The complete mitogenome of the omei toothed toad, Oreolalax omeimontis (Anura: Megophryidae) with phylogenetic analysis.

ABSTRACT: We sequenced and annotated the complete mitogenome sequence of Oreolalax omeimontis (17,675 bp long). The mitogenome encoded 13 protein-coding genes (PCG), 2 ribosomal RNA (rRNA) genes, 23 transfer RNA (tRNA) genes, and a control region (GenBank accession number MN803321). The phylogenetic tree conforms the close relationship of O. omeimontis and O. multipunctatus, and a monophyletic clade of genus Oreolalax.

A new species of the toothed toad Oreolalax (Anura, Megophryidae) from Sichuan Province, China.

Project description:The toad genus Oreolalax is widely distributed in southwest China and northern Vietnam. A new species of the genus is described from Sichuan Province, China. Phylogenetic analyses based on the mitochondrial 12S rRNA and 16S rRNA gene sequences supported the new species as an independent clade clustered into the clade also containing O. nanjiangensis and O. chuanbeiensis. The new species can be distinguished from its congeners by a combination of the following characters: body size moderate (SVL 51.2-64.2 mm in males); head broad; tympanum hidden; interorbital region with dark triangular pattern; belly with marbling; male lacking spines on lip margin; spiny patches on chest small with thick sparse spines in male; nuptial spines thick and sparse; tibio-tarsal articulation reaching beyond nostril when leg stretched forward; toe webbing at base.

First near complete mitochondrial genome of large toothed toad, Oreolalax major (Anura: Megophryidae) from southwest China.

Project description:In this study, the near complete mitogenome sequence (15,469 bp) of Oreolalax major was determined using polymerase chain reaction (PCR). It includes 13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes and 19 transfer RNA (tRNA) genes (GenBank accession number KU310894). The features of O. major have one more tRNA gene (tRNAMet ) behind the original one before ND2 which is similar to Leptobrachium boringii. Phylogenetic analyses were based on the concatenated sequences of the 13 protein-encoding genes of O. major and other related species.

The mitogenome has a length of 17,110 bp, containing 13 protein-coding genes, 2 rRNA genes, 23 tRNA genes, and a control region. It has a tandem duplication of tRNA-Met gene and a translocation of tRNA-Trp gene compared with typical vertebrate mitogenomes. Phylogenetic analyses based on the mitogenome sequences of 18 Megophryidae species were conducted to access phylogenetic assignments of O. xiangchengensis.

Complete Mitogenome of Oreolalax Omeimontis Reveals Phylogenetic Status and Novel Gene Arrangement of Archaeobatrachia.

Project description:Species of the genus Oreolalax displayed crucial morphological characteristics of vertebrates transitioning from aquatic to terrestrial habitats; thus, they can be regarded as a representative vertebrate genus for this landing phenomenon. But the present phylogenetic status of Oreolalax omeimontis has been controversial with morphological and molecular approaches, and specific gene rearrangements were discovered in all six published Oreolalax mitogenomes, which are rarely observed in Archaeobatrachia. Therefore, this study determined the complete mitogenome of O. omeimontis with the aim of identifying its precise phylogenetic position and novel gene arrangement in Archaeobatrachia. Phylogenetic analysis with Bayesian inference and maximum likelihood indicates O. omeimontis is a sister group to O. lichuanensis, which is consistent with previous phylogenetic analysis based on morphological characteristics, but contrasts with other studies using multiple gene fragments. Moreover, although the duplication of trnM occurred in all seven Oreolalax species, the translocation of trnQ and trnM occurred differently in O. omeimontis to the other six, and this unique rearrangement would happen after the speciation of O. omeimontis. In general, this study sheds new light on the phylogenetic relationships and gene rearrangements of Archaeobatrachia.

Compared with most other vertebrates, this mitogenome appears a tandem duplication of tRNAMet gene. The tRNATrp gene of Oreolalax major translocates from the "WANCY" tRNA cluster to upstream of D-loop. As the first report of the mitogenome sequence from the genus Oreolalax, it will provide fundamental data for further research of phylogeny and biogeography with this genus.

This study will facilitate the further research of the population genetics of this species and systematic study of the genus Oreolalax.

A new toad of Oreolalax Myers & Leviton, 1962 (Anura, Megophryidae) from Sichuan Province, southwest China.

Project description:A new species of the genus Oreolalax Myers & Leviton, 1962 is described from Sichuan Province, southwest China. Molecular phylogenetic analyses based on mitochondrial gene sequences clustered the new species as an independent clade nested with O.rugosus, O.liangbeiensis, and O.major. The new species could be distinguished from its congeners by a combination of the following characters: body size moderate (39.8-52.8 mm in male); head broad; tympanum absent; interorbital region with dark triangular pattern; 1/3 toes webbed, with broad lateral fringes, belly smooth, brown yellow or medium yellow scattered variable brown spots; skin on dorsum relatively rough with fine tiny and large warts granules; middle pectoral glands are evident in males; flanks with dark-brown warts granules; upper surface of limbs with dark bars; and iris orange above and creamy-white below. The new species inhabits subtropical alpine scrub and swamp.

The complete mitochondrial genome of the Baishanzu horned toad Boulenophrys baishanzuensis (Anura: Megophryidae).

Project description:The mitochondrial genome (mitogenome) of Boulenophrys baishanzuensis (Anura: Megophryidae) was sequenced by the Illumina platform. The assembled circular mitogenome of B. baishanzuensis had a total length of 17,040 bp, with a GC content of 41.25%. It consisted of 13 protein-coding genes (PCGs), two rRNA genes, 22 tRNA genes, and a D-loop region. The majority of the PCGs were encoded by the H-strand, while one PCG (nad6) and eight tRNA genes (tRNA-Gln, tRNA-Ala, tRNA-Asn, tRNA-Cys, tRNA-Tyr, tRNA-Ser2, tRNA-Glu, and tRNA-Pro) were encoded in the L-strand. Phylogenetic analysis revealed that the newly sequenced species formed a clade with other Boulenophrys species, while the genus Boulenophrys itself formed a sister group with the genus Atympanophrys.
