Complete Genome Sequences of Two Type 3 Porcine Circoviruses, WB17KW and WB20GG, Isolated from Korean Wild Boar.

ABSTRACT: Porcine circovirus type 3 (PCV3) in domestic pigs was first reported in South Korea in 2017. Here, we report the first complete genome sequences of two PCV3 strains isolated from Korean wild boar, which enhance our understanding about the genetic relatedness of PCV3 in domestic pigs and wild boar.

Genetic Diversity of Porcine Circovirus Isolated from Korean Wild Boars.

Project description:In Korea, three genotypes of porcine circovirus type 2 (PCV2a, PCV2b, and PCV2d) have been identified on domestic pig farms, while two genotypes (PCV2a and PCV2b) have been identified in wild boar populations. Here, we investigated genotype diversity and genotypic shift in 91 PCV2 isolates from 1340 wild boars captured in South Korea between 2013 and 2017. Phylogenetic analyses based on the complete ORF2 showed that the 91 PCV2 strains were detected as four genotypes by qPCR screening assay: PCV2a (2.2%, 2/91), PCV2b (16.5%, 15/91), PCV2d (80.2%, 73/91), and PCV2h (1.1%, 1/91). Only one intergenotype recombinant event was detected between PCV2 ORF2 in wild boars (PCV2b) and domestic pigs (PCV2a). Amino acid positions 86-89 within ORF2, which distinguishes the different genotypes, were conserved in all PCV2 genotypes isolated from South Korean wild boars, including TNKI in PCV2a/PCV2h, SNPR in PCV2b, and SNPL in PCV2d. The estimated nucleotide substitution rates in the ORF2 region of viruses from South Korean wild boars and domestic pigs were 5.8145 × 10-4 and 4.5838 × 10-4 substitutions per site per year (s/s/y), respectively. The times to the most recent common ancestor (tMRCA) for South Korean domestic pig PCV2 were 1937 (PCV2a), 1972 (PCV2b), 1999 (PCV2d-1), and 2000 (PCV2d-2). By contrast, the tMRCA for South Korean wild boar PCV2b and PCV2d were 1989 and 2001, respectively. Thus, the PCV2d genotype is prevalent among South Korean wild boars and domestic pigs.

Too many wild boar? Modelling fertility control and culling to reduce wild boar numbers in isolated populations.

Project description:Wild boar and feral swine number and range are increasing worldwide in parallel with their impact on biodiversity and human activities. The ecological and economic impact of this species include spread of diseases, vehicle collisions, damage to crops, amenities and infrastructures and reduction in plant and animal abundance and richness. As traditional methods such as culling have not contained the growth and spread of wild boar and feral pigs, alternative methods such as fertility control are now advocated. We used empirical data on two isolated wild boar populations to model and compare the effects of different regimes of culling and fertility control on population trends. We built a Bayesian population model and applied it to explore the implications for population control of various management options combining culling and/or contraception. The results showed that, whilst fertility control on its own was not sufficient to achieve the target reduction in wild boar number, adding fertility control to culling was more effective than culling alone. In particular, using contraceptives on 40% of the population to complement the culling of 60% of the animals, halved the time to achieve our target reduction compared with culling only. We conclude that, assuming the effort of adding fertility control to culling was found to be cost-effective in terms of population reduction, these two methods should be used simultaneously if a rapid decrease in wild boar number is required for a closed population.

High prevalence of porcine Hokovirus in German wild boar populations.

Project description:Porcine Hokovirus (PHoV) was recently discovered in Hong Kong. This new Parvovirus of pigs is closely related to the human Parvoviruses 4 and 5 (PARV4/5) and bovine Hokovirus (BHoV). So far, nothing is known about the presence and prevalence of PHoV in regions of the world other than Hong Kong. A study was initiated to investigate PHoV in German wild boars from five different geographical regions, using a newly established quantitative real-time PCR assay. Analysis of collected liver and serum samples revealed high overall prevalence (32.7%; 51/156) of PHoV in wild boars. The prevalence differed between the regions and increased with age. Two near full-length genomes and a large fragment for three additional isolates from different regions were sequenced and used for phylogenetic analysis. The German PHoV sequences from wild boars showed a close relationship with sequences of isolates from Hong Kong.

Based on comparative sequence analysis of the F gene, these strains were classified as NDV class II, genotypes VIId and VIb/2, respectively.

Complete genome sequences of a Korean virulent porcine epidemic diarrhea virus and its attenuated counterpart.

Project description:A virulent porcine epidemic diarrhea virus (PEDV) strain, DR13, was obtained from suckling pigs suspected of having porcine epidemic diarrhea in 1999 in Korea, and its attenuated counterpart was derived from virulent strain DR13 by serial propagation in Vero cells. This report describes the first complete genome sequences of virulent PEDV and its attenuated counterpart, which will provide important insights into the molecular basis of the attenuation of PEDV.
