The complete mitochondrial genome of Eutrichosiphum pasaniae (Okajima, 1908) (Hemiptera: Aphididae: Greenideinae).

ABSTRACT: In this study, we sequenced the complete mitochondrial genome of Eutrichosiphum pasaniae through Illumina platform. The circular mitogenome is 16,500 bp in length and composed of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), a large control region and a special repeat region. The nucleotide composition of whole mitogenome is strongly AT-biased (85.5%). All PCGs start with ATN and end with TAA except for cox1 which terminates with an incomplete stop codon T. All tRNAs have a typical clover-leaf secondary structure except for trnS (AGN). The lengths of rrnL, rrnS and control region are 1276, 774 and 996 bp, respectively. The repeat region with a length of 909 bp is located between trnE and trnF and consists of 4.1 repeat units. The phylogenetic tree supports the sister relationship of Eutrichosiphum pasaniae and Greenidea psidii.

Project description:Here we report the complete mitochondrial genome of the aphid species Mollitrichosiphum tenuicorpus. The M. tenuicorpus mitogenome is 15,727 bp in length and comprising 37 genes typically present in insect mitogenomes, a control region, and a unique repeat region. All protein-coding genes (PCGs) terminate with TAA or TAG except for cox1, which is terminated with T-. The secondary structure of trnS (AGN) loses the dihydrouridine (DHU) arm, but all the other transfer RNAs show the typical clover-leaf secondary structure. The repeat region between trnE and trnF is 458 bp, with a 217-bp repeat unit repeating 2.11 times. Phylogenetic analysis of the M. tenuicorpus mitogenome using the maximum-likelihood optimality criterion places it in a strongly supported sister position to Eutrichosiphum pasaniae. These data show that mitogenome sequences could be useful in resolving phylogenetic relationships of the Greenideinae.

Project description:We sequenced the complete mitochondrial genome of Schoutedenia ralumensis. The mitogenome is 16,051 bp long with an A + T content of 84.5%, including 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, a control region, and an aphid-specific repeat region located between trnE and trnF. All protein-coding genes are initiated by ATN and terminated with TAA or TAG except for cox1 and nad5. All transfer RNAs display the typical clover-leaf secondary structure except for trnS (AGN). The unique repeat region is 974 bp long, in which a 305-bp repeat unit repeats 3.19 times. The phylogenetic tree supports a sister relationship of S. ralumensis and Greenidea psidii.

Project description:In this study, the complete mitochondrial genome of the pest aphid Greenidea ficicola was determined. The mitogenome was 17,361 bp in length, containing 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, 1 long control region located between srRNA and tRNAIle , and a repeat region located between tRNAGlu and tRNAPhe . Thirteen protein-coding genes have typical ATN start codon and TAA termination codon. All tRNAs were predicted to contain typical clover-leaf secondary structures except tRNASer(gct). The length of lrRNA and srRNA are 1270 bp and 773 bp, respectively. Phylogenetic analysis shows that Greenideinae speices form a highly supported clade.

The complete mitochondrial genome of Brevicoryne brassicae (Hemiptera: Aphididae).

Project description:The mitochondrial genome of Brevicoryne brassicae (Hemiptera: Aphididae) was determined by Illumina paired-end sequencing. The genome size is 15,927 bp with 45.19% A, 39.03% T, 9.92% C and 5.86% G. It is encoded with 13 protein-coding genes, 22 transfer RNAs and two ribosome RNAs. The phylogenetic tree showed that B. brassicae clustered within clade of Aphididae, had a closer relationship with genus Diuraphis than the rest genera. This study provides important information for future study on the evolution, genetic and molecular biology of Brevicoryne.

The Complete Mitochondrial Genome of Mindarus keteleerifoliae (Insecta: Hemiptera: Aphididae) and Comparison with Other Aphididae Insects.

