Deciphering the chemical instability of sphaeropsidin A under physiological conditions - degradation studies and structural elucidation of the major metabolite.

ABSTRACT: The fungal metabolite sphaeropsidin A (SphA) has been recognised for its promising cytotoxicity, particularly towards apoptosis- and multidrug-resistant cancers. Owing to its intriguing activity, the development of SphA as a potential anticancer agent has been pursued. However, this endeavour is compromised since SphA exhibits poor physicochemical stability under physiological conditions. Herein, SphA's instability in biological media was explored utilizing LC-MS. Notably, the degradation tendency was found to be markedly enhanced in the presence of amino acids in the cell medium utilized. Furthermore, the study investigated the presence of degradation adducts, including the identification, isolation and structural elucidation of a major degradation metabolite, (4R)-4,4',4'-trimethyl-3'-oxo-4-vinyl-4',5',6',7'-tetrahydro-3'H-spiro[cyclohexane-1,1'-isobenzofuran]-2-ene-2-carboxylic acid. Considering the reduced cytotoxic potency of aged SphA solutions, as well as that of the isolated degradation metabolite, the reported antiproliferative activity has been attributed primarily to the parent compound (SphA) and not its degradation species. The fact that SphA continues to exhibit remarkable bioactivity, despite being susceptible to degradation, motivates future research efforts to address the challenges associated with this instability impediment.

Project description:The loss of germination, viability, and vigor of seeds under storage conditions are the main causes of the need to multiply the seed material for leguminous crops. For crop establishment, seeds obtained in propagation fields are usually used, and the coating comes from the basic seed. In the case of leguminous species, the seeds quickly lose their viability, and in accordance with international regulations, for legumes, the number of seeds increases only in the first year of propagation. Therefore, the main objective of this study was to assess the effects of variations in the storage period, temperature and humidity on the morphophysiological and chemical traits of two pea seed varieties (Gloriosa and Kelvedon Wonder). The pea seeds were harvested at the end of June 2017, 2018 and 2019 and stored for 32, 20 and 8 months at T = 4 °C and H = 8%; T = 4 °C and H = 12%; T = 8 °C and H = 8%; T = 8 °C and H = 12%; and T = 22 °C and H = 65%. The results of the morphological, chemical, and biochemical analyses showed that the highest germination rate; hypocotyl length; radicle length; lipid content; dietetic fiber content; caloric value; and Ca, Mg, K, Na, Fe and Zn contents were detected in the Gloriosa and KW seeds stored for 8 months at 4 °C and 8% humidity. Analysis of the experimental data by statistical methods revealed that increasing the storage time had an individual significant negative influence only on the germination rate of both pea varieties and on the hypocotyl length and radicle length of the KW variety, while humidity and temperature variation had individual significant influences on the lipid content. The significant effects of humidity and temperature on the germination rate, hypocotyl length and root length of KW plants were also determined. For the remaining morphophysiological and chemical traits of pea seeds, the individual and combined effects of the factors were not statistically significant. Furthermore, the comparison of means using the Tukey test showed that storage conditions related to temperature and humidity generally used by farmers (T = 22 °C × H = 65%) did not significantly affect the majority of the nutritional properties of the pea seeds. However, maintaining pea seeds under these conditions for a longer period of time significantly affects seed germination and vigor.

Project description:Surface sterilization and hazardous chemical degradation under ambient conditions can provide significant benefits for public and environmental health. Materials with sterilization and chemical degradation capacity under sunlight can efficiently reduce infectious disease incidence rates and toxic chemical exposure. Utilizing renewable energy for sustainable sterilization and degradation is more desirable as it reduces the potential secondary contamination. Herein, we report functional structure design using lignin, a renewable carbon heterogeneous polymer, to synthesize a highly efficient and stable photocatalyst that degrades environmentally hazardous organic compounds rapidly. Through a hydrolysis reaction between Ti-OH and the hydroxyl groups of lignin, Ti-O-C and Ti-O-Ti bonds were established and a lignin based photocatalyst with a hollow sphere structure (Clignin@H-TiO2) was formed. The presence of a homozygous carbon modified TiO2 structure contributes to the enhanced photodegradation activity with solar light. The close hetero-interfacial contact between carbonized lignin and TiO2 further improves the photocatalytic efficiency by facilitating effective charge carrier separation. After synthesis optimization, the resulting Clignin@H-TiO2 photocatalyst exhibits excellent performance in the degradation of atenolol under solar light irradiation with 100% degradation within five minutes. Additionally, it efficiently removes approximately 50% of PFOA and kills about 90% of bacteria within three hours. The uniform distribution of lignin within the crosslinking structures ensures a durable carbon modified TiO2 framework, which remains stable after 10 cycles of usage. The robustness of the lignin-based photocatalyst enables incorporating the catalyst into diversified material formats and various usages. Coating of the photocatalyst onto device surfaces shows bacterial killing efficacy under sunlight. The photocatalysts based on lignin valorization present a green chemistry approach for environmental remediation and surface sterilization, which has long-term environmental protection benefits, with broad applications in toxin treatment and health protection against pathogen infection.
