Complete mitochondrial genome of Penthimia melanocephala Motschulsky, 1863 (Hemiptera: Cicadellidae: Deltocephalinae).

ABSTRACT: Penthimia is the largest genus in the tribe Penthimiini of the subfamily Deltocephalinae. To date, there are no available mitogenome sequences from Penthimia. In this study, we have sequenced and annotated the complete mitogenome of Penthimia melanocephala (Motschulsky 1863), the mitogenome is 15,308 bp in length, which including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), 22 transfer RNAs (tRNAs) and a long non-coding region (control Region), base composition of whole sequences are A (50.4%), C (15.3%), G (10.0%), and T (24.3%). All PCGs started with the typical ATN codon and stopped with the typical TAN codon except for COII and COIII stopped with single T. Within phylogenetic tree, Deltocephalinae members were clustered into a clade. The complete mitogenome of P. melanocephala can provide essential DNA molecular data for further evolutionary and phylogenetic analysis.

Project description:The complete mitochondrial genome of Fieberiella septentrionalis was annotated for the first time in the present study. The mitogenome was found to have circular shape, with 16,175 bp in size, containing 13 protein coding genes (PCGs), 22 transfer-RNA genes, 2 ribosomal-RNA genes, and 1 non-coding region. The nucleotide composition biases toward A and T is 77.9% of the entirety, which is a typical structure of Cicadellidae. All PCGs have ATN as the start codon, TAA and single T as the stop codon. The resulting phylogenetic tree confirms that the F. septentrionalis belongs to the subfamily of Deltocephalinae and Fieberiellini as sister to the remaining tribes of this subfamily.

The complete mitochondrial genome of Aconurella prolixa (Lethierry 1885) (Hemiptera: Cicadellidae: Deltocephalinae: Chiasmini).

Project description:The complete mitochondrial genome of the widespread leafhopper species Aconurella prolixa (Hemiptera: Cicadellidae: Deltocephalinae: Chiasmini) was obtained via next-generation sequencing. This mitochondrial genome is 14,832 bp in length with the 37 classical eukaryotic mitochondrial genes and a control region. All 13 protein-coding genes (PCGs) are initiated with ATN, except ND5 uses TTG as the start codon, and terminate with TAA or TAG with the exception of COX2 and ND4 which use a single T residue as the stop codon. Twenty-one of the 22 transfer RNA (tRNAs) genes have the typical clover-leaf structure except for trnS1. Unlike some other species of deltocephalinae, no tRNA rearrangements were detected. The monophyly of Cicadellidae and Deltocephalinae, as well as the monophyly of Chiasmini, with a sister relationship between A. prolixa and (Exitianus indicus + Nephotettix cincticeps) is supported by Bayesian inference phylogenetic analyses based on 13 PCGs.

Complete mitochondrial genome of Mukaria splendida Distant (Hemiptera: Cicadellidae: Deltocephalinae: Mukariini) and phylogenetic analysis.

Project description:The complete mitochondrial genome (mitogenome) of Mukaria splendida Distant, 1908 (Hemiptera: Cicadellidae: Deltocephalinae) was first reported in Mukariini. The length of this mitogenome is 16,711 bp, which has an A + T content of 79% (A = 44.5%, T = 34.5%, G = 8.8%, C = 12.2%). A total of 37 genes were annotated [13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), and 2 ribosomal RNA genes (rRNA)]. Among the 37 genes, 4 protein coding genes (ND1, ND4, ND4L, ND5), 8 tRNA genes (trnQ, trnC, trnY, trnF, trnH, trnP, trnL2, trnV), and 2 rRNA (12S rRNA, 16S rRNA) were encoded by N chain, and the remaining genes were encoded by J chain. Overall, there were 14 gene overlaps and 9 gene gaps in the mitochondrial genome of this species. All PCGs were started with ATD (ATA/ATT/ATG), and stopped with TAR, except ATP6, which ends with single T. The phylogenetic analysis confirms that M. splendida clustered with other Deltocephalinae species.

Characterization of the Complete Mitochondrial Genome of Drabescus ineffectus and Roxasellana stellata (Hemiptera: Cicadellidae: Deltocephalinae: Drabescini) and Their Phylogenetic Implications.

