Sequencing and characterization of mitochondrial genome of Eurytoma sp. (Hymenoptera: Eurytomidae).

ABSTRACT: The mitochondrial genome (mitogenome) of Eurytoma sp. (Hymenoptera: Eurytomidae: Eurytominae) was determined in this study. The sequenced region is 14,946?bp with an A?+?T content of 82.65%, including 13 protein-coding genes, 2 ribosomal RNAs, 18 transfer RNAs, and a partial sequence of AT-rich region. All PCGs begin with typical ATN codons and use standard canonical TAA as their termination codons, except for atp6 gene, which stops with an incomplete codon T. A comparison of all the sequenced hymenopteran mitogenomes revealed that Eurytoma sp. has dramatic gene rearrangements, some of which is peculiar to Chalcidoidea. What's more, Maximum likelihood (ML) analysis highly supported the monophyly of both Chalcidoidea and Apocrita.

Project description:The mitochondrial genome (mitogenome) of Megachile strupigera (Hymenoptera: Megachilidae: Megachilinae) was determined in our study. The sequenced region is 15,193 bp with an A + T content of 83.44%, including 13 protein-coding genes, two ribosomal RNAs and 19 transfer RNAs. All PCGs are initiated by typical ATN codons and stop with the complete termination codon TAA, except for nad3 gene, which has an incomplete stop codon T. Bayesian method supported the monophyly of both Megachilidae and Apoidea. And within the Apoidea, Apidae and Megachilidae formed a sister clade to Colletidae.

Characterization of the mitochondrial genome of Arge bella Wei & Du sp. nov. (Hymenoptera: Argidae).

Project description:We describe Arge bella Wei & Du sp. nov., a large and beautiful species of Argidae from south China, and report its mitochondrial genome based on high-throughput sequencing data. We present the gene order, nucleotide composition of protein-coding genes (PCGs), and the secondary structures of RNA genes. The nearly complete mitochondrial genome of A. bella has a length of 15,576 bp and a typical set of 37 genes (22 tRNAs, 13 PCGs, and 2 rRNAs). Three tRNAs are rearranged in the A. bella mitochondrial genome as compared to the ancestral type in insects: trnM and trnQ are shuffled, while trnW is translocated from the trnW-trnC-trnY cluster to a location downstream of trnI. All PCGs are initiated by ATN codons, and terminated with TAA, TA or T as stop codons. All tRNAs have a typical cloverleaf secondary structure, except for trnS1. H821 of rrnS and H976 of rrnL are redundant. A phylogenetic analysis based on mitochondrial genome sequences of A. bella, 21 other symphytan species, two apocritan representatives, and four outgroup taxa supports the placement of Argidae as sister to the Pergidae within the symphytan superfamily Tenthredinoidea.

Characterization of the mitochondrial genome of Analcellicampa xanthosoma gen. et sp. nov. (Hymenoptera: Tenthredinidae).

Project description:A new genus with a new species of the tribe Hoplocampini of Hoplocampinae was described from China: Analcellicampa xanthosoma Wei & Niu, gen. et sp. nov. Hoplocampa danfengensis G. Xiao 1994 was designated as the type species of the new genus. The characters of Analcellicampa danfengensis (G. Xiao) comb. nov. were briefly discussed. A key to the tribes and known genera of Hoplocampinae was provided. The nearly complete mitochondrial genome of A. xanthosoma was characterized as having a length of 15,512 bp and containing 37 genes (22 tRNAs, 13 protein-coding genes (PCGs), and 2 rRNAs). The gene order of this new specimen was the same as that in the inferred insect ancestral mitochondrial genome. All PCGs were initiated by ATN codons and ended with TAA or T stop codons. All tRNAs had a typical cloverleaf secondary structure, except for trnS1. Remarkably, the helices H991 of rrnS and H47 of rrnL were redundant, while helix H563 of rrnL was highly conserved. A phylogeny based on previously reported symphytan mitochondrial genomes showed that A. xanthosoma is a sister group to Monocellicampa pruni, with high support values. We suggest that A. xanthosoma and M. pruni belong to the tribe Hoplocampini of Hoplocampinae.

The mitochondrial genome of Vulgichneumon sp. (Hymenoptera: Ichneumonidae).

Project description:The Vulgichneumon sp. belongs to the subfamily Ichneumoninae of Ichneumonidae. The mitogenome (GenBank accession number: MN178162) of Vulgichneumon sp. was sequenced, the first representative of the mitogenome of the subfamily. The nearly complete mitogenome is 15,306 bp totally, consisting of 13 protein-coding genes, 2 rRNAs and 22 transfer RNAs. The nucleotide composition biases toward A and T, which together made up 83.6% of the entirety. Bayesian inference analysis supported the monophyly of Evanioidea, Chalcidoidea, Platygastroidea, Proctotrupoidea, Ceraphronoidea, and Ichneumonoidea. This result also suggested that the clade that contains other six superfamilies was the sister group to the clade that consist of Trigonaloidea, Megalyroidea and Ichneumonoidea.

