all the other transfer RNAs show the typical clover-leaf secondary structure. The repeat region between trnE and trnF is 458 bp, with a 217-bp repeat unit repeating 2.11 times. Phylogenetic analysis of the M. tenuicorpus mitogenome using the maximum-likelihood optimality criterion places it in a strongly supported sister position to Eutrichosiphum pasaniae. These data show that mitogenome sequences could be useful in resolving phylogenetic relationships of the Greenideinae.
