Complete mitochondrial genome of the black-winged fly, Felderimyia fuscipennis (Diptera: Tephritidae) and its phylogenetic relationship within family Tephritidae.

ABSTRACT: The black-winged fly, Felderimyia fuscipennis (Diptera: Tephritidae), is an insect pest of bamboo shoot, mainly distributed in Thailand, Malaysia and Yunnan Province and Guangxi Autonomous Region, China. The complete sequence of the mitogenome of F. fuscipennis has been determined in this study. The whole mitogenome sequence is 16,536 bp in length, which totally contains 13 protein-coding genes (PCGs), 2 rRNA genes, 22 tRNA genes, and a non-coding region (putative control region, CR). The phylogeny indicates that F. fuscipennis of subfamily Trypetinae was monophyletic and clearly separated from both Dacinae and Tephritinae with high bootstrap value supported.

Project description:Acrotaeniostola dissimilis (Diptera: Tephritidae) is an insect pest of bamboo shoot and distributed in the Yunnan and Sichuan Provinces, Southwest China. Complete mitogenome sequence has been determined in this study. The circular genome is 15384 bp long and contains a standard gene complement, that is, the large and small ribosomal RNA subunits, 22 transfer RNA genes, 13 genes encoding mitochondrial proteins, and a non-coding A + T-rich control region. The phylogeny showed that A. dissimilis in tribe Gastrozonini was a monophyletic branch and clearly separated from both tribe Dacini and tribe Ceratitidini with high bootstrap value supported.

Complete sequence of the mitogenome of C. vesuviana has been determined in this study. The circular genome is 15,267?bp long and contains a standard gene complement, that is, the large and small ribosomal RNA subunits, 22 transfer RNA genes, 13 genes encoding mitochondrial proteins, and a non-coding A?+?T-rich control region. The phylogeny showed that C. vesuviana of subfamily Trypetinae was monophyletic and clearly separated from both Dacinae and Tephritinae with maximum support (p?=?1).

Complete mitochondrial genome of the pumpkin fruit fly, Bactrocera depressa (Diptera: Tephritidae).

Project description:In this study, we sequenced the complete mitochondrial genome (mitogenome) of the pumpkin fruit fly, Bactrocera depressa (Diptera: Tephritidae), which is an economically damaging pest of pumpkin and turban squash. The 15,832-bp-long complete mitogenome of the species consists of a typical set of genes, with an arrangement typical of insects. Of the 13 protein-coding genes (PCGs), 12 have a typical ATN start codon, whereas the COI gene begins with TCG, which has been identified as the start codon for all Bactrocera COI genes. The 1004-bp A?+?T-rich region of B. depressa is the third longest, after B. minax and B. scutellata, of the Bactrocera species for which the whole mitogenome has been sequenced. Phylogenetic analysis using the 13 PCGs of Bactrocera species indicated that B. depressa is a sister to the sister group containing B. tau and B. cucurbitae with the highest nodal support (Bayesian posterior probability =1.0).

Project description:The melon fly, Zeugodacus cucurbitae (Diptera:Tephritidae) is an important invasive pest and distributed throughout tropical, subtropical countries and areas. In this study, we report the complete mitochondrial genome of the fly with 15, 685 base pair in length, which includes 37 genes (the large and small ribosomal RNA subunits, 22 transfer RNA genes, 13 genes encoding mitochondrial proteins) and a non-coding A + T-rich control region. Molecular phylogeny indicated that there was a high bootstrap value supported among Z. cucurbitae and Z. tau belonging to the Z. tau complex.

Project description:The striped fruit fly, Bactrocera (Zeugodacus) scutellata occurred in East and Southeast Asia and is one of the most serious pest insects in China. Complete mitochondrial genome was sequenced and characterized for the fruit fly from Anshun, Guizhou Province in Southwest China. The pest fruit fly has a total length of 15,904?bp, consisting of 13 protein-coding genes (PCGs), 22 tRNA, 2 rRNA genes and a non-coding region (A?+?T-rich control region). The phylogenetic position of B. scutellata was closely clustered with B. cucurbitae, B. tau, B. caudata and B. diaphora. The all species of the subgenus Zeugodacus formed a sister group which was monophyletic. The complete mitochondrial genome of B. scutellata will provide helpful information for genetics, systematics and phylogeny of tephritid fruit flies, Particularly Bactrocera genus.

Τhe Complete Mitochondrial Genome of Bactrocera carambolae (Diptera: Tephritidae): Genome Description and Phylogenetic Implications.