Sequencing and characterization of mitochondrial genome of Fusarium sp. (Hypocreales: Nectriaceae).

Project description:In this study, we presented the complete mitochondrial genome of Fusarium sp. It has a total length of 47, 74?bp, and the base composition of the mitogenome is as follows: A (34.1%), T (33.5%), C (14.7%), and G (17.7%). The mitogenome contains 23 protein-coding genes, 1 ribosomal RNA (rRNA), and 26 transfer RNA (tRNA) genes, all coded on the same strand of DNA. The gene order is identical to that of the other Fusarium mitogenomes. The taxonomic status of the Fusarium sp. mitogenome exhibits a closest relationship with F. oxysporum, but varied in the structure of mitochondrial genome.

Mitochondrial genome of Chalcidoidea sp. (Hymenoptera: Apocrita: Chalcidoidea) and phylogenetic analysis.

Project description:Chalcidoidea (Hymenoptera) are minute wasps which can attack immature and adult stages of virtually all insect orders. Here, we sequenced and annotated the mitochondrial genome (mitogenome) of Chalcidoidea sp. This mitogenome was 15,152 bp long and encoded 13 protein-coding genes (PCGs), 20 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). All 13 PCGs were initiated by the ATN (ATG, ATT, ATA, and ATC) codon. All PCGs terminate with the stop codons TAA or TAG except for nad4 which ended with the incomplete codon T-. Phylogenetic analysis showed that Chalcidoidea sp. got together with the species Encyrtus infelix and Eurytoma sp., and species in Chalcidoidea formed a sister group to other Cynipoidea and Proctotrupoidea species.

Sequencing and analysis of the complete mitochondrial genome of Habrobracon hebetor (Hymenoptera: Braconidae).

Project description:We determined the complete mitochondrial genome sequence of Habrobracon hebetor (Say). The complete mitogenome sequence of H. hebetor was observed to be a circular molecule 15,708 bp long and consists of 13 protein-coding genes (PCG), 2 ribosomal RNA (rRNA) genes, and 22 transfer RNA (tRNA) genes (GenBank accession no. MN842279). This nucleotide composition is biased toward adenine and thymine (85.2% A + T). The A + T-rich region is found between trnM and trnQ, and this entire region was 864 bp long.

Here, we sequenced and analyzed the full mitogenome of Tetrastichus howardi, an important natural enemy of many lepidopteran pests. Next-generation sequencing of the mitochondrial genome of Bombus longipennis Friese, 1918 (Hymenoptera: Apidae).

Project description:Bombus longipennis is the species of Bumblebees (Hymenoptera: Apidae), which are important pollinators for wild plants and greenhouses crops. The complete mitochondrial genome (mitogenome) of B. longipennis was determined by next-generation sequencing. The mitogenome was 18,458 bp in size with 87.2% A + T content, containing 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and an AT-rich control region (D-loop). Gene arrangement was found to be identical to those of other mitogenomes of bumblebees (e.g. Bombus terrestris and Bombus ignitus). All 13 PCGs initiated with typical ATN codons. Among them, 11 PCGs terminated with TAA or TAG; only cox2 and nad4 have incomplete stop codon T. All 22 tRNAs can be folded into typical cloverleaf structure. Phylogenetic analysis based on the concatenated nucleotide sequences of all 13 PCGs indicated that B. longipennis was more closely related to other species of subgenus Bombus, which clustered into a monophyletic group.

Mitochondrial genome evolution in fire ants (Hymenoptera: Formicidae).

Project description:BackgroundComplete mitochondrial genome sequences have become important tools for the study of genome architecture, phylogeny, and molecular evolution. Despite the rapid increase in available mitogenomes, the taxonomic sampling often poorly reflects phylogenetic diversity and is often also biased to represent deeper (family-level) evolutionary relationships.ResultsWe present the first fully sequenced ant (Hymenoptera: Formicidae) mitochondrial genomes. We sampled four mitogenomes from three species of fire ants, genus Solenopsis, which represent various evolutionary depths. Overall, ant mitogenomes appear to be typical of hymenopteran mitogenomes, displaying a general A+T-bias. The Solenopsis mitogenomes are slightly more compact than other hymentoperan mitogenomes (~15.5 kb), retaining all protein coding genes, ribosomal, and transfer RNAs. We also present evidence of recombination between the mitogenomes of the two conspecific Solenopsis mitogenomes. Finally, we discuss potential ways to improve the estimation of phylogenies using complete mitochondrial genome sequences.ConclusionsThe ant mitogenome presents an important addition to the continued efforts in studying hymenopteran mitogenome architecture, evolution, and phylogenetics. We provide further evidence that the sampling across many taxonomic levels (including conspecifics and congeners) is useful and important to gain detailed insights into mitogenome evolution. We also discuss ways that may help improve the use of mitogenomes in phylogenetic analyses by accounting for non-stationary and non-homogeneous evolution among branches.