Project description:To explore the mitogenome characteristics and shed light on the phylogenetic relationships and molecular evolution of Drabescini species, we sequenced and analyzed the complete mitochondrial genome of two species including Drabescus ineffectus and Roxasellana stellata. The complete mitogenomes of D. ineffectus and R. stellata are circular, closed and double-stranded molecules with a total length of 15744 bp and 15361 bp, respectively. These two newly sequenced mitogenomes contain the typical 37 genes. Most protein-coding genes (PCGs) began with the start codon ATN and terminated with the terminal codon TAA or TAG, with an exception of a special initiation codon of ND5, which started with TTG, and an incomplete stop codon T-- was found in the Cytb, COX2, ND1 and ND4. All tRNAs could be folded into the canonical cloverleaf secondary structure except for the trnS1, which lacks the DHU arm and is replaced by a simple loop. The multiple tandem repeat units were found in A + T-control region. The sliding window, Ka/Ks and genetic distance analyses indicated that the ATP8 presents a high variability and fast evolutionary rate compared to other PCGs. Phylogenetic analyses based on three different datasets (PCG123, PCG12R and AA) using both Bayesian inference (BI) and maximum likelihood (ML) methods showed strong support for the monophyly of Drabescini.

Project description:The complete mitogenome of Idioscopus nitidulus (Cicadellidae) was sequenced. It comprises 15,287 base pairs (bp), including 13 protein-coding genes (PCGs), 2 rRNA genes, 22 tRNA genes and a control region. The phylogenetic analyses based on concatenated thirteen protein-coding genes of mitogenomes recover the monophyly of Auchenorrhyncha (Fulgoromorpha + Cicadomorpha) and Sternorrhyncha as a sister group to Auchenorrhyncha. The complete mitogenome sequence of Idioscopus nitidulusis available in the GenBank with accession number: KR024406.

Chromosome-Level Genome Assembly of Nephotettix cincticeps (Uhler, 1896) (Hemiptera: Cicadellidae: Deltocephalinae).

Project description:The green rice leafhopper, Nephotettix cincticeps (Uhler), is an important rice pest and a vector of the rice dwarf virus in Asia. Here, we produced a high-quality chromosome-level genome assembly of 753.23 Mb using PacBio (∼110×) and Hi-C data (∼94×). It contained 163 scaffolds and 950 contigs, whose scaffold/contig N50 lengths reached 85.36/2.57 Mb. And 731.19 Mb (97.07%) of the assembly was anchored into eight pseudochromosomes. Genome completeness was attained to 97.0% according to the insect reference Benchmarking Universal Single-Copy Orthologs (BUSCO) gene set (n = 1,367). We masked 347.10 Mb (46.08%) of the genome as repetitive elements. Nine hundred sixty-two noncoding RNAs were identified and 14,337 protein-coding genes were predicted. We also assigned GO term and KEGG pathway annotations for 10,049 and 9,251 genes, respectively. Significantly expanded gene families were primarily involved in immunity, cuticle, digestion, detoxification, and embryonic development. This study provided a crucial genomic resource for better understanding on the biology and evolution in family Cicadellidae.

Project description:The complete mitochondrial genome of the Aster leafhopper Macrosteles quadrilineatus was sequenced using an Illumina-based next-generation sequencing approach. The genome is 16,626 bp in length with 78.0% AT content. It encodes 37 typical mitochondrial genes including 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 A + T-rich region. Two tandem repeats were identified within the A + T-rich region. One tRNA gene rearrangement (trnW-trnC-trnY→trnC-trnW-trnY) was found between nd2 and cox1.

Complete mitochondrial genome of Populicerus confusus (Hemiptera: Cicadellidae: Idiocerinae).

Project description:In this study, we firstly reported the complete mitochondrial genome of Populicerus confuses. The complete mitochondrial genome was 16,395 bp in length which overall base composition was 41.43% A, 36.30% T, 11.54% C, and 10.73% G. It consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes (12S and 16S rRNA), and a control region (D-loop region). The complete mitochondrial genomes of P. confuses and other 9 species were used for phylogenetic analysis using the Bayesian method. The resulting phylogenetic tree confirms that the Populicerus populi is most closely related to P. confuses. The mitogenome provided the valuable evidence on phylogenetic relationship of the Idiocerinae at the molecular level.

Complete mitochondrial genome sequence of Empoascanara dwalata (Hemiptera: Cicadellidae: Typhlocybinae).

Project description:In this study, the complete mitochondrial genome of the leafhopper species Empoascanara dwalata (Hemiptera: Cicadellidae: Typhlocybinae) was sequenced and annotated, and the sequence is available in the GenBank with accession number: MT350235. The circular mitogenome with 15,271 bp in length, including 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one AT-rich region. The nucleotide composition is as follows: A (41.7%), T (34.7%), G (10.6%), and C (13.0%). 11 PCGs have ATN as the start codon, except for atp8 and nad5 genes have TTG. The conventional termination codons (TAA or TAG) occur in 11 PCGs, while cox2 and nad5 use incomplete codon (T--) as termination codon. Phylogenetic analysis using 13 PCGs showed that E. dwalata was clustered with three other Typhlocybine species, which was consistent with the conventional classification.
