Complete mitochondrial genome of an Asian longicorn beetle, Olenecamptus bilobus Fabricius (Coleoptera: Cerambycidae: Lamiinae).

ABSTRACT: Olenecamptus bilobus Fabricius is widely distributed in some parts of Southeast and East Asia whose larvae bore under the bark of at least eleven plant families. The complete mitochondria genome of O. bilobus was 15,262 bp in length, with 37 genes, including 12 protein-coding genes (PCGs), 23 tRNA genes (tRNAs), and 2 rRNA genes (rRNAs). The A + T content is 76.91%, showing strong AT skew. Phylogenetic analysis indicated that O. bilobus had a close relationship with Olenecamptus subobliteratus Pic.

The nucleotide composition of the D. granulosus mitogenome was A: 39.5%, T: 31.7%, G: 10.9%, C: 17.9%. The A＋T content was 71.2%, showing strong AT skew. Phylogenetic analysis indicated that D. granulosus have sister relationship with Dorysthenes paradoxus.

The complete mitochondrial genomes of five longicorn beetles (Coleoptera: Cerambycidae) and phylogenetic relationships within Cerambycidae.

Project description:Cerambycidae is one of the most diversified groups within Coleoptera and includes nearly 35,000 known species. The relationships at the subfamily level within Cerambycidae have not been convincingly demonstrated and the gene rearrangement of mitochondrial genomes in Cerambycidae remains unclear due to the low numbers of sequenced mitogenomes. In the present study, we determined five complete mitogenomes of Cerambycidae and investigated the phylogenetic relationship among the subfamilies of Cerambycidae based on mitogenomes. The mitogenomic arrangement of all five species was identical to the ancestral Cerambycidae type without gene rearrangement. Remarkably, however, two large intergenic spacers were detected in the mitogenome of Pterolophia sp. ZJY-2019. The origins of these intergenic spacers could be explained by the slipped-strand mispairing and duplication/random loss models. A conserved motif was found between trnS2 and nad1 gene, which was proposed to be a binding site of a transcription termination peptide. Also, tandem repeat units were identified in the A + T-rich region of all five mitogenomes. The monophyly of Lamiinae and Prioninae was strongly supported by both MrBayes and RAxML analyses based on nucleotide datasets, whereas the Cerambycinae and Lepturinae were recovered as non-monophyletic.

Complete mitochondrial genome of Pseudoechthistatus hei (Coleoptera: Cerambycidae: Lamiinae) and its phylogenetic analysis.

Project description:The little-known genus Pseudoechthistatus Pic, 1917 belongs to the subfamily Lamiinae of the family Cerambycidae. Adult Pseudoechthistatus hei Xie and W. Wang, 2019 specimens were collected from Bāijì Hill, Xīntángfáng Village, Wéixī County, Yúnnán Province, China. The complete mitochondrial genome (GenBank accession number: ON641973.1) of P. hei was sequenced, annotated, and characterized; it is a circular DNA molecule of 16,103 bp with a 75.71% AT content, and it comprised 13 protein-coding genes (PCG), 22 tRNA genes, two rRNA genes, and 1 control region. The PCGs initiated with the typical ATN (Met) start codons, and were terminated by typical TAN stop codons. The Bayesian Inference phylogenetic tree was first constructed using JTT + F + I + G4 model for P. hei, which showed that P. hei was closely related to Monochamus alternatus alternatus.

Hispaniolan hemilophini (coleoptera, cerambycidae, lamiinae).

Project description:The Tribe Hemilophini (Lamiinae) is reviewed for Hispaniola and an identification key is provided. Fifteen species are now known from the island, including one new species of Adesmus (Adesmus fortunei from Pedernales and La Vega Provinces, Dominican Republic), one new species of Oedudes (Oedudes anulatus from Peravia and La Vega Provinces, Dominican Republic), and five new species of Calocosmus (Calocosmus contortus from San Cristóbal Province, Calocosmus punctatus from Peravia Province, Calocosmus rawlinsi from Elías Piña Province, Calocosmus robustus from La Vega Province, and Calocosmus thonalmus from La Altagracia Province, all in the Dominican Republic). Oedudes and Adesmus are new island and country records for Hispaniola and Dominican Republic, respectively. Calocosmus holosericeus Gahan is a new synonym of Calocosmus janus Bates. In addition to the new species, five new country records and four new island records are presented for Calocosmus.

