Characterization of the complete mitochondrial DNA sequence of the Ariosoma meeki (Anguilliformes, Congridae).

ABSTRACT: In this study, the complete mitochondrial genome of Ariosoma meeki was sequenced, assembled and annotated. The circular genome is 16,154 bp in length with nucleotide composition is 28.42% A, 26.53% T, 19.65% G, and 25.40% C and contains 13 protein-coding genes (PCGs), 21 transfer RNA genes (tRNAs), 2 ribosomal RNA unit genes and a large non-coding region (putative control region). To further explore the evolution relationship of the Anguilliformes, we constructed the phylogenetic tree and found that the A. meeki had closer relationship with Ariosoma shiroanago. This study provided the valuable evidence on phylogenetic relationship of the A. meeki at the molecular level and essential resource for further study the molecular phylogenetic, biogeography and adaptive evolution of this lineage.

The mitochondrial genome of B. crocodilinus is of 17,818 bp in length, with overall base composition of 32.11% A, 24.69% T, 16.22% G, and 26.98% C. The genome content includes 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 2 control regions. The result of phylogenetic analysis indicates that B. crocodilinus mitogenome is close to that of Ophisurus macrorhynchos, which are nested within the family Ophichthidae.

The complete mitochondrial genome of Conger erebennus (Jordan & Snyder, 1901) (Anguilliformes: Congridae) from Liaoning Province, China.

Project description:This study aims to explore the complete mitochondrial genome and phylogenetic relationship of Conger erebennus (Jordan & Snyder, 1901) (Anguilliformes: Congridae). The information about the mitogenome was obtained using Illumina's next-generation sequencing technologies. The results revealed that the length of the circular mitogenome was 18,777 bp and it contained 37 genes and three unidentical control regions (CRs). The mitogenome contained 62.78% AT bases with an anti-G bias similar to that in most marine fishes. The maximum-likelihood phylogenetic tree of Anguiliformes based on encoding genes revealed that C. erebennus was in the same clade as other species of Congridae. The results of this study will progress the future of evolutionary research and species identification.

The genome sequence is 17,785 bp in length, comprising 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 2 control regions. Overall base composition is 30.59% A, 24.94% T, 17.01% G, and 27.46% C. The result of phylogenetic analysis indicates that O. rotundus mitogenome is close to that of M. maculosus, which are nested within the family Ophichthidae.

The complete mitochondrial genome of Muraenesox cinereus (Anguilliformes Muraenesocidae).

Project description:In this study, we present the complete mitogenome and a phylogenetic analysis of Muraenesox cinereus determined by long PCR and primer walking methods. The complete mitochondrial genome is a circular molecule of 17,987 bp in length and contains the same set of 37 mitochondrial genes [13 protein-coding genes, 2 ribosomal RNA (rRNA), and 22 transfer RNA (tRNA)], and a control region as other bony fishes. The base composition of the entire mitogenome showed a slight excess of AT bias. The entire mitogenome data produced in this study provides the genomic resources available for future evolutionary studies.

Complete mitochondrial DNA sequence of the endangered fish (Bahaba taipingensis): Mitogenome characterization and phylogenetic implications.

Project description:To understand the systematic status of Bahaba taipingensis within Sciaenidae, the complete mitochondrial genome (mitogenome) sequence of Chinese bahaba has recently been determined by long PCR and primer walking methods. The complete mitochondrial genome is 16500 bp in length and contains 37 mitochondrial genes (13 protein-coding genes, 2 ribosomal RNA genes and 22 transfer RNA genes) as well as a control region (CR) as other bony fishes. Within the control region, we identified the extended termination associated sequence domain (ETAS), the central conserved sequence block domain (CSB-D, SCB-E and CSB-F) and the conserved sequence block domain (CSB-1, CSB-2 and CSB-3). Phylogenetic analyses revealed that Bahaba taipingensis is more closely related to Pseudosciaeniae than Argyrosominae and Sciaeninae. Additionally, Bahaba taipingensis is the sister taxon of Miichthys miiuy, and those two are sister to Collichthys plus Larimichthys.

It consists of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNA genes and a non-coding control region. The overall base composition of the genome is 27.54% for A, 24.80% for T, 31.23% for C and 16.43% for G. The phylogenetic tree, which is based on 12 protein-coding gene sequences, suggested that L. guentheri was closest to L. spadiceus. This study could give impetus to studies focused on population structure and molecular evolution of L. guentheri.

The overall base composition of the genome is 27.58% for A, 25.07% for T, 30.83% for C and 16.52% for G. The phylogenetic tree, which is based on 12 protein coding gene sequences, suggested that L. gloveri was closest to L. lagocephalus. This study could give impetus to studies focused on population structure and molecular evolution of L. gloveri.

Genetic identification of eggs from four species of Ophichthidae and Congridae (Anguilliformes) in the northern East China Sea.

Project description:We report the first genetic identification of eggs of four species of Anguilliformes caught in the northern East China Sea during August 2016, where leptocephali and adults have been collected. The species were Ophisurus macrorhynchos and Echelus uropterus belonging to the Ophichthidae, and Ariosoma majus and Gnathophis heterognathos belonging to the Congridae. The eggs were identified using three molecular genetic markers (mitochondrial 12S rRNA, 16S rRNA, and cytochrome c oxidase subunit 1), sequences obtained from local adult specimens, and geographical distribution data. All eggs were in the early or middle developmental stages. For all species except A. majus, the eggs were found near the range of small leptocephali in the East China Sea and the southern Korean Peninsula, which indicates these species had spawned along the continental near these areas during the summer.

Phylogenetic analysis of the complete mitochondrial genome of Anguilla japonica (Anguilliformes, Anguillidae).

Project description:The Japanese eel (Anguilla japonica), which was listed as Endangered (EN) under the International Union for the Conservation of Nature due to the declines in its abundance, overfishing, and its narrow range distribution. To better protect this species, we described the complete mitogenome of A. japonica in this study. The mitogenome is 16717bp in length and contains 13PCGs, two rRNA genes (12S rRNA and 16S rRNA), 22 tRNA genes, and a putative control region (CR) and one origin of replication on the light-strand (OL). Moreover, the 13 PCGs encode 3813 amino acids in total, all the protein-coding genes use the initiation codon ATG except COI uses GTG. Most of them have TAA or TAG as the stop codon, except COI uses AGG and two PCGs (COII and ND4) use an incomplete stop codon T. A phylogenetic tree based on the Neighbour Joining method was constructed to provide relationship within Anguilla, which could be a useful basis for conservation of this species.
