Project description:BackgroundThe risks during early growth on becoming overweight in adulthood are widely studied in humans. However, early-life predictive factors for canine adult overweight and obesity have not yet been studied. To identify factors that may help explain the development of overweight and obesity at adulthood in dogs, a longitudinal study of 2 years was conducted in 24 female Beagle dogs of the same age, sexual status, and raised under identical environmental conditions. By means of a hierarchical classification on principal components with the following quantitative values: fat-free mass (FFM), percentage fat mass and pelvic circumference at 2 years of age, three groups of dogs were established and were nominally named: ideal weight (IW, n = 9), slightly overweight (OW1, n = 6) and overweight (OW2, n = 9). With the aim of identifying predictive factors of development of obesity at adulthood parental characteristics, growth pattern, energy balance and plasma factors were analysed by logistic regression analysis.ResultsAt 24 months, the group compositions were in line with the body condition scores (BCS 1-9) values of the IW (5 or 6/9), the OW1 (6/9) and the OW2 (7 or 8/9) groups. Logistic regression analysis permitted the identification of neonatal growth rate during the first 2 weeks of life (GR2W) and BCS at 7 months as predictors for the development of obesity at adulthood. Seventy percent of dogs with either GR2W >125% or with BCS > 6/9 at 7 months belonged to the OW2 group. Results from energy intake and expenditure, corrected for FFM, showed that there was a greater positive energy imbalance between 7 and 10 months for the OW2, compared to the IW group.ConclusionThis study expands the understanding of previously reported risk factors for being overweight or obese in dogs, establishing that (i) 15 out of 24 of the studied dogs became overweight and (ii) GR2W and BCS at 7 months of age could be used as predictive factors as overweight adult dogs in the OW2 group had higher values compared the other groups of dogs.

Project description:The body condition score (BCS) in sheep (Ovis aries) is a widely used subjective measure of body condition. Body condition score and liveweight have been reported to be statistically and often linearly related in ewes. Therefore, it was hypothesized that current BCS could be accurately and indirectly predicted using a ewe's lifetime liveweight, liveweight change, and previous BCS record. Ewes born between 2011 and 2012 (n = 11,798) were followed from 8 months to approximately 67 months of age in New Zealand. Individual ewe data was collected on liveweight and body condition scores at each stage of the annual cycle (pre-breeding, pregnancy diagnosis, pre-lambing, and weaning). Linear regression models were fitted to predict BCS at a given ewe age and stage of the annual cycle using a ewe's lifetime liveweight records (liveweight alone models). Further, linear models were then fitted using previous BCS and changes in liveweight, in addition to the lifetime liveweight records (combined models). Using the combined models improved (p < 0.01) the R2 value by 39.8% (from 0.32 to 0.45) and lowered the average prediction error by 10% to 12% (from 0.29 to 0.26 body condition scores). However, a significant portion of the variability in BCS remained unaccounted for (39% to 89%) even in the combined models. The procedures found in this study, therefore, may overestimate or underestimate measures by 0.23 to 0.32 BCS, which could substantially change the status of the ewe, leading to incorrect management decisions. However, the findings do still suggest that there is potential for predicting ewe BCS from liveweight using linear regression if the key variables affecting the relationship between BCS and liveweight are accounted for.

Project description:Since dogs are part of many peoples' lives, research and industry related to their health and longevity are becoming a rising topic. Although gut microbiota (GM) is a key contributor to host health, limited information is available for canines. Therefore, this study characterized GM according to individual signatures (e.g., breed, age, and body condition score-BCS) of dogs living in the same environment. Fresh fecal samples from 96 healthy dogs were analyzed by sequencing the V3-V4 region of the 16S rRNA gene. The major microbial phyla were Firmicutes, Bacteroidetes, Fusobacteria, Proteobacteria, and Actinobacteria. In the comparison by breeds, relative abundance of Fusobacterium was significantly differed. Interestingly, Fusobacterium perfoetens abundance was positively correlated with age (p = 0.018), being significantly more enriched in the 6-10-year-old group (14.3%) than in the 0.5-1-year-old group (7.2%). Moreover, despite the healthy appearance of dogs in all age (0.5-10 years) and BCS (3-6) groups, the gut microbial environment may be disadvantageous in older dogs or in dogs with an abnormal BCS. These findings broaden our understanding of gut microbial ecology according to individual characteristics of dogs and may be used as a reference for providing customized-care to companion animals.

