Complete mitochondrial genome of Cyllorhynchites ursulus (Coleoptera: Rhynchitidae).

ABSTRACT: The complete mitochondrial genome of Cylloryhunchites ursulus was sequenced for the first time from its whole body using the next-generation sequencing method. This mitogenome was 14,508 base pairs in length, containing 13 protein-coding genes, 20 transfer RNA genes, and two ribosomal RNA genes. Its overall A, C, G, and T contents were 37.8, 17.8, 11.1, and 33.4%, respectively. Its A?+?T content (71.2%) was higher than G?+?C content. The phylogenetic result showed that although C. ursulus is a species of family Rhynchitidae, its phylogenetic position is closely related to family Attlabidae.

The genome was a circular molecule of 15,499 base pairs (bp), with 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an estimated 830 bp A + T-rich control region. The total base composition of the mitogenome was A-T bias, with 40.9% for A, 37.0% for T, 13.4% for C, and 8.7% for G. The molecular data presented here would be useful for further study of O. diversipes.

Complete mitochondrial genome of Niphades castanea (Coleoptera: Curculionidae).

Project description:Niphades castanea Chao is an important insect pest on many plants which belong to genus Castanea. The complete mitochondrial genome of N. castanea was sequenced and analyzed. The phylogenetic relationships between N. castanea and other 41 species in the family Curculionidae were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that N. castanea is closely related to Hylobitelus xiaoi, which is in accordance with the traditional morphological classification.

First Complete Mitochondrial Genome of Melyridae (Coleoptera, Cleroidea): Genome Description and Phylogenetic Implications.

Project description:To explore the characteristics of the mitogenome of Melyridae and reveal phylogenetic relationships, the mitogenome of Cordylepherus sp. was sequenced and annotated. This is the first time a complete mitochondrial genome has beengenerated in this family. Consistent with previous observations of Cleroidea species, the mitogenome of Cordylepherus sp. is highly conserved in gene size, organization and codon usage, and secondary structures oftRNAs. All protein-coding genes(PCGs) initiate withthe standard start codon ATN,except ND1, which starts with TTG, and terminate with the complete stop codons of TAA and TAG, or incomplete forms, TA- and T-. MosttRNAs have the typical clover-leaf structure, except trnS1 (Ser, AGN), whose dihydrouridine (DHU) armis reduced. In the A+T-rich region, three types of tandem repeat sequence units are found, including a 115 bp sequence tandemly repeated twice, a 16 bp sequence tandemly repeated three timeswith a partial third repeat and a 10 bp sequence tandemly repeated seven times. Phylogenetic analyses based on 13 protein-coding genes by both Bayesian inference (BI) and maximum likelihood (ML) methods suggest that Melyridae sensu lato is polyphyletic, and Dasytinae and Malchiinae are supported as independent families.

Project description:Beetles in the weevil subfamily Platypodinae are among the dominant groups of insects in wet tropical forests, which together with bark and ambrosia beetles in the subfamily Scolytinae. Easily recognised by the circle-shaped entrance holes in fallen logs and a very elongated body shape, they have earned the common name, "pinhole borers". All except two Platypodinae species are ambrosia beetles that cultivate fungi in wood tunnels as the sole food for their larvae. Platypodinae is a peculiar weevil subfamily of species that cultivate fungi in tunnels excavated in dead wood. The Platypodinae is likely the oldest known lineage of fungus-cultivating insects, with an origin of the ambrosial habit more than 80?Ma. Here, we sequenced and characterized the complete mitochondrial genome of E. parallelus, which was collected from logs imported from SierraLeone. The complete circular mitochondrial genome (mitogenome) of Euplatypus parallelus is 16,095?bp in size, containing 37 typical genes and one non-coding AT-rich region. The AT content of the AT-rich region is 87.5%. All protein-coding genes (PCGs) start with standard ATN initiation codons except for nad1and end with complete termination codons TAA except for cox1genes using an incomplete stop codon T. tRNA genes are predicted with a characteristic cloverleaf secondary structure except for trnS1(AGN) , whose dihydrouridine (DHU) arm is replaced by a simple loop. The size of the large and small ribosomal RNA genes are 1386 and 741?bp, respectively.

The complete mitochondrial genome of Sympiezomias velatus (Coleoptera: Curculionidae).

