Complete mitochondrial genome of Xiphidiopsis (Xiphidiopsis) gurneyi (Orthoptera, Tettigoniidae, Meconematinae).

ABSTRACT: Xiphidiopsis (Xiphidiopsis) gurneyi belongs to Meconematinae. The complete mitochondrial genome of X. (X.) gurneyi was sequenced by the next-generation sequencing (NGS) technology. The total length of the mitogenome was 16,225?bp and contains the typical gene arrangement, base composition, codon usage found in Meconematinae species. Phylogenetic tree was constructed based on concatenated datasets of PCGs and rRNAs of X. (X.) gurneyi and 19 Tettigoniidae species to assess their phylogenic relationship. Phylogenetic analysis showed that X. (X.) gurneyi was more closely related to the genus of Xizicus.

Project description:The length of Alloxiphidiopsis emarginata complete mitogenome was 16,207 bp and contained the typical gene arrangement, base composition, codon usage found in other related species. The overall base composition exhibited obvious anti-G (10.6%) and AT bias (71.6%). The initiation codons of all PCGs were typical ATN (ATA/ATG/ATT), and the termination codons were TAA, TAG, or incomplete stop codon T. All tRNAs could be folded into typical cloverleaf secondary structures, except tRNASer (AGN). Phylogenetic analyses showed that A. emarginata was closer with Xizicus howardi.

The complete mitochondrial genome of Ruspolia yunnana (Orthoptera: Tettigoniidae: Conocephalinae).

Project description:The complete mitochondrial genome of Ruspolia yunnana was 15,794 bp and consisted of 37 genes and a control region. The A + T content of the mitogenome was as high as 71.3%. Most protein-coding genes started with the codon ATN and ended with stop codons TAA. Phylogenetic analyses based on mitogenomes suggested that Ruspolia yunnana and Ruspolia dubia have the closest genetic relationship. In general, this study provided meaningful genetic information for Ruspolia yunnana and validated the phylogenetic relationships within the Conocephalinae.

Sequencing and analysis of the complete mitochondrial genome of Metrioptera bonneti (Orthoptera: Tettigoniidae).

Project description:Here, we report the complete mitochondrial genome sequence of Metrioptera bonneti and its phylogenetic status. The complete mitogenome of M. bonneti is 16,273 bp in size and consisted of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one D-loop. The overall nucleotide composition of the mitogenome was 35% A, 20.4% C, 11.7% G, and 32.9% T, with an A?+?T bias of 67.9%. Using the 13 PCGs and two rRNA of M. bonneti, together with 39 Tettigoniidae and two outgroup species, we constructed a phylogenetic tree based on the Bayesian inference method. The tree showed that M. bonneti forms a sister clade with Metrioptera ussuriana and supported the monophyly of Tettigoniinae.

The complete mitochondrial genome of Xizicus (Haploxizicus) maculatus revealed by Next-Generation Sequencing and phylogenetic implication (Orthoptera, Meconematinae).

Project description:Xizicus Gorochov, 1993, the quiet-calling katydid, is a diverse genus with 68 species in world, which includes more than 45 species in China, has undergone numerous taxonomic revisions with contradicting conclusions. In this study the complete mitochondrial genome of Xizicus (Haploxizicus) maculatus collected from Hainan for the first time was sequenced using the Next-Generation Sequencing (NGS) technology. The length of whole mitogenome is 16,358 bp and contains the typical gene arrangement, base composition, and codon usage found in other related species. The overall base composition of the mitochondrial genome is 37.0 % A, 32.2 % T, 20.2 % C, and 10.6 % G. All 13 protein-coding genes (PCGs) began with typical ATN initiation codon. Nine of the 13 PCGs have a complete termination codon, but the remaining four genes (COI, COIII, ND5, and ND4) terminate with an incomplete T. Phylogenetic analyses are carried out based on the concatenated dataset of 13 PCGs and two rRNAs of Tettigoniidae species available in GenBank. Both Bayesian inference and Maximum Likelihood analyses recovered each subfamily as a monophyletic group. Regardless of the position of Lipotactinae, the relationships among the subfamilies of Tettigoniidae were as follows: ((((Tettigoniinae, Bradyporinae) Meconematinae) Conocephalinae) Hexacentrinae). The topological structure of the phylogeny trees showed that the Xizicus (Haploxizicus) maculatus is closer to Xizicus (Xizicus) fascipes than Xizicus (Eoxizicus) howardi.

