The complete mitochondrial genome of the Antarctic marbled rockcod, Notothenia rossii (Perciformes, Nototheniidae).

ABSTRACT: The complete mitochondrial genome of Notothenia rossii was obtained using PacBio Sequel long-read sequencing platform. The mitogenome of N. rossii was circular form and 18,274 bp long, which consists of 13 protein-coding genes, 24 tRNAs, 2 rRNAs, and non-coding control region. Particularly, we found duplicated tRNA^Thr and tRNA^Pro in addition to the typical 22 tRNAs. The phylogenetic tree revealed that N. rossii was most closely related to N. coriiceps among species in the Nototheniidae clade within the suborder Notothenioidei.

The complete mitochondrial genome of P. borchgrevinki was sequenced in this study. The genome sequence is 17,299?bp in length, which comprises 13 protein-coding genes, 22 tRNAs, 2 rRNAs and a control region. The overall base composition is 20.45% G, 25.11% A, 29.46% T and 24.98 C%, with an A:T content of 54.57%.

Complete mitochondrial genome of Trematomus newnesi (Perciformes, Nototheniidae).

Project description:The complete mitochondrial genome of Trematomus newnesi was sequenced using an Illumina platform. The 18,602 bp mitogenome contains 13 protein-coding genes, two rRNAs, and 23 tRNAs (tRNAMet is duplicated). The eight stop codons are TAA, TAG, CTT, GTA, AAT, ACT, AGG, and TTA. Two start codons ATG and GTG are present. The GC content is 44.4% and AT content is 55.6%. A phylogenetic tree was generated using 13 species from three families. The results showed that T. newnesi is closely related to Pagothenia borchgrevinki in Nototheniidae. This study provides fundamental data for further genetic evolutionary studies on T. newnesi.

The complete mitochondrial genome of Trematomus loennbergii (Perciformes, Nototheniidae).

Project description:The complete mitochondrial genome of Trematomus loennbergii was studied using NGS technology with PacBio platform. The mitochondrial genome size was 19,374bp and it had 13 protein-coding genes, 22 tRNAs and 2 rRNAs. There were 4 types of stop codons which were TAA, TAG, AGG and T(AA) but start codon type was only one (ATG). The contents of GC were 44.09% and AT contents were 55.91%. To conduct phylogenetic analysis, 12 species in 3 families were used. The result suggested that T. loennbergii was close to Pagothenia borchgrevinki in Nototheniidae. This study would provide a fundamental data for molecular evolution of T. loennbergii.

Project description:In this study, the complete mitochondrial genome of the marbled eelpout, Lycodes raridens Taranetz & Andriashev, 1937 was sequenced using the primer walking method. The mitogenome was 16,569 bp in length and encoded with 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and one Non-Coding Region. The overall nucleotide composition of L. raridens is 25.5%, 25.3%, 18.7%, and 30.5% for A, T, G, and C, respectively. Phylogenetic analysis using the ML method showed that L. raridens was clustered into one branch with L. ygreknotatus and L. toyamensis.

The complete mitochondrial genome sequence and gene organization of Trematomus bernacchii (Perciformes: Nototheniidae) with phylogenetic consideration.

Project description:In this study, the complete mitochondrial DNA sequence was obtained from Trematomus bernacchii. The full length of mitogenome was 16 018 bp, including 20 transfer RNA genes, a 16S ribosomal RNA genes and an incomplete 12S rRNA, 12 protein-coding genes and an incomplete ND6 protein-coding gene, and 2 non-coding regions: control region (D-loop) and origin of light-strand (L-strand) replication (OL). Most genes were located on the heavy-strand (H-strand), while ND6 and seven tRNA genes were located on the L-strand. T. bernacchii's mitogenome was comprised of 24.52% for A, 25.44% for C, 20.38% for G and 29.66% for T, with a higher A + T content (54.18%). From the NJ phylogenetic tree, it revealed that T. bernacchii's was genetically closest to species Dissostichus eleginoides among seven species within suborder Notothenioidei. This study could lay the foundation for the future studies in species identification, taxonomic status, molecular systematics, stock evaluation and conservation genetics for T. bernacchii's.

Project description:Antarctic icefish Chionodraco hamatus is an ideal model for studying cold adaptation mechanisms. The complete mitochondrial genome of C. hamatus was sequenced in this study. The genome sequence is 17 373 bp in length, which comprises 13 protein-coding genes, 22 tRNAs, two rRNAs and a control region. The overall base composition is A: 26.25%, T: 26.05%, G: 17.49% and C: 30.21%, with an A:T content of 52.3%. The phylogenetic analyses based on the concatenated nucleotide sequences of all the genes and control region supported a relatively close relationship with Parachaenichthys. charcoti. The complete mitochondrial genome sequence will be useful for evolutionary and functional studies of Antarctic Notothenioids.

Comparison of Fatty Acid Contents and MMP-1 Inhibitory Effects of the Two Antarctic Fish, Notothenia rossii and Champsocephalus gunnari.

Project description:Total fatty-acid (FA) contents of different organs (stomach, liver, brain, and skin) of two Antarctic fish, marbled rockcod (Notothenia rossii) and mackerel icefish (Champsocephalus gunnari), were examined using gas chromatography-mass spectrometry (GC-MS). N. rossii possessed higher contents of total omega-3, where eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), the most represented omega-3 FAs, were distributed throughout all parts of the fish. The highest level of EPA was observed in the skin and that of DHA was observed in the brain of N. rossii. C. gunnari showed organ peculiarity in that most of the omega-3 FAs were found in stomach and skin. Specifically, the highest levels of EPA and DHA were both observed in the stomach. Although N. rossii and C. gunnari both inhabit the Antarctic Southern Oceans, their characteristics in terms of the composition of fatty acids were shown to vary. The extracts were also evaluated for matrix metalloproteinase-1 (MMP-1)-inhibitory activities in UVB-induced human dermal fibroblasts, where extracts of the skin and liver of N. rossii showed the most significant inhibition upon MMP-1 production. These findings provide experimental evidence that the extracts of the Antarctic fish could be utilized as bioactive nutrients, particularly in the enhancement of skin health.

Project description:The complete mitochondrial genome of Lycodes tanakae was sequenced for the first time from its muscle tissue using the next-generation sequencing method. Its mitochondrial genome was 16,594 base pairs in length, containing 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and one control region. Its overall A, C, G, and T contents were 25.6%, 30.6%, 18.7%, and 25.2%, respectively. Its, A + T content (50.8%) was slightly higher than its G + C content (49.2%). A phylogenetic tree was built using 10 belonging to the order Perciformes and two species belonging to the order Scorpaeniformes.

The complete mitochondrial genome was 16,513 bp in length and contained 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and 2 non-coding region (the control region and the origin of light strand replication). The overall base composition was A 26.37%, T 25.61%, C 30.80%, and G 17.22%. All protein-coding genes started with an ATG initiation codon, except COI used GTG. With the exception of ND6, all other genes were encoded on the heavy strand, the NJ tree demonstrated that A. macropterus has a closest relationship with Cheilodipterus quinquelineatus and Apogon semilineatus.