Project description:The mitogenome of Mindarus keteleerifoliae Zhang (Hemiptera: Aphididae) is a 15,199 bp circular molecule. The gene order and orientation of M. keteleerifoliae is similarly arranged to that of the ancestral insect of other aphid mitogenomes, and, a tRNA isomerism event maybe identified in the mitogenome of M. keteleerifoliae. The tRNA-Trp gene is coded in the J-strand and the same sequence in the N-strand codes for the tRNA-Ser gene. A similar phenomenon was also found in the mitogenome of Eriosoma lanigerum. However, whether tRNA isomers in aphids exist requires further study. Phylogenetic analyses, using all available protein-coding genes, support Mindarinae as the basal position of Aphididae. Two tribes of Aphidinae were recovered with high statistical significance. Characteristics of the M. keteleerifoliae mitogenome revealed distinct mitogenome structures and provided abundant phylogenetic signals, thus advancing our understanding of insect mitogenomic architecture and evolution. But, because only eight complete aphid mitogenomes, including M. keteleerifoliae, were published, future studies with larger taxon sampling sizes are necessary.

Phylosymbiotic Structures of the Microbiota in Mollitrichosiphum tenuicorpus (Hemiptera: Aphididae: Greenideinae).

Project description:Aphids harbor an array of symbionts that provide hosts with ecological benefits. Microbial community assembly generally varies with respect to aphid species, geography, and host plants. However, the influence of host genetics and ecological factors on shaping intraspecific microbial community structures has not been fully understood. In the present study, using Illumina sequencing of the V3 - V4 hypervariable region of the 16S rRNA gene, we characterized the microbial compositions associated with Mollitrichosiphum tenuicorpus from different regions and plants in China. The primary symbiont Buchnera aphidicola and the secondary symbiont Arsenophonus dominated the microbial flora in M. tenuicorpus. Ordination analyses and statistical tests suggested that geography and aphid genetics primarily contributed to the variation in the microbiota of M. tenuicorpus. We further confirmed the combined effect of aphid genetics and geography on shaping the structures of symbiont and secondary symbiont communities. Moreover, the significant correlation between aphid genetic divergence and symbiont community dissimilarity provides evidence for intraspecific phylosymbiosis in natural systems. Our study helped to elucidate the eco-evolutionary relationship between symbiont communities and aphids within one given species.

Project description:We have determined the mitochondrial genome of Paracolopha morrisoni (Baker, 1919), a gall-forming aphid collected from Korea. The circular mitogenome of Paracolopha morrisoni is 16,330 bp long including 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNAs, and a single large non-coding region of 932 bp. The base composition was AT-biased (84.9%). Gene order of P. morrisoni is identical to all other aphid mitochondrial genomes. Phylogenetic trees show that P. morrisoni is sister to Eriosoma lanigerum both belonging to tribe. The mitochondrial genome of P. morrisoni will be useful in understanding the genetic backgrounds of the species.

Project description:We have sequenced mitochondrial genome of Uroleucon erigeronense (Thomas, 1878) isolated from Erigeron canadensis in Korea. The circular mitogenome of U. erigeronense is 15,691 bp long including 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single control region of 968 bp. AT ratio is 84.2%. Additional phylogenetic studies of aphid mitogenomes are required due to the inconsistency found in the three phylogenetic trees.

Complete mitochondrial genome of Sichuan's population of Aphis aurantii (Hemiptera: Aphididae).

Project description:The complete mitochondrial genome of the black citrus aphid from Sichuan Province of China, Aphis aurantii, was sequenced and analyzed. The mitochondrial genome was a double strand, circular molecule with 15,296 bp and an A + T content of 83.5%, comprising 13 PCGs, 22 tRNA genes, and two rRNA genes. Gene arrangement was conserved in the mitogenome of A. aurantii. A 631-bp long control region was found, with a high A + T content of 82.6%. All PCGs used standard ATN start codons and most PCGs ended with complete TAA stop codons. The phylogenetic analysis supported that A. aurantii was closely related to other five congeners of the genus Aphis.

Complete mitochondrial genome of Rhopalosiphum maidis (Hemiptera: Aphididae) and its phylogenetic implications.

Project description:Rhopalosiphum maidis Fitch, 1856 is widespread in tropical and temperate regions. R. maidis can spread viral diseases in maize and harm various important crops. In the present study, we report the first complete mitochondrial genome of R. maidis. The circular genome is found to be 17,021 bp in length, includes a standard set of 22 transfer RNAs, two ribosomal RNAs, 13 protein-coding genes, and two non-coding control regions. The base composition is 84.32% AT and 15.79% GC. The phylogenetic tree of the 17 Aphidini families constructed based on the nucleotide sequences of complete mitochondrial genomes strongly supports the conclusion that R. maidis is closely related to R. rufiabdominalis.
