Exploitation of Tolerance of Wheat Kernel Weight and Shape-Related Traits from Aegilops tauschii under Heat and Combined Heat-Drought Stresses.

ABSTRACT: Kernel weight and shape-related traits are inherited stably and increase wheat yield. Narrow genetic diversity limits the progress of wheat breeding. Here, we evaluated kernel weight and shape-related traits and applied genome-wide association analysis to a panel of wheat multiple synthetic derivative (MSD) lines. The MSD lines harbored genomic fragments from Aegilops tauschii. These materials were grown under optimum conditions in Japan, as well as under heat and combined heat-drought conditions in Sudan. We aimed to explore useful QTLs for kernel weight and shape-related traits under stress conditions. These can be useful for enhancing yield under stress conditions. MSD lines possessed remarkable genetic variation for all traits under all conditions, and some lines showed better performance than the background parent Norin 61. We identified 82 marker trait associations (MTAs) under the three conditions; most of them originated from the D genome. All of the favorable alleles originated from Ae. tauschii. For the first time, we identified markers on chromosome 5D associated with a candidate gene encoding a RING-type E3 ubiquitin-protein ligase and expected to have a role in regulating wheat seed size. Our study provides important knowledge for the improvement of wheat yield under optimum and stress conditions. The results emphasize the importance of Ae. tauschii as a gene reservoir for wheat breeding.

Project description:The inconsistent prevalence of abiotic stress in most of the agroecosystems can be addressed through deployment of plant material with stress adaptive plasticity. The present study explores water stress induced plasticity for early root-shoot development, proline induction and cell membrane injury in 57 accessions of Aegilops tauschii (DD-genome) and 26 accessions of Triticum dicoccoides (AABB-genome) along with durum and bread wheat cultivars. Thirty three Ae. tauschii accessions and 18 T. dicoccoides accessions showed an increase in root dry weight (ranging from 1.8 to 294.75%) under water stress. Shoot parameters- length and biomass, by and large were suppressed by water stress, but genotypes with stress adaptive plasticity leading to improvement of shoot traits (e.g., Ae tauschii accession 14191 and T. dicoccoides accession 7130) could be identified. Water stress induced active responses, rather than passive repartitioning of biomass was indicated by better shoot growth in seedlings of genotypes with enhanced root growth under stress. Membrane injury seemed to work as a trigger to activate water stress adaptive cellular machinery and was found positively correlated with several root-shoot based adaptive responses in seedlings. Stress induced proline accumulation in leaf tissue showed marked inter- and intra-specific genetic variation but hardly any association with stress adaptive plasticity. Genotypic variation for early stage plasticity traits viz., change in root dry weight, shoot length, shoot fresh weight, shoot dry weight and membrane injury positively correlated with grain weight based stress tolerance index (r = 0.267, r = 0.404, r = 0.299, r = 0.526, and r = 0.359, respectively). In another such trend, adaptive seedling plasticity correlated positively with resistance to early flowering under stress (r = 0.372 with membrane injury, r = 0.286 with change in root length, r = 0.352 with change in shoot length, r = 0.268 with change in shoot dry weight). Overall, Ae. tauschii accessions 9816, 14109, 14128, and T. dicoccoides accessions 5259 and 7130 were identified as potential donors of stress adaptive plasticity. The prospect of the study for molecular marker tagging, cloning of plasticity genes and creation of elite synthetic hexaploid donors is discussed.

Project description:As the post-transcriptional regulators of gene expression, microRNAs or miRNAs comprise an integral part of understanding how genomes function. Although miRNAs have been a major focus of recent efforts, miRNA research is still in its infancy in most plant species. Aegilops tauschii, the D genome progenitor of bread wheat, is a wild diploid grass exhibiting remarkable population diversity. Due to the direct ancestry and the diverse gene pool, A. tauschii is a promising source for bread wheat improvement. In this study, a total of 87 Aegilops miRNA families, including 51 previously unknown, were computationally identified both at the subgenomic level, using flow-sorted A. tauschii 5D chromosome, and at the whole genome level. Predictions at the genomic and subgenomic levels suggested A. tauschii 5D chromosome as rich in pre-miRNAs that are highly associated with Class II DNA transposons. In order to gain insights into miRNA evolution, putative 5D chromosome miRNAs were compared to its modern ortholog, Triticum aestivum 5D chromosome, revealing that 48 of the 58 A. tauschii 5D miRNAs were conserved in orthologous T. aestivum 5D chromosome. The expression profiles of selected miRNAs (miR167, miR5205, miR5175, miR5523) provided the first experimental evidence for miR5175, miR5205 and miR5523, and revealed differential expressional changes in response to drought in different genetic backgrounds for miR167 and miR5175. Interestingly, while miR5523 coding regions were present and expressed as pre-miR5523 in both T. aestivum and A. tauschii, the expression of mature miR5523 was observed only in A. tauschii under normal conditions, pointing out to an interference at the downstream processing of pre-miR5523 in T. aestivum. Overall, this study expands our knowledge on the miRNA catalog of A. tauschii, locating a subset specifically to the 5D chromosome, with ample functional and comparative insight which should contribute to and complement efforts to develop drought tolerant wheat varieties.
