The complete mitochondrial genome sequence of Himalayan toad Duttaphrynus himalayanus (Anura: Bufonidae).

ABSTRACT: In this study, the complete mitochondrial genome of Duttaphrynus himalayanus was sequenced adopting Illumina high-throughput sequencing method. The complete mitogenome of the species was 17,172 bp in length, including 13 protein-coding genes, 22 transfer RNA (tRNA) genes, two ribosomal RNA genes, and a non-coding control region (CR). The overall base composition of mitogenome was 29.7% A, 29.6% T, 26.0% C, and 14.7% G. Most mitochondrial genes are encoded on the heavy strand, only ND6 and eight tRNA genes on the light strand. The complete mitogenome of D. himalayanus can provide an important data for future studies on phylogenetic relationship and population genetics of this species.

Project description:The Asiatic toad Bufo gargarizans belongs to Bufonidae. This species is known from the Russian Far East, central, northern and north-eastern China, the Democratic People's Republic of Korea and Japan. In this study, the complete mitochondrial genome of B. gargarizans was sequenced. The mitogenome was 17,407 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a non-coding control region. As in other vertebrates, most mitochondrial genes are encoded on the heavy strand, except for ND6 and eight tRNA genes which are encoded on the light strand. The overall base composition of the B. gargarizans is 28.9% A, 28.2% T, 27.5% C, and 15.3% G. Phylogenetic analysis showed B. gargarizans was closely related to B. bankorensis and B. tibetanus. The complete mitogenome of B. gargarizans can provide an important data for the studies on phylogenetic relationship and population genetics to further explore the taxonomic status of this species.

The complete mitochondrial genome of Bufotes zamdaensis (Anura: Bufonidae).

Project description:The complete mitochondrial genome sequence of Bufotes zamdaensis was reported in the present research by shotgun sequencing. The total length of mitogenome is 17,189 bp, and contains 13 protein-coding genes, 22 tRNA genes, two ribosome RNA genes, and one D-loop region. All gene conformations were consistent with the gene arrangement in vertebrates. Most of the genes of B. zamdaensis were distributed on the H-strand, except for the ND6 subunit gene and eight tRNA genes which were encoded on the L-strand. Phylogenetic analysis of B. zamdaensis and 12 other closely amphibian species was reconstructed using Maximum-likelihood methods. The sequences of B. zamdaensis were clustered in genus Bufotes. The phylogenetic analyses based on these mitogenomes presented here will be useful to further insights on the evolutionary relationships of Bufonidae.

Project description:The complete mitogenome of Leishan moustache toad, Vibrissaphora leishanensis, is determined. The mitogenome size is 17,485 bp, containing 13 protein-coding genes, 2 rRNA genes, 23 tRNA genes and a control region (D-loop). The base composition of the whole genome is 28.1% A, 32.6% T, 24.4% C and 14.8% G. As observed in this genus before, this mitogenome also appears a tandem duplication of tRNAMet gene. This study will provide fundamental data for further research to resolve population genetics of this species and systematic problems of the genus Vibrissaphora.

The complete mitochondrial genome of the Baishanzu horned toad Boulenophrys baishanzuensis (Anura: Megophryidae).

Project description:The mitochondrial genome (mitogenome) of Boulenophrys baishanzuensis (Anura: Megophryidae) was sequenced by the Illumina platform. The assembled circular mitogenome of B. baishanzuensis had a total length of 17,040 bp, with a GC content of 41.25%. It consisted of 13 protein-coding genes (PCGs), two rRNA genes, 22 tRNA genes, and a D-loop region. The majority of the PCGs were encoded by the H-strand, while one PCG (nad6) and eight tRNA genes (tRNA-Gln, tRNA-Ala, tRNA-Asn, tRNA-Cys, tRNA-Tyr, tRNA-Ser2, tRNA-Glu, and tRNA-Pro) were encoded in the L-strand. Phylogenetic analysis revealed that the newly sequenced species formed a clade with other Boulenophrys species, while the genus Boulenophrys itself formed a sister group with the genus Atympanophrys.

Complete mitochondrial genome of the Alpine Metacarpal-tubercled Toad Leptobrachella alpina (Amphibia, Anura, Megophryidae).

