Complete mitochondrial genome of Chironomus flaviplumus (Diptera: Chironomidae) collected in Korea.

ABSTRACT: The complete mitochondrial genome of Chironomus flaviplumus was sequenced. The circular mitochondrial genome is 15,739 bp and consists of 13 protein-coding, two ribosomal RNAs, and 22 transfer RNA genes (GenBank accession no. MW770891). Results of phylogenetic analysis indicate that the species clustered with other species of the family Chironomidae. This study is helpful to the identification of C. flaviplumus larvae, which is difficult to be identified by morphology.

Project description:A non-biting midge Chironomus yoshimatsui has been widely used in ecotoxicology and chemical risk assessments. In this study, the complete mitochondrial genome (mitogenome) sequence of C. yoshimatsui was determined using short-read next-generation sequencing technologies. The mitogenome was 15,734 bp in length and consisted of 13 protein coding genes, 2 ribosomal RNAs, and 22 transfer RNAs. The A + T content was 77.8%. The gene order was identical to the pattern conserved across Diptera. The mitocgenome sequence obtained in this study provides a useful resource for further evolutionary and ecological studies.

Complete mitochondrial genome of Limnophyes minimus (Diptera: Chironomidae).

Project description:The complete mitochondrial genome of Limnophyes minimus (Meigen 1818) was sequenced and annotated, and its general features and base composition were analyzed. The phylogenetic relationships of the families Chironomidae, Simuliidae, Sciaridae and Culicidae based on 25 metagenomes were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that L. minimus belongs family Chironomidae, which is consistent with the traditional morphological classification.

The complete mitochondrial genome of Propsilocerus akamusi (Diptera, Chironomidae).

Project description:Propsilocerus akamusi is one of the most important genera of Chironomidae. However, the systemically classification and taxonomic studies have so far been limited. In this study, we report the complete mitochondrial genome sequence of P. akamusi. The mitogenome has 15,632 base pairs (77.58% A + T content) and made up of total of 37 genes (13 protein-coding, 22 transfer RNAs, and 2 ribosomal RNAs). This study will provide useful genetic information for future phylogenetic and taxonomic classification of Chironomidae.

Karyotypes of Chironomus Meigen (Diptera: Chironomidae) species from Africa.

Project description:The karyotypes of six African Chironomus species (Chironomus alluaudi Kieffer, 1913, Chironomus transvaalensis Kieffer, 1923, Chironomus sp. Nakuru, Chironomus formosipennis Kieffer, 1908, Chironomus prope pulcher Wiedemann, 1830, Chironomus sp. Kisumu) were investigated; four of these karyotypes were described for the first time (Chironomus sp. Nakuru, Chironomus formosipennis, Chironomus prope pulcher, Chironomus sp. Kisumu). Of the six Chironomus karyotypes, three had "pseudothummi" cytocomplex chromosome arms combinations AE CD BF G (Chironomus alluaudi, Chironomus transvaalensis, Chironomus sp. Nakuru), two had "thummi"cytocomplex arms combinations AB CD EF G (Chironomus formosipennis, Chironomus prope pulcher), and one had "parathummi"armcombinations AC BF DE G (Chironomus sp. Kisumu). Thus, three of the ten main cytocomplexes known were detected in Africa. Detailed photomaps of all chromosome arms, with the exception of arms B and G, were prepared for the karyotypes of Chironomus alluaudi, Chironomus transvaalensis, Chironomus sp. Nakuru, Chironomus prope pulcher; the karyotypes of Chironomus formosipennis, Chironomus sp. Kisumucould only be fragmentarily mapped. Endemic African banding sequences were characteristic for most of the chromosomal arms in all species studied. However, basic sequences, which can be present in different Chironomus species on different continents (Wülker, 1980; Kiknadze et al. 2008), were also detected also in several African species (Chironomus alluaudi, Chironomus sp. Nakuru, and Chironomus formosipennis). The banding sequences of African species studied allow discussion of the derivation of modern banding patterns from hypothetical species, living before separation of cytocomplexes and continents.

Complete mitochondrial genome of Aedes flavopictus (Yamada, 1921) (Diptera: Culicidae) collected in South Korea.

