The complete mitogenome of the large toothed toad, Oreolalax major (Anura: Megophryidae) with phylogenetic analysis.

ABSTRACT: The complete mitochondrial genome of the Oreolalax major (17,786 bp long) was obtained in this study. It includes 13 protein-coding genes (PCG), two ribosomal RNA (rRNA) genes, and 23 transfer RNA (tRNA) genes (GenBank accession number MN803320). The phylogenetic tree indicates that the O. major is closely related to the O. xiangchengensis.

Project description:We sequenced and annotated the complete mitogenome sequence of Oreolalax omeimontis (17,675 bp long). The mitogenome encoded 13 protein-coding genes (PCG), 2 ribosomal RNA (rRNA) genes, 23 transfer RNA (tRNA) genes, and a control region (GenBank accession number MN803321). The phylogenetic tree conforms the close relationship of O. omeimontis and O. multipunctatus, and a monophyletic clade of genus Oreolalax.

Project description:In this study, the near complete mitogenome sequence (15,469?bp) of Oreolalax major was determined using polymerase chain reaction (PCR). It includes 13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes and 19 transfer RNA (tRNA) genes (GenBank accession number KU310894). The features of O. major have one more tRNA gene (tRNAMet ) behind the original one before ND2 which is similar to Leptobrachium boringii. Phylogenetic analyses were based on the concatenated sequences of the 13 protein-encoding genes of O. major and other related species.

A new species of the toothed toad Oreolalax (Anura, Megophryidae) from Sichuan Province, China.

Project description:The toad genus Oreolalax is widely distributed in southwest China and northern Vietnam. A new species of the genus is described from Sichuan Province, China. Phylogenetic analyses based on the mitochondrial 12S rRNA and 16S rRNA gene sequences supported the new species as an independent clade clustered into the clade also containing O. nanjiangensis and O. chuanbeiensis. The new species can be distinguished from its congeners by a combination of the following characters: body size moderate (SVL 51.2-64.2 mm in males); head broad; tympanum hidden; interorbital region with dark triangular pattern; belly with marbling; male lacking spines on lip margin; spiny patches on chest small with thick sparse spines in male; nuptial spines thick and sparse; tibio-tarsal articulation reaching beyond nostril when leg stretched forward; toe webbing at base.

Project description:Oreolalax xiangchengensis is an alpine toad species of Megophryidae. We determined its complete mitogenome. The mitogenome has a length of 17,110 bp, containing 13 protein-coding genes, 2 rRNA genes, 23 tRNA genes, and a control region. It has a tandem duplication of tRNA-Met gene and a translocation of tRNA-Trp gene compared with typical vertebrate mitogenomes. Phylogenetic analyses based on the mitogenome sequences of 18 Megophryidae species were conducted to access phylogenetic assignments of O. xiangchengensis.

Compared with most other vertebrates, this mitogenome appears a tandem duplication of tRNAMet gene. The tRNATrp gene of Oreolalax major translocates from the "WANCY" tRNA cluster to upstream of D-loop. As the first report of the mitogenome sequence from the genus Oreolalax, it will provide fundamental data for further research of phylogeny and biogeography with this genus.

The base composition on light strand was 28.0% A, 32.2% T, 24.9% C and 14.9% G. Compared with most other vertebrates, this mitogenome appear a tandem duplication of tRNA-Met gene. This study will facilitate the further research of the population genetics of this species and systematic study of the genus Oreolalax.

Complete mitochondrial genome of the Alpine Metacarpal-tubercled Toad Leptobrachella alpina (Amphibia, Anura, Megophryidae).

Project description:The complete mitochondrial genome of the Leptobrachella alpina Fei, Ye, and Li 1990, was assembled for the first time. The mitogenome of this species was 17,763 bp in length, containing 13 protein-coding genes, two ribosomal RNA genes (12S rRNA and 16S rRNA), 22 transfer RNA genes (tRNA), and a non-coding control region (D-loop). The base content of the mitogenome was that A, T, G, and C occupied 28.5%, 30.8%, 15.1%, and 25.6%, respectively. The phylogenetic analysis was conducted based on 17 complete mitogenome sequences of the family Megophryidae by the Bayesian inference approach. The phylogenetic tree suggested that Leptobrachium and Oreolalax clustered into a clade and formed a sister group with Leptobrachella. This work is critical for the further genetic research and conservation of this species.

Complete Mitogenome of Oreolalax Omeimontis Reveals Phylogenetic Status and Novel Gene Arrangement of Archaeobatrachia.

Project description:Species of the genus Oreolalax displayed crucial morphological characteristics of vertebrates transitioning from aquatic to terrestrial habitats; thus, they can be regarded as a representative vertebrate genus for this landing phenomenon. But the present phylogenetic status of Oreolalax omeimontis has been controversial with morphological and molecular approaches, and specific gene rearrangements were discovered in all six published Oreolalax mitogenomes, which are rarely observed in Archaeobatrachia. Therefore, this study determined the complete mitogenome of O. omeimontis with the aim of identifying its precise phylogenetic position and novel gene arrangement in Archaeobatrachia. Phylogenetic analysis with Bayesian inference and maximum likelihood indicates O. omeimontis is a sister group to O. lichuanensis, which is consistent with previous phylogenetic analysis based on morphological characteristics, but contrasts with other studies using multiple gene fragments. Moreover, although the duplication of trnM occurred in all seven Oreolalax species, the translocation of trnQ and trnM occurred differently in O. omeimontis to the other six, and this unique rearrangement would happen after the speciation of O. omeimontis. In general, this study sheds new light on the phylogenetic relationships and gene rearrangements of Archaeobatrachia.

Two complete mitochondrial genomes of Boulenophrys (Anura: Megophryidae: Megophryinae): characteristics and phylogenetic implications.

Project description:The Chinese horned toads, Boulenophrys boettgeri (Boulenger, 1899) and Boulenophrys kuatunensis (Pope, 1929), are two captivating species within the family Megophryidae, which inhabit the mountainous streams in the Eastern of China. In this study, two new complete mitochondrial genomes of B. boettgeri and B. kuatunensis were sequenced, assembled, and annotated using next-generation sequencing. The length of mitochondrial genomes of B. boettgeri and B. kuatunensis was 16,597 and 17,921 bp, respectively, with both containing 13 protein coding genes, 22 tRNA genes, two rRNA genes, and one putative control region. Phylogenetic relationships based on protein-coding mitochondrial genes showed that the two Boulenophrys species formed a cluster with other Boulenophrys species. The two new sequences provide valuable insights into the mitochondrial genomes of these two species, offering important data for understanding the phylogenetic relationships of the genus Boulenophrys.