Project description:The complete mitochondrial genome of the Leptobrachella alpina Fei, Ye, and Li 1990, was assembled for the first time. The mitogenome of this species was 17,763 bp in length, containing 13 protein-coding genes, two ribosomal RNA genes (12S rRNA and 16S rRNA), 22 transfer RNA genes (tRNA), and a non-coding control region (D-loop). The base content of the mitogenome was that A, T, G, and C occupied 28.5%, 30.8%, 15.1%, and 25.6%, respectively. The phylogenetic analysis was conducted based on 17 complete mitogenome sequences of the family Megophryidae by the Bayesian inference approach. The phylogenetic tree suggested that Leptobrachium and Oreolalax clustered into a clade and formed a sister group with Leptobrachella. This work is critical for the further genetic research and conservation of this species.

It has a tandem duplication of tRNA-Met gene and a translocation of tRNA-Trp gene compared with typical vertebrate mitogenomes. Phylogenetic analyses based on the mitogenome sequences of 18 Megophryidae species were conducted to access phylogenetic assignments of O. xiangchengensis.

First near complete mitochondrial genome of large toothed toad, Oreolalax major (Anura: Megophryidae) from southwest China.

Project description:In this study, the near complete mitogenome sequence (15,469 bp) of Oreolalax major was determined using polymerase chain reaction (PCR). It includes 13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes and 19 transfer RNA (tRNA) genes (GenBank accession number KU310894). The features of O. major have one more tRNA gene (tRNAMet ) behind the original one before ND2 which is similar to Leptobrachium boringii. Phylogenetic analyses were based on the concatenated sequences of the 13 protein-encoding genes of O. major and other related species.

The complete mitochondrial genome sequence of Rana dabieshanensis (Anura: Ranidae).

Project description:Rana dabieshanensis is a species of frog within the family Ranidae. In this study, we assembled a complete mitochondrial genome (mito-genome) for R. dabieshanensis by high-throughput sequencing technology. It is 18,291 bp and includes 13 protein-coding genes, 22 tRNA genes, two rRNA genes and one control region. The nucleotide composition is A: 27.0%, T: 28.6%, C: 29.3% and G: 15.2%. Two overlaps among the 13 protein-coding genes were found: ATP8/ATP6, ND4L/ND4. The study of phylogenetic analysis based on complete mitochondrial genome showed that there was close genetic relationship between R. dabieshanensis and R. omeimontis and it is useful for systematic analyses of genus Rana.

Anti-Trypanosomal Bufadienolides from the Oocytes of the Toad Rhinella alata (Anura, Bufonidae).

Project description:Amphibians are widely known as a prolific source of bioactive metabolites. In this work, we isolated and characterized compounds with antiparasitic activity from the oocytes of the toad Rhinella alata collected in Panama. Bio-guided isolation and structural elucidation were carried out using chromatographic and spectroscopic techniques, respectively. The organic extract was subjected to solid phase extraction followed by HPLC purification of the fraction with in vitro activity against Trypanosoma cruzi trypomastigotes. Seven steroids (1-7) of the bufadienolide family were isolated, and their structures were determined using NMR and MS analyses; of these 19-formyl-dyscinobufotalin, (3) is reported as a new natural product. Compounds 1 and 3-7 resulted in a good anti-trypanosomal activity profile. Among these, 16β-hydroxyl-hellebrigenin (1) and bufalin (7) showed significant selectivity values of >5 and 2.69, respectively, while the positive control benznidazole showed a selectivity of 18.81. Furthermore, molecular docking analysis showed compounds 1, 3 and 7 interact through H-bonds with the amino acid residues GLN-19, ASP-158, HIS-159 and TRP-177 from cruzipain at the catalytic site. Given the lack of therapeutic options to treat American trypanosomiasis, this work can serve as the basis for further studies that aim for the development of bufadienolides or their derivatives as drugs against Chagas disease.

Reconstruction of past distribution for the Mongolian toad, Strauchbufo raddei (Anura: Bufonidae) using environmental modeling.

Project description:The use of ecological models enables determining the current distribution of species, but also their past distribution when matching climatic conditions are available. In specific cases, they can also be used to determine the likelihood of fossils to belong to the same species-under the hypothesis that all individuals of a species have the same ecological requirements. Here, using environmental modeling, we reconstructed the distribution of the Mongolian toad, Strauchbufo raddei, since the Last Glacial Maximum and thus covering the time period between the Late Pleistocene and the Holocene. We found the range of the species to have shifted over time, with the LGM population clustered around the current southern range of the species, before expanding east and north during the Pleistocene, and reaching the current range since the mid-Holocene. Finally, we determined that the ecological conditions during the life-time of the mid-Pleistocene fossils attributed to the species in Europe were too different from the one of the extant species or fossils occurring at the same period in Asia to belong to the same species.