Project description:The relationship between ewe body condition score (BCS) and liveweight (LW) has been exploited previously to predict the former from LW, LW-change, and previous BCS records. It was hypothesized that if fleece weight and conceptus-free liveweight and LW-change, and in addition, height at withers were used, the accuracy of current approaches to predicting BCS would be enhanced. Ewes born in 2017 (n = 429) were followed from 8 mo to approximately 42 mo of age in New Zealand. Individual ewe data were collected on LW and BCS at different stages of the annual production cycle (i.e., prebreeding, at pregnancy diagnosis, prelambing, and weaning). Additionally, individual lambing dates, ewe fleece weight, and height at withers data were collected. Linear regression models were fitted to predict current BCS at each ewe age and stage of the annual production cycle using two LW-based models, namely, unadjusted for conceptus weight and fleece weight (LW alone1) and adjusted (LW alone2) models. Furthermore, another two models based on a combination of LW, LW-change, previous BCS, and height at withers (combined models), namely, unadjusted (combined1) and adjusted for conceptus and fleece weight (combined2), were fitted. Combined models gave more accurate (with lower root mean square error: RMSE) BCS predictions than models based on LW records alone. However, applying adjusted models did not improve BCS prediction accuracy (or reduce RMSE) or improve model goodness of fit (R 2) (P > 0.05). Furthermore, in all models, both LW-alone and combined models, a great proportion of variability in BCS, could not be accounted for (0.25 ≥ R 2 ≥ 0.83) and there was substantial prediction error (0.33 BCS ≥ RMSE ≥ 0.49 BCS) across age groups and stages of the annual production cycle and over time (years). Therefore, using additional ewe data which allowed for the correction of LW for fleece and conceptus weight and using height at withers as an additional predictor did not improve model accuracy. In fact, the findings suggest that adjusting LW data for conceptus and fleece weight offer no additional value to the BCS prediction models based on LW. Therefore, additional research to identify alternative methodologies to account for individual animal variability is still needed.

Project description:In dogs, optimal growth is critical for future health and wellbeing. Recently, a series of evidence-based growth standards, based on bodyweight, were developed for male and female dogs across 5 different size categories. The aim of the current study was to compare growth curves depicted by the standards with patterns of growth in dogs that were either healthy, had abnormal body condition, or had various diseases with the potential to affect growth. The data came from 2 research colonies in Europe (France and UK), and a large corporate network of primary care veterinary hospitals across the USA. Age and bodyweight data were used to model growth in healthy dogs, in dogs that became overweight or underweight by 3 years of age, and in dogs with diseases associated with altered growth. Centile line crossing during the growth phase was uncommon in healthy dogs, with <5% of dogs crossing >2 centile lines. In contrast, centile line crossing was more frequent in dogs with abnormal growth patterns or abnormal body condition. Dogs that developed obesity by 3 years grew faster than the growth standards predicted, and 68% crossed ≥2 centile lines in an upwards direction. Dogs with conditions associated with accelerated growth also grew faster than expected, and 54% crossed ≥2 centile lines. In contrast dogs that became underweight by 3 years gained weight slower than expected, and 49% crossed ≥2 centile lines in a downwards direction. These results suggest that the growth standards are useful for monitoring healthy growth in dogs. Prospective studies are now required to confirm these findings and to determine whether early intervention can prevent the development of diseases.

