The complete mitochondrial genome of Gracilaria textorii (Gracilariales, Florideophyceae).

ABSTRACT: In this study, we determined the mitochondrial genome sequence of Gracilaria textorii. The mitogenome of G. textorii is 25,743?bp in length, with GC content of 28.2%, and includes 25 protein-coding genes, two rRNA genes (rns and rnl), and 22 tRNA genes. The overall nucleotide composition of G. textorii includes 33.9% A, 14.1% C, 14.1% G, and 28.2% T. The newly sequenced mitogenome data were used for reconstruction of phylogenetic relationships in red algae, and the results suggested that G. textorii was closely related to G. salicornia.

Project description:Gracilaria chilensis is an economically important marine alga. In this study, we obtained complete mitogenome of G. chilensis by high-throughput sequencing, which was mapped as a circular molecule of 26,897?bp with 27.56% GC content and was identified 53 genes, including 25 protein-coding genes, 2 rRNA genes, 26 tRNA genes, and 1 group II intron inserted into the trnI gene. In addition, a 162-bp stable stem loop was found in intergenic regions, which was most likely associated with DNA transcription and replication. The Bayesian phylogenetic tree of Gracilariaceae revealed that G. chilensis and G. salicornia and G. changii shared a closer relationship than G. vermiculophylla in the genus Gracilaria.

Complete sequence of mitochondrial DNA of Gracilaria tenuistipitata (Rhodophyta).

Project description:We sequenced and analysed the complete mitogenome of Gracilaria tenuistipitata. The complete G. tenuistipitata mitogenome is 25,819 bp long, containing 50 genes, including 24 protein-coding genes, one intron, two rRNA genes, 23 tRNA genes, and one unidentified open reading frame. Twenty-three of the 24 (95.83%) protein-coding genes ended with the TAA stop codon, and one (4.17%) with TAG (rps3 gene). All protein-coding genes in G. tenuistipitata were found to contain the start codon ATG. Phylogenetic analysis revealed that G. tenuistipitata clustered with G. chilensis. The complete mitochondrial genome sequence provided here should be useful for elucidation of Gracilaria evolution.

Project description:Gracilaria edulis, a marine red macroalgae, is a rich source of sulfated polysaccharides, carbohydrate, vitamins, and minerals, and showed multiple bioactivities such as antibacterial, antitumour, and cholinesterase inhibitory activity. The plastid genome sequence of G. edulis is 179,410?bp. A total of 235 genes were determined, including 201 protein-encoding genes, 30 tRNA genes, 3 rRNA genes, 1 ribonuclease gene, and 1 intron inserted into the trnM gene. Phylogenetic analysis showed that G. edulis clustered together with Gracilaria salicornia, Gracilaria tenuistipitata var. liui and Gracilaria chilensis. The plastid genome analysis will help in the understanding of Gracilaria evolution.

Phylogenetic analysis of the complete chloroplast genome of Gracilaria vermiculophylla.

Project description:In this study, we sequenced and annotated the complete chloroplast genome of Gracilaria vermiculophylla (GenBank accession number: MN853882). The total length of the chloroplast genome of G. vermiculophylla is 180,262 bp, including 191 protein-encoding genes, 30 tRNA genes and 3 rRNA genes. The phylogenetic tree, which is based on core genes, shows that G. vermiculophylla clustered into the Gracilaria clade and has close genetic relationships with algae Gracilaria tenuistipitata and Gracilaria changii. These data will enable a better understanding of the phylogenetic status of G. vermiculophylla.

Phylogenetic analysis showed that G. bailiniae clustered together with Gracilaria chorda and Gracilariopsis lemaneiformis. The complete plastid genome provided in this work would be useful for elucidation of Gracilaria evolution.

The complete G. tenuistipitata var. liui mitogenome length was 25,879 bp and contained 50 genes including 24 protein-coding, 2 rRNA, and 23 tRNA genes and one unidentified open reading frame (ORF). Of the 24 protein-coding genes, 23 (95.83%) terminated with the stop codon TAA, and one (4.17%) with TAG (rps3 gene). All protein-coding genes in G. tenuistipitata var. liui started with ATG codon. Mitogenome phylogenetic analysis revealed that G. tenuistipitata var. liui firstly clustered together with Gracilaria tenuistipitata. The complete mitogenome sequence would help in understanding Gracilaria evolution.

Highly Conserved Mitochondrial Genomes among Multicellular Red Algae of the Florideophyceae.

Project description:Two red algal classes, the Florideophyceae (approximately 7,100 spp.) and Bangiophyceae (approximately 193 spp.), comprise 98% of red algal diversity in marine and freshwater habitats. These two classes form well-supported monophyletic groups in most phylogenetic analyses. Nonetheless, the interordinal relationships remain largely unresolved, in particular in the largest subclass Rhodymeniophycidae that includes 70% of all species. To elucidate red algal phylogenetic relationships and study organelle evolution, we determined the sequence of 11 mitochondrial genomes (mtDNA) from 5 florideophycean subclasses. These mtDNAs were combined with existing data, resulting in a database of 25 florideophytes and 12 bangiophytes (including cyanidiophycean species). A concatenated alignment of mt proteins was used to resolve ordinal relationships in the Rhodymeniophycidae. Red algal mtDNA genome comparisons showed 47 instances of gene rearrangement including 12 that distinguish Bangiophyceae from Hildenbrandiophycidae, and 5 that distinguish Hildenbrandiophycidae from Nemaliophycidae. These organelle data support a rapid radiation and surprisingly high conservation of mtDNA gene syntheny among the morphologically divergent multicellular lineages of Rhodymeniophycidae. In contrast, we find extensive mitochondrial gene rearrangements when comparing Bangiophyceae and Florideophyceae and multiple examples of gene loss among the different red algal lineages.