The Phylogenetic Relationship of Lamiinae (Coleoptera: Cerambycidae) Using Mitochondrial Genomes.

Project description:Lamiinae is the largest subfamily of the Cerambycidae (longhorn beetles), with approximately 21,863 described species. Previous phylogenetic studies of Lamiinae showed that this subfamily was monophyletic, but the relationship between the tribes of Lamiinae is still controversial. Partial molecular data and species morphological characteristics are not sufficient to resolve species phylogenetic studies perfectly. At the same time, the full mitochondrial genome contains more comprehensive genetic data. Benefiting from the development of next-generation sequencing (NGS), mitochondrial genomes can be easily acquired and used as reliable molecular markers to investigate phylogenetic relationships within Cerambycidae. Using NGS technology, we obtained 11 mitochondrial genome sequences of Lamiinae species. Based on this newly generated mitochondrial genome dataset matrix, we reconstructed the phylogeny of Lamiinae. The Bayesian Inference and Maximum Likelihood analyses strongly support the monophyly of four tribes (Lamiini, Batocerini, Mesosini, and Saperdini), whereas the tribe Acanthocinini was identified as paraphyletic. Other mitochondrial structural features were also observed: the start codon in the nad1 gene of all 11 mitochondrial genomes is TTG; 17-22 bp intergenic spacers (IGS) with a 'TACTA' motif were found between trnS2 and nad1. Moreover, two long IGS were found in Mesosa myops and Batocera sp. Tandem repeats were found in the IGS of Batocera sp.

Cytoplasmic incompatibility in the semivoltine longicorn beetle Acalolepta fraudatrix (Coleoptera: Cerambycidae) double infected with Wolbachia.

Project description:Wolbachia are obligatory endosymbiotic α-proteobacteria found in many arthropods. They are maternally inherited, and can induce reproductive alterations in the hosts. Despite considerable recent progress in studies on the associations between Wolbachia and various taxonomic groups of insects, none of the researches have revealed the effects of Wolbachia on longicorn beetles as the host insect. Acalolepta fraudatrix is a forest longicorn beetle that is distributed in East Asia. In this study, the relationship between Wolbachia and A. fraudatrix was investigated. Out of two populations of A. fraudatrix screened for Wolbachia using the genes ftsZ, wsp, and 16S rRNA, only one of the populations showed detection of all three genes indicating the presence of Wolbachia. Electron microscopy and fluorescent in situ hybridization also confirmed that the A. fraudatrix population was infected with Wolbachia. Sequencing the wsp genes derived from single insects revealed that two strains of Wolbachia coexisted in the insects based on the detection of two different sequences of the wsp gene. We designated these strains as wFra1 and wFra2. The bacterial titers of wFra1 were nearly 2-fold and 3-fold higher than wFra2 in the testes and ovaries, respectively. The two strains of Wolbachia in the insects were completely eliminated by rearing the insects on artificial diets containing 1% concentration of tetracycline for 1 generation. Reciprocal crosses between Wolbachia-infected and Wolbachia-uninfected A. fraudatrix demonstrated that only eggs produced by the crosses between Wolbachia-infected males and Wolbachia-uninfected females did not hatch, indicating that Wolbachia infecting A. fraudatrix causes cytoplasmic incompatibility in the host insect. This is the first report showing the effect of Wolbachia on reproductive function in a longicorn beetle, A. fraudatrix.

Complete mitochondrial genome of Oberea diversipes (Coleoptera, Cerambycidae).

Project description:The complete mitochondrial DNA genome of the Oberea diversipes was reported. The genome was a circular molecule of 15,499 base pairs (bp), with 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an estimated 830 bp A + T-rich control region. The total base composition of the mitogenome was A-T bias, with 40.9% for A, 37.0% for T, 13.4% for C, and 8.7% for G. The molecular data presented here would be useful for further study of O. diversipes.
