Prion diseases are characterized by the self-templated misfolding of the cellular prion protein (PrP<sup>C</sup>) into infectious aggregates (PrP<sup>Sc</sup>). The detailed molecular basis of the misfolding and aggregation of PrP<sup>C</sup> remains incompletely understood.