Project description:Here, we determined the complete mitogenome sequence of Sympiezomias velatus (Coleoptera: Curculionidae: Entiminae). This mitogenome was 15,592?bp long with an A?+?T content of 74.1% and contains 13 protein-coding genes (PCGs), 21 transfer RNA genes (tRNAs), 2 ribosomal RNA unit genes and a large non-coding region (putative control region). The trnI gene was not found in the S. velatus mitogenome. The order and orientation of the mitochondrial genes were identical to the inferred ancestral arrangement of insects except for trnR which was changed to be adjacent the nad3 gene. All tRNAs had the typical cloverleaf structure, except for trnS1 which lacked the dihydrouridine arm. The Bayesian phylogenetic tree of 10 Entiminae species based on the concatenated nucleotide sequences of 13 PCGs strong supported a sister relationship of S. velatus and Barynotus obscures.

The complete mitochondrial genome of Spondylis buprestoides (Coleoptera: Cerambycidae).

Project description:Spondylis buprestoides is a major boring pest of Cerambycidae, which mainly damaging conifers and also can carry pine wood nematode, Bursaphelenchus xylophilus. In this study, the complete mitochondrial genome of S. buprestoides was determined by Illumina sequencing technology. The whole mitogenome was 15,837 bp in length with 20.05% GC content, which contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes (rRNAs). Phylogenetic analysis showed that S. buprestoides was closely related to Lepturinae. The sequence data of S. buprestoides could provide useful genetic information for the studies on phylogenetic and evolutionary of Cerambycidae.

The complete mitochondrial genome of Anoplistes halodendri (Coleoptera: Cerambycidae).

Project description:In this study, we report the complete mitochondrial genome of Anoplistes halodendri, which covers a total of 15,697 bp in length with 28.27% GC content. The complete mitochondrial genome is composed of 12 protein-coding genes (PCGs) and also contains 22 transfer RNA genes (tRNAs) and two ribosomal RNA genes (rRNAs). Phylogenetic analysis of the A. halodendri with other 21 different species of Cerambycidae indicated that A. halodendri formed an isolated clade and belong to Cerambycinae. The results will be helpful to study the evolutionary relationship among the subfamilies of Cerambycidae.

Project description:Epicauta ruficeps is widely distributed in China and some countries in Southeast Asia, and plays an important role in medicine and biological control. The complete mitochondria genome of E. ruficeps was 15,813 bp in length, with 37 genes, including 13 PCGs, 22 tRNA genes (tRNAs), and two rRNA genes (rRNAs). The positions and sequences of genes were consistent with those of known Meloidae species. The nucleotide composition was highly A + T biased, accounting for ∼65% of the whole mitogenome. The complete mitogenome of E. ruficeps would help understand Meloidae evolution.

Project description:In this study, the complete mitochondrial genome of Arhopalus unicolor was first sequenced. The length of the complete mitochondria genome of A. unicolor was 15,760 bp with 19.0% GC content, including 40.5% A, 10.8% C, 8.2% G and 40.5% T. There were 13 protein-coding genes (CDS), 22 transfer RNA genes (tRNA), 2 ribosomal RNA genes (rRNA), and an AT-rich region. Phylogenetic analysis showed that A. unicolor was closely related to Spondylis buprestoides. This study provides useful genetic information for subsequent prevention of A. unicolor.

Complete mitochondrial genome of the Themus foveicollis (Coleoptera: Cantharidae).

Project description:The genus Themus Motschulsky, 1858 is one of the largest genera of beetle family Cantharidae, however, no complete mitochondrial genome is available in the public database so far. Here, we sequenced and analyzed the first complete mitochondrial genome of Themus, with T. foveicollis as the representative species. The mitochondrial genome was 16,469 bp in size with 37 genes including 13 protein-coding genes (PCGs), 22 transfer RNA genes, and two ribosomal RNA genes, as well as a non-coding region, which are arranged in the same way as the hypothetical ancestral insect. It exhibited a typical high A + T bias and a higher proportion of bases A and C than T and G (A: 41.1%, T: 37.5%, G: 8.6%, C: 12.9%). The phylogenetic trees of Elateroidea were reconstructed based on 13 PCGs sequences using the Bayesian inference (BI) and maximum-likelihood (ML) analyses to investigate its phylogenetic position. The results showed that Themus was consistently grouped with the remaining genera of Cantharidae and placed in a monophyletic clade of Cantharinae in high supporting values, which are congruent with the morphological classification. The newly sequenced mitochondrial genome in this study constitutes important reference data for reconstructing phylogeny of the family Cantharidae or the superfamily Elateroidea.