Project description:Bactrocera carambolae is one of the approximately 100 sibling species of the Bactrocera dorsalis complex and considered to be very closely related to B. dorsalis. Due to their high morphological similarity and overlapping distribution, as well as to their economic impact and quarantine status, the development of reliable markers for species delimitation between the two taxa is of great importance. Here we present the complete mitochondrial genome of B. carambolae sourced from its native range in Malaysia and its invaded territory in Suriname. The mitogenome of B. carambolae presents the typical organization of an insect mitochondrion. Comparisons of the analyzed B. carambolae sequences to all available complete mitochondrial sequences of B. dorsalis revealed several species-specific polymorphic sites. Phylogenetic analysis based on Bactrocera mitogenomes supports that B. carambolae is a differentiated taxon though closely related to B. dorsalis. The present complete mitochondrial sequences of B. carambolae could be used, in the frame of Integrative Taxonomy, for species discrimination and resolution of the phylogenetic relationships within this taxonomically challenging complex, which would facilitate the application of species-specific population suppression strategies, such as the sterile insect technique.

Proteomics and Lipidomics of Black Soldier Fly (Diptera: Stratiomyidae) and Blow Fly (Diptera: Calliphoridae) Larvae.

Project description:Farming insects has recently emerged as a new source of protein and lipid production. To date, research has mostly focused on food applications of insects. Focusing on nonfood potential of oil and proteins of insects, high-throughput studies of insect lipids and proteins are needed. We performed proteomics and lipidomics investigation on black soldier fly (Hermitia illucens) and blow fly (Lucilia sericata) larvae to investigate new potential and applications. We used mass spectrometry for proteomics and lipidomics analysis of control and treated larvae. Treatment was performed by incubation with a biological decomposer. We provide the list of all fatty acids with their concentration in control and treated larvae. This result showed high levels of lauric acid in black soldier fly, which could even increase after biological decomposition. Proteomics analysis showed the presence of proteins like collagen of cosmetic interest, and proteins with antimicrobial properties such as phenoloxidases and enzymatic activities, such as amylase and trypsin. Insects harbor high potential for nonfood usage as additives, antimicrobial effects, and even pharmaceuticals and cosmetics. These data open avenues for future research in pharmacological and cosmetic approaches to find new molecules of interests.

Complete Mitochondrial Genome of Three Bactrocera Fruit Flies of Subgenus Bactrocera (Diptera: Tephritidae) and Their Phylogenetic Implications.

Project description:Bactrocera latifrons is a serious pest of solanaceous fruits and Bactrocera umbrosa is a pest of Artocarpus fruits, while Bactrocera melastomatos infests the fruit of Melastomataceae. They are members of the subgenus Bactrocera. We report here the complete mitochondrial genome of these fruit flies determined by next-generation sequencing and their phylogeny with other taxa of the subgenus Bactrocera. The whole mitogenomes of these three species possessed 37 genes namely, 13 protein-coding genes (PCGs), 2 rRNA and 22 tRNA genes. The mitogenome of B. latifrons (15,977 bp) was longer than those of B. melastomatos (15,954 bp) and B. umbrosa (15,898 bp). This difference can be attributed to the size of the intergenic spacers (283 bp in B. latifrons, 261 bp in B. melastomatos, and 211 bp in B. umbrosa). Most of the PCGs in the three species have an identical start codon, except for atp8 (adenosine triphosphate synthase protein 8), which had an ATG instead of GTG in B. umbrosa, whilst the nad3 (NADH dehydrogenase subunit 3) and nad6 (NADH dehydrogenase subunit 6) genes were characterized by an ATC instead of ATT in B. melastomatos. The three species had identical stop codon for the respective PCGs. In B. latifrons and B. melastomatos, the T?C (thymidine-pseudouridine-cytidine)-loop was absent in trnF (phenylalanine) and DHU (dihydrouracil)-loop was absent in trnS1 (serine S1). In B. umbrosa, trnN (asparagine), trnC (cysteine) and trnF lacked the T?C-loop, while trnS1 lacked the DHU-stem. Molecular phylogeny based on 13 PCGs was in general concordant with 15 mitochondrial genes (13 PCGs and 2 rRNA genes), with B. latifrons and B. umbrosa forming a sister group basal to the other species of the subgenus Bactrocera which was monophyletic. The whole mitogenomes will serve as a useful dataset for studying the genetics, systematics and phylogenetic relationships of the many species of Bactrocera genus in particular, and tephritid fruit flies in general.

Characterisation of the complete mitochondrial genome of Bactrocera ruiliensis (Diptera: Tephritidae), with its phylogenetic analysis.

Project description:The complete mitochondrial genome (mitogenome) of the Bactrocera ruiliensis (Diptera: Tephritidae: Dacinae) is sequenced and annotated. The mitochondrial genome is 15,870 bp (GenBank No. MN477221) and has an A + T content of 73.3% (A 39.2%; C 16.2%; G 10.3%, and T 34.4%), which is the classical structure for insect mitogenome. All PCGs started with ATN, except ATP8, which is started with TTG; 12 PCGs use TAR (TAA/TAG) as the stop codon, except COX1, which ends with single T--. The phylogenetic tree confirms that B. ruiliensis clustered with other Bactrocera species. This study enriches the mitogenomes of the fruit flies.