Complete mitochondrial genome of Conocephalus melaenus (Orthoptera: Tettigoniidae): a tRNA-like sequence on the J-strand.

Project description:In this study, the complete mitogenome sequence of Conocephalus melaenus (Orthoptera: Tettigoniidae) has been decoded for the first time by next generation sequencing method. The overall base composition of C. melaenus mitogenome are 37.1% for A, 17.3% for C, 11.1% for G and 34.4% for T, and has high AT content of 71.5%. The complete mitochondrial genome of C. melaenus is 15,923?bp in length, and consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes and one A?+?T-rich region. An unusual feature of the C. melaenus mitogenome is the presence of a tRNASer(UCN) -like sequence on the J-strand, and the tRNASer(UCN) -like sequence have proper folding structures and anticodons sequences. Using the 13 PCGs and 2 rRNA of C. melaenus, together with 17 other close-related and two outgroup species, we constructed Bayesian inference phylogenetic tree to verify the accuracy and utility of new determined mitogenome sequences. The complete mitogenome of C. melaenus provides valuable molecular data for further phylogenetic and evolutionary analysis in Tettigoniidae.

Illumina next-generation sequencing reveals the mitochondrial genome of Ducetia japonica (Orthoptera: Tettigoniidae).

Project description:Using Illumina next-generation sequencing (NGS), the complete mitochondrial genome (mitogenome) of Ducetia japonica was sequenced in the present study. The mitogenome of D. japonica (Genbank accession no. KU885974) is 16,276?bp in size, had the typical invertebrate mitochondrial gene arrangement and containing 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), 2 ribosomal RNA genes (rRNA) and a control region. Except for the control region (868?bp), one novel larger intergenic (616?bp) was found between nad2 and trnW. Phylogenetic results unambiguously support the monophyly of Phaneropterinae, although the gene order of two Sinochlora species different with other Tettigoniidae species. Using Illumina NGS platforms for mitogenome sequencing will provide rather essential and important DNA molecular data for the further phylogenetic analysis across major ensiferan lineages.

Complete Mitochondrial Genome Sequence of Acrida cinerea (Acrididae: Orthoptera) and Comparative Analysis of Mitochondrial Genomes in Orthoptera.

Project description:The complete 15,599-bp mitogenome of Acrida cinerea was determined and compared with that of the other 20 orthopterans. It displays characteristic gene content, genome organization, nucleotide composition, and codon usage found in other Caelifera mitogenomes. Comparison of 21 orthopteran sequences revealed that the tRNAs encoded by the H-strand appear more conserved than those by the L-stand. All tRNAs form the typical clover-leaf structure except trnS (agn), and most of the size variation among tRNAs stemmed from the length variation in the arm and loop of T?C and the loop of DHU. The derived secondary structure models of the rrnS and rrnL from 21 orthoptera species closely resemble those from other insects on CRW except a considerably enlarged loop of helix 1399 of rrnS in Caelifera, which is a potentially autapomorphy of Caelifera. In the A+T-rich region, tandem repeats are not only conserved in the closely related mitogenome but also share some conserved motifs in the same subfamily. A stem-loop structure, 16 bp or longer, is likely to be involved in replication initiation in Caelifera and Grylloidea. A long T-stretch (>17?bp) with conserved stem-loop structure next to rrnS on the H-strand, bounded by a purine at either end, exists in the three species from Tettigoniidae.