Project description:BackgroundThere is an association between overweight status and life span in kenneled dogs, but a similar association has not been reported for pet dogs.ObjectivesTo examine the effects of being overweight in middle age on the life span of neutered client-owned dogs.AnimalsFifty-thousand seven-hundred eighty seven middle-aged neutered client-owned dogs attending a network of approximately 900 veterinary hospitals across North America.MethodsRetrospective case-control study. For each of 12 breeds, groups of dogs aged between 6.5 and 8.5 years were identified as being in "overweight" or "normal" body condition. Within each breed and sex, differences in life span between dogs in normal body condition and overweight body condition in the 2 groups were then analyzed by Cox proportional hazards models.ResultsFor all breeds, instantaneous risk of death for dogs in overweight body condition was greater than those in normal body condition throughout the age range studied, with hazard ratios ranging from 1.35 (99.79% confidence interval [CI] 1.05-1.73) for German Shepherd dog to 2.86 (99.79% CI 2.14-3.83) for Yorkshire Terrier. In all breeds, median life span was shorter in overweight compared with normal weight dogs, with the difference being greatest in Yorkshire Terriers (overweight: 13.7 years, 99.79% CI 13.3-14.2; normal: 16.2 years, 99.79% CI 15.7-16.5) and least in German Shepherd dogs (overweight: 12.1 years, 99.79% CI 11.8-12.4; normal: 12.5 years, 99.79% CI 12.2-12.9).Conclusions and clinical importanceVeterinary professionals should consider promoting healthy body condition for dogs, particularly from midlife onward.

Project description:The assessment of animals' health and nutritional status using a Body Condition Score (BCS) has become a common and reliable tool in lab-animal science. It enables a simple, semi-objective, and non-invasive assessment (palpation of osteal prominences and subcutaneous fat tissue) in routine examination of an animal. In mammals, the BCS classification contains 5 levels: A low score describes a poor nutritional condition (BCS 1-2). A BCS of 3 to 4 is considered optimum, whereas a high score (BCS = 5) is associated with obesity. While BCS are published for most common laboratory mammals, these assessment criteria are not directly applicable to clawed frogs (Xenopus laevis) due to their intracoelomic fat body instead of subcutaneous fat tissue. Therefore, this assessment tool is still missing for Xenopus laevis. The present study aimed to establish a species-specific BCS for clawed frogs in terms of housing refinement in lab-animal facilities. Accordingly, 62 adult female Xenopus laevis were weighed and sized. Further, the body contour was defined, classified, and assigned to BCS groups. A BCS 5 was associated with a mean body weight of 193.3 g (± 27.6 g), whereas a BCS 4 ranged at 163.1 g (±16.0 g). Animals with a BCS = 3 had an average body weight of 114.7 g (±16.7 g). A BCS = 2 was determined in 3 animals (103 g, 110 g, and 111 g). One animal had a BCS = 1 (83 g), equivalent to a humane endpoint. In conclusion, individual examination using the presented visual BCS provides a quick and easy assessment of the nutritional status and overall health of adult female Xenopus laevis. Due to their ectothermic nature and the associated special metabolic situation, it can be assumed that a BCS ≥3 is to be preferred for female Xenopus laevis. In addition, BCS assessment may indicate underlying subclinical health problems that require further diagnostic investigation.

Project description:BackgroundThe kidneys have many essential metabolic functions, and metabolic disturbances during decreased renal function have not been studied extensively.ObjectivesTo identify metabolic changes in blood samples with increased serum creatinine concentration, indicating decreased glomerular filtration.AnimalsClinical samples analyzed using a nuclear magnetic resonance (NMR) based metabolomics platform. The case group consisted of 23 samples with serum creatinine concentration?>125??mol/L, and the control group of 873 samples with serum creatinine concentration within the reference interval.MethodsBiomarker association with increased serum creatinine concentration was evaluated utilizing 3 statistical approaches: Wilcoxon rank-sum test, logistic regression analysis (false discovery rate (FDR)-corrected P-values), and random forest classification. Medians of the biomarkers were compared to reference intervals. A heatmap and box plots were used to represent the differences.ResultsAll 3 statistical approaches identified similar analytes associated with increased serum creatinine concentrations. The percentages of citrate, tyrosine, branched-chain amino acids, valine, leucine, albumin, linoleic acid and the ratio of phenylalanine to tyrosine differed significantly using all statistical approaches, acetate differed using the Wilcoxon test and random forest, docosapentaenoic acid percentage only using logistic regression (P <?.05), and alanine only using random forest.Conclusions and clinical importanceWe identified several metabolic changes associated with increased serum creatinine concentrations, including prospective diagnostic markers and therapeutic targets. Further research is needed to verify the association of these changes with the clinical state of the dog. The NMR metabolomics test is a promising tool for improving diagnostic testing and management of renal diseases in dogs.

