Complete mitochondrial genome analysis of Lingula anatina from Korea (Brachiopoda, Lingulida, Lingulidae).

ABSTRACT: In this study, complete mitochondrial genome of the Lingula anatina (Lamark, 1801) from Korea has been sequenced and analysed, and compared with previous complete mitochondrial genome record from Japan. The mitogenome is 25,790?bp and composed of 13 protein-coding genes, two ribosomal RNA and 34 tRNA. In comparison with previous record, there are dramatically changes in structure between two records. Additionally, phylogenetic tree of L. anatina in Brachiopoda reconstructed due to 12 protein-coding genes of mitochondrial genome. The results showed that the Korean L. anatina positioned in Brachiopoda and the closest species is the L. anatina from the Japan. This study provides the second complete mitochondrial genome for the species.

Project description:In this study a complete mitochondrial genome of the species, Laqueus japonicus was sequenced and analysed. The mitochondrial genome size is 14,267?bp with 20.2% A, 15.7% C, 27.1% G, and 37.0% T nucleotide distributions. This is the second complete mitochondrial genome record from the genus Laqueus and first record for the species. Genome structure and gene orientation are identical with previous record of the genus. In addition, phylogenetic relationship of L. japonicus in the subphylum Rhynchonelliformea was investigated by using protein coding genes of complete mitochondrial genome. The present study suggests that the closest species to L. japonicus is L. rubellus and they belong to the family Laqueidae.

Analysis of the complete mitochondrial DNA sequence of the brachiopod terebratulina retusa places Brachiopoda within the protostomes.

Project description:Brachiopod phylogeny is still a controversial subject. Analyses using nuclear 18SrRNA and mitochondrial 12SrDNA sequences place them within the protostomes but some recent interpretations of morphological data support a relationship with deuterostomes. In order to investigate brachiopod affinities within the metazoa further, we compared the gene arrangement on the brachiopod mitochondrial genome with several metazoan taxa. The complete (15 451 bp) mitochondrial DNA (mtDNA) sequence of the articulate brachiopod Terebratulina retusa was determined from two overlapping long polymerase chain reaction products. All the genes are encoded on the same strand and gene order comparisons showed that.only one major rearrangement is required to interconvert the T. retusa and Katharina tunicata (Mollusca: Polvplacophora) mitochondrial genomes. The partial mtDNA sequence of the prosobranch mollusc Littorina saxatilis shows complete congruence with the T. rehtusa gene arrangement with regard to the ribosomal and protein coding genes. This high similarity in gene arrangement is the first to be reported within the protostomes. Sequence analyses of mitochondrial protein coding genes also support a close relationship of the brachiopod with molluscs and annelids, thus supporting the clade Lophotrochozoa. Though being highly informative, sequence analyses of the mitochondrial protein coding genes failed to resolve the branching order within the lophotrochozoa.

Complete mitochondrial genome of Gloydius saxatilis (Viperidae: Crotalinae) from Korea.

Project description:In this study, we sequenced the complete mitochondrial genome of Gloydius saxatilis using Illumina next-generation sequencing. The total length of the mitogenome was 17,223 bp, and contained 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), two non-coding control regions (CRs), and the origin of light (OL)-strand replication. The genome structure and order of the genes were similar to other Crotalinae species. Phylogenetic analysis based on the 13 concatenated PCGs indicated that G. saxatilis closely related to G. intermedius and, G. shedaoensis.

The overall base composition of the complete genome is 22.94% A, 43.32% T, 25.72% G, and 8.02% C, with a high A + T content of 66.26%.

Complete mitochondrial genome and phylogenetic analysis of Callorhinus ursinus: an endangered species from South Korea.

Project description:We present the complete mitochondrial genome and a phylogenetic analysis of Callorhinus ursinus, the northern fur seal, determined using Illumina next-generation sequencing (NGS) technology. The total length of the mitogenome was 17,154 bp, which consisted of 13 protein-coding genes, two ribosomal RNA genes, 22 tRNA genes, and one control region. The base composition of the entire mitogenome was 33.5% (A), 26.3% (C), 13.9% (G), and 26.3% (T) with an A + T bias of 59.8%. The control region contained two types of tandem repeats. A neighbour-joining (NJ) tree was constructed and comprised two clades with C. ursinus forming a monophyletic group. Data produced in this study will aid exploration of the genetic diversity of endangered C. ursinus and contribute to molecular identification of this species.

The complete mitochondrial genome of Ricania shantungensis (Hemiptera: Ricaniidae) in Korea.

Project description:In this study, we have sequenced and annotated the complete mitochondrial genome of Ricania shantungensis (Hemiptera: Ricaniidae) for the first time. The circular mitogenome of R. shantungensis was 15,789 bp, including 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single control region of 1,363 bp. Its AT ratio was 74.6%. According to the phylogenetic tree, R. shantungensis was clustered with the genus Ricania.

Complete mitochondrial genome of Acanthochitona defilippii (Polyplacophora: Chitonida) from South Korea.

Project description:The chiton (Polyplacophora) occupies a significant position in molluscan evolutionary history as one of the most primitive groups within the phylum Mollusca. Acanthochitona defilippii (Tapparone-Canefri 1874) (Chitonida: Acanthochitonidae) is a commonly found intertidal chiton species in South Korea. In this study, we characterized the complete mitochondrial genome of A. defilippii (14,999 bp long), comprising 13 protein-coding genes (PCGs), 22 transfer RNA genes, two ribosomal RNA genes, and an A + T rich region (166 bp). The base composition is as follows: 31.82% for A, 11.63% for C, 16.69% for G, and 39.86% for T. We reconstructed a maximum likelihood (ML) tree to elucidate phylogenetic relationships among the eight chitonid families using the nucleotide sequences of all PCGs. The ML tree revealed that A. defilippii clustered with Acanthochitona avicula (BP 100) within the family Acanthochitonidae. Acanthochitonidae formed a sister group with Mopaliidae. The results could provide a valuable understanding the phylogenetic relationships of chitonid species.

The circular mitochondrial genome is 15,739 bp and consists of 13 protein-coding, two ribosomal RNAs, and 22 transfer RNA genes (GenBank accession no. MW770891). Results of phylogenetic analysis indicate that the species clustered with other species of the family Chironomidae. This study is helpful to the identification of C. flaviplumus larvae, which is difficult to be identified by morphology.

The complete mitochondrial genome of Buccinum bayani (Gastropoda: Buccininae) in Korea.

Project description:This study is the first to report the complete 15,262 base pair mitochondrial genome of Buccinum bayani, consisting of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNA genes, and a control region (D-loop). The overall base composition of the complete genome is A (29.87%), T (39.11%), G (15.86%), and C (15.16%), with a high AT content of 68.98%. Phylogenetic analysis showed that B. bayani was most closely related to B. undatum. The mitogenome of B. bayani will provide helpful information to further our phylogenetic and evolutionary understanding of the family Buccininae.
