ABSTRACT:

Background

Spexin (SPX) is a peptide hormone that regulates body weight, adipose tissue metabolism, and food intake.

Hypothesis

Serum SPX concentration correlates with body condition score (BCS) and markers of obesity in dogs.

Animals

Fifty-seven dogs of varying body condition assessed using a 5-point BCS.

Methods

Prospective, nonblinded, observational cohort study. Serum SPX concentration was measured using commercially available radioimmunoassay (RIA) in dogs with varying BCS. Spexin mRNA and protein expression were detected using real-time quantitative polymerase chain reaction and immunofluorescence staining.

Results

Serum SPX concentration was lower in dogs with BCS4 (8.56 +/- 2.86) and BCS5 (6.7 +/- 2.12) compared to BCS2 (11.96 +/- 2.23) and BCS3 (10.51 +/- 2.19; BCS2 vs BCS5, P < .001 and BCS2 vs BCS4, P = .005; BCS3 vs BCS5, P = .002). Spexin mRNA was detected in adipose tissue, liver and pancreas. Spexin protein was expressed in adipose tissue and liver but not in pancreas. There were negative correlations between SPX and serum concentration of insulin (P < .05); leptin (P < .01), triglycerides (P < .01), total cholesterol (P < .01), nonesterified fatty acids (P < .01), and fructosamine (P < .01). There was a positive correlation between SPX and serum concentration of adiponectin (P < .01).

Conclusions and clinical importance

Spexin could be involved in pathogenesis of obesity in dogs, and might be considered as a potential marker for obesity.

Project description:BackgroundThe risks during early growth on becoming overweight in adulthood are widely studied in humans. However, early-life predictive factors for canine adult overweight and obesity have not yet been studied. To identify factors that may help explain the development of overweight and obesity at adulthood in dogs, a longitudinal study of 2 years was conducted in 24 female Beagle dogs of the same age, sexual status, and raised under identical environmental conditions. By means of a hierarchical classification on principal components with the following quantitative values: fat-free mass (FFM), percentage fat mass and pelvic circumference at 2 years of age, three groups of dogs were established and were nominally named: ideal weight (IW, n = 9), slightly overweight (OW1, n = 6) and overweight (OW2, n = 9). With the aim of identifying predictive factors of development of obesity at adulthood parental characteristics, growth pattern, energy balance and plasma factors were analysed by logistic regression analysis.ResultsAt 24 months, the group compositions were in line with the body condition scores (BCS 1-9) values of the IW (5 or 6/9), the OW1 (6/9) and the OW2 (7 or 8/9) groups. Logistic regression analysis permitted the identification of neonatal growth rate during the first 2 weeks of life (GR2W) and BCS at 7 months as predictors for the development of obesity at adulthood. Seventy percent of dogs with either GR2W >125% or with BCS > 6/9 at 7 months belonged to the OW2 group. Results from energy intake and expenditure, corrected for FFM, showed that there was a greater positive energy imbalance between 7 and 10 months for the OW2, compared to the IW group.ConclusionThis study expands the understanding of previously reported risk factors for being overweight or obese in dogs, establishing that (i) 15 out of 24 of the studied dogs became overweight and (ii) GR2W and BCS at 7 months of age could be used as predictive factors as overweight adult dogs in the OW2 group had higher values compared the other groups of dogs.

Project description:The relationship between ewe body condition score (BCS) and liveweight (LW) has been exploited previously to predict the former from LW, LW-change, and previous BCS records. It was hypothesized that if fleece weight and conceptus-free liveweight and LW-change, and in addition, height at withers were used, the accuracy of current approaches to predicting BCS would be enhanced. Ewes born in 2017 (n = 429) were followed from 8 mo to approximately 42 mo of age in New Zealand. Individual ewe data were collected on LW and BCS at different stages of the annual production cycle (i.e., prebreeding, at pregnancy diagnosis, prelambing, and weaning). Additionally, individual lambing dates, ewe fleece weight, and height at withers data were collected. Linear regression models were fitted to predict current BCS at each ewe age and stage of the annual production cycle using two LW-based models, namely, unadjusted for conceptus weight and fleece weight (LW alone1) and adjusted (LW alone2) models. Furthermore, another two models based on a combination of LW, LW-change, previous BCS, and height at withers (combined models), namely, unadjusted (combined1) and adjusted for conceptus and fleece weight (combined2), were fitted. Combined models gave more accurate (with lower root mean square error: RMSE) BCS predictions than models based on LW records alone. However, applying adjusted models did not improve BCS prediction accuracy (or reduce RMSE) or improve model goodness of fit (R 2) (P > 0.05). Furthermore, in all models, both LW-alone and combined models, a great proportion of variability in BCS, could not be accounted for (0.25 ≥ R 2 ≥ 0.83) and there was substantial prediction error (0.33 BCS ≥ RMSE ≥ 0.49 BCS) across age groups and stages of the annual production cycle and over time (years). Therefore, using additional ewe data which allowed for the correction of LW for fleece and conceptus weight and using height at withers as an additional predictor did not improve model accuracy. In fact, the findings suggest that adjusting LW data for conceptus and fleece weight offer no additional value to the BCS prediction models based on LW. Therefore, additional research to identify alternative methodologies to account for individual animal variability is still needed.

Project description:Animal robustness may be defined as a complex trait characterizing the ability of an individual to be adapted, productive and healthy under contrasted and fluctuating environmental situations. Such a trait is now considered an essential criterion in order to meet the more ambitious goals of farming sustainability. In ruminants, one of the key mechanisms via which robustness is expressed is the capacity to mobilize or restore body reserves (BR) to cope with the challenges of negative energy balances. The objectives of this work were as follows: 1) to estimate the genetic parameters related to BR dynamics in ewes over successive production cycles and 2) to investigate BR management relationships between different physiological stages. For this, historical individual BW and BCS data from 2,920 phenotyped ewes were used for genetic analysis. The changes in BW (∆BW) and BCS (∆BCS) over time were analyzed. Eight physiological stages were considered to investigate these changes over time: mating, early pregnancy, mid-pregnancy, lambing, early suckling, mid-suckling, weaning, and postweaning. The estimated heritability were low for both ∆BW (h2 = 0.13 to 0.18) and ∆BCS (h2 = 0.04 to 0.16). Moderate to high positive genetic correlations (from 0.48 to 0.91) were obtained between BR mobilization phases and between BR accretion phases. Similarly, moderate to high negative genetic correlations (from -0.36 to -0.75) were estimated between the BR mobilization and accretion periods, suggesting that mechanisms driving BR mobilization and accretion processes were genetically correlated. This is the first study in ruminants that demonstrate that the extent and temporal changes in profiles of BR mobilization and accretion are heritable and genetically linked, indicating that such traits could be considered in genetic programs aimed at improving robustness. Nevertheless, further research is needed for a more comprehensive understanding of BR dynamics, notably by including other physiological parameters (i.e., metabolites and hormones) and additional information on the productive and reproductive life of the ewe.