Project description:Animal robustness may be defined as a complex trait characterizing the ability of an individual to be adapted, productive and healthy under contrasted and fluctuating environmental situations. Such a trait is now considered an essential criterion in order to meet the more ambitious goals of farming sustainability. In ruminants, one of the key mechanisms via which robustness is expressed is the capacity to mobilize or restore body reserves (BR) to cope with the challenges of negative energy balances. The objectives of this work were as follows: 1) to estimate the genetic parameters related to BR dynamics in ewes over successive production cycles and 2) to investigate BR management relationships between different physiological stages. For this, historical individual BW and BCS data from 2,920 phenotyped ewes were used for genetic analysis. The changes in BW (∆BW) and BCS (∆BCS) over time were analyzed. Eight physiological stages were considered to investigate these changes over time: mating, early pregnancy, mid-pregnancy, lambing, early suckling, mid-suckling, weaning, and postweaning. The estimated heritability were low for both ∆BW (h2 = 0.13 to 0.18) and ∆BCS (h2 = 0.04 to 0.16). Moderate to high positive genetic correlations (from 0.48 to 0.91) were obtained between BR mobilization phases and between BR accretion phases. Similarly, moderate to high negative genetic correlations (from -0.36 to -0.75) were estimated between the BR mobilization and accretion periods, suggesting that mechanisms driving BR mobilization and accretion processes were genetically correlated. This is the first study in ruminants that demonstrate that the extent and temporal changes in profiles of BR mobilization and accretion are heritable and genetically linked, indicating that such traits could be considered in genetic programs aimed at improving robustness. Nevertheless, further research is needed for a more comprehensive understanding of BR dynamics, notably by including other physiological parameters (i.e., metabolites and hormones) and additional information on the productive and reproductive life of the ewe.

Project description:BackgroundThe ACTH stimulation has low sensitivity for the diagnosis of hypercortisolism possibly as a result of biological and analytical variability.Hypothesis/objectivesTo report the components of biological and analytical variability in serum cortisol concentration post-ACTH stimulation ([cortisol]) in healthy dogs.AnimalsFourteen healthy harrier hound dogs.MethodsThe data were extracted from a separate, prospective, randomized, double-blinded, controlled discovery study in which dogs treated with vehicle control and 4 different doses of cortisone acetate (CA) for 7 days had an ACTH stimulation test performed to confirm the dose-dependent effect of CA. The index of individuality (IoI), the critical difference between sequential measurements (CD ), and the number of measurements required to assess the homeostatic set point (HSP) of [cortisol] with confidence intervals (CI) of 90 and 95% were estimated.ResultsThe IoI was equal to 1.1 and the CD was 3.3 ?g/dL (92 nmol/L). The number of measurements required to assess the HSP of [cortisol] with CI of 90 and 95% were 3 and 15, respectively. Additionally, mean [cortisol] was higher in males than in females (13.3 ± 4 ?g/dL [366 ± 114 nmol/L] vs. 11.5 ± 2.5 ?g/dL [318 ± 65 nmol/L], respectively; P = .046). As expected, treatment with CA resulted in a dose-dependent suppression of [cortisol].Conclusions and clinical importanceFalse-negative test results in hypercortisolism could occur when [cortisol] is outside of the individual's HSP and within the reference interval. The large CD emphasizes the importance of assessing clinically relevant parameters in the diagnosis and monitoring of HC.