Project description:In this study, we determined for the first time the mitochondrial genome sequence of an Aedes flavopictus specimen collected in South Korea. Its mitochondrial genome was 16,060 bp in length, consisting of 13 protein-coding, 22 tRNA, and 2 rRNA genes and a non-coding A + T rich region. The overall base composition in the heavy strand was 39.7, 8.6, 12.7, and 39% of A, G, C, and T, respectively, and the G + C content was 21.2%. Phylogenetic analysis revealed that Aedes spp. formed a monophyletic clade.

Complete mitochondrial genome of the non-biting midge Polypedilum henicurum Wang, 1995 (Diptera: Chironomidae).

Project description:The complete mitochondrial genome of Polypedilum henicurum was sequenced and analyzed. The whole mitogenome is 15,964 bp in length and contains 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), and two ribosomal RNAs (rRNAs). The overall nucleotide composition is 39.5% A, 39.0% T, 13.1% C, and 8.4% G. Most PCGs start with ATN codon excluding COX1 (TTG) and all PCGs end with TAA codon. Gene arrangement stays the same with the ancestral mitochondrial genome. Bayesian inference phylogenetic tree supports its close relationship with P. unifascium. This work enriches the library of Chironomidae mitochondrial genomes and provides a valuable resource for understanding the evolutionary history of Polypedilum.

Chromosomal organization of the ribosomal RNA genes in the genus Chironomus (Diptera, Chironomidae).

Project description:Chromosomal localization of ribosomal RNA coding genes has been studied by using FISH (fluorescence in situ hybridization) in 21 species from the genus Chironomus Meigen, 1803. Analysis of the data has shown intra- and interspecific variation in number and location of 5.8S rDNA hybridization sites in 17 species from the subgenus Chironomus and 4 species from the subgenus Camptochironomus Kieffer, 1914. In the majority of studied species the location of rDNA sites coincided with the sites where active NORs (nucleolus organizer regions) were found. The number of hybridization sites in karyotypes of studied chironomids varied from 1 to 6. More than half of the species possessed only one NOR (12 out of 21). Two rDNA hybridization sites were found in karyotypes of five species, three - in two species, and five and six sites - in one species each. NORs were found in all chromosomal arms of species from the subgenus Chironomus with one of them always located on arm G. On the other hand, no hybridization sites were found on arm G in four studied species from the subgenus Camptochironomus. Two species from the subgenus Chironomus - Chironomusbalatonicus Devai, Wuelker & Scholl, 1983 and Chironomus "annularius" sensu Strenzke, 1959 - showed intraspecific variability in the number of hybridization signals. Possible mechanisms of origin of variability in number and location of rRNA genes in the karyotypes of species from the genus Chironomus are discussed.

The complete mitochondrial genome of a non-biting midge Polypedilum unifascium (Tokunaga, 1938) (Diptera: Chironomidae).

Project description:The complete mitochondrial genome of Polypedilum unifascium (Diptera: Chironomidae) was determined by Illumina sequencing technology. The whole mitogenome is 16,452 bp in length with an A + T bias of 79.3%, and contains 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), and two ribosomal RNAs (rRNAs). All PCGs start with ATN codon and use TAA as the stop codon. Gene arrangement of the 13 PCGs is identical to that of other known Chironomidae mitochondrial genomes. The resultant Bayesian inference and maximum-likelihood trees based on the sequence data of 13 PCGs support its close relationship with P. vanderplanki.

Results of maximum likelihood analysis showed that this species clustered with other species of the family Chironomidae. This study will contribute to the phylogenetics of genus Stictochironomus and the other genera of Chironomidae.

Characterization of the complete mitochondrial genome of Odontobutis platycephala collected from Nakdong River, South Korea.

Project description:The complete mitochondrial genome of Odontobutis platycephala collected from a native Korean river was determined by the bioinformatics assembly of the next-generation sequencing (NGS) reads. The circular mitogenome was 17,590 bp length which harbored canonical 13 protein-coding genes, 22 tRNAs, and 2 rRNAs, which was identical to those of family Odontobutidae. Twenty-eight genes were located on H strand, whereas remaining nine genes were on L strand. Except for COX1 gene (GTG), other 12 protein-coding genes were predicted typical start codons (ATG). Among the currently known mitogenome sequences, O. platycephala showed highest identity (96.98%) to Korean haplotype of O. platycephala (NC010199).
