The complete mitochondrial genome of Calliptamus barbarus Costa 1836 (Orthoptera: Acrididae) from Qinghai Lake, China and its phylogeny.

ABSTRACT: The complete mitochondrial genome of Calliptamus barbarus (Orthoptera: Acrididae: Calliptaminae) from Qinghai Lake, Qinghai province, China is a circular molecule of 15,668 bp in size, and contains 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and one AT-rich region. The overall nucleotide composition is 41.8% of A, 30.9% of T, 11.3% of G, and 16.0% of C. All PCGs started with typical ATN codon, e.g. one with ATA, two with ATT and ATC, and eight with ATG. Eleven PCGs ended with complete stop codon TAA, and the other two genes (ND1 and ND4L) ended with TAG. Phylogenetic trees were reconstructed with 13 PCGs using Bayesian Inference (BI) and maximum likelihood (ML) to validate the taxonomic status of C. barbarus, exhibiting the close relationships with C. abbreviates + C. italicus.

Molecular Features and Expression Patterns of Vitellogenin Receptor in Calliptamus italicus (Orthoptera: Acrididae).

Project description:Vitellogenin receptor (VgR) mediates the intake of vitellin via oocytes, thus exerting an important role in vitellogenesis. In this study, reverse transcription-polymerase chain reaction (RT-PCR) and rapid-amplification of cDNA ends techniques were adopted to clone the CiVgR gene, namely the VgR gene of Calliptamus italicus, i.e., Orthopteran. The full length of CiVgR was 5,589 bp, and the open reading frame was estimated to be 5,265 bp, which encoded 1,754 amino acids (aa). Sequence alignment analysis showed that CiVgR belonged to the superfamily of low-density lipoprotein receptor genes, which contained several conserved domains, including ligand-binding domains, epidermal growth factor precursor homology domains, transmembrane domains, and cytoplasmic domains. However, no O-linked sugar domain was identified. Phylogenetic analysis showed that CiVgR had the closest genetic relationship to Blattarias. RT-PCR showed that CiVgR was only specifically expressed in the ovarian tissue of females. quantitative real time polymerase chain reaction showed that the transcription of CiVgR already appeared in the fourth-instar nymph of C. italicus, which gradually increased after adult emergence, peaked at the previtellogenesis stage, and then started to decrease. The expression pattern of CiVgR was closely associated with vitellogenesis. The findings of this study further our understanding of the molecular mechanisms involved in the reproduction of C. italicus, and provide new ideas to control this insect.

Preliminary Study of the Intestinal Microbial Diversity of Three Acridoidae: Oedipoda fuscocincta, Dociostaurus moroccanus, and Calliptamus barbarus (Acrididae: Orthoptera), in the Moroccan Middle Atlas.

Project description:Locusts are known for their herbivorous diet that constitutes a nuisance to agriculture worldwide, in Morocco these insects are considered a real threat and are widely distributed in the country. These insects are equipped with a digestive system that allows them to digest huge amounts of plant tissue. To understand the mechanisms allowing this voracity, the current study has focused on the diversity of gut microbiome using biochemical and molecular analysis tools, different bacterial isolates were identified and studied. The present study results showed the presence of four important bacterial families that are present in the intestine of these insects, namely Micrococcaceae, Dermabacteraceae, Bacillaceae, and Pseudomonadaceae. The results of Gram staining showed that 2 of 11 isolates were Gram-negative bacteria, however, only 9 bacterial strains were catalase positive. While, 3 strains (Pseudomonas stutzeri S12, Kocuria rhizophila, and Bacillus thuringiensis S4 and S8) had pectinase activity, while only one strain (Pseudomonas stutzeri S12) had cellulase activity.

The Survival and Physiological Response of Calliptamus abbreviatus Ikonn (Orthoptera: Acrididae) to Flavonoids Rutin and Quercetin.

Project description:Insect-resistant substances from plants are important natural resources that human beings can potentially develop and use to control pests. In this study, we explored the adverse effects of rutin and quercetin on grasshopper (Calliptamus abbreviatus), as well as the insect's physiological response to these substances in laboratory and field experiments. These two plant compounds exhibited toxic effects on C. abbreviatus, with quercetin showing a stronger toxicity, indicated by a lower survival, slower development, and higher induced gene expression and activities of UDP-glucuronosyltransferase, cytochrome P450s, superoxide dismutase, peroxidase and catalase, compared to rutin. These compounds, especially quercetin, have the potential to be developed as biopesticides to control grasshoppers.

The complete mitochondrial genome of Calliptamus abbreviatus Ikovnnikov (Orthoptera: acridoidea).

Project description:Calliptamus abbreviatus Ikovnnikov (Orthoptera: Acridoidea) is one of the important pests in the grasslands in northern China. The complete mitochondrial genome of this insect was sequenced. This genome is 16,615 bp long, with an AT content of 73.3%, containing 37 typical animal mitochondrial genes and an AT-rich region. All 13 PCGs share the start codon ATN, and the usual termination codons (TAA) are found from 13 protein-coding genes. All of the 22 typical animal tRNA genes were found in C. abbreviatus mt-genome, and most of the tRNAs could be folded into the classic cloverleaf secondary structure except for tRNA-Ser (AGN), which lacks the dihydrouracil (DHU) stem. The sizes of the large and small ribosomal RNA genes are 1555 and 799 bp long, respectively. The AT content of the AT-rich region is 87.0%. Phylogenetic analysis supports that the coleopteran insects from the same family cluster in the same group.

Selection of Reference Genes for Normalization of Real-Time PCR Data in Calliptamus italicus (Orthoptera: Acrididae) Under Different Temperature Conditions.

Project description:Global warming has dominated worldwide climate change trends, and adaptability to high temperatures is the main factor underlying the spread of the pest Calliptamus italicus in Xinjiang Province, China. However, knowledge about the molecular mechanisms responsible for this adaptability and other related biological properties of C. italicus remain relatively unclear. Real-time quantitative polymerase chain reaction (RT-qPCR) is a key tool for gene expression analysis associated with various biological processes. Reference genes are necessary for normalizing gene expression levels across samples taken from specific experimental conditions. In this study, transcript level of five genes (GAPDH, 18S, TUB, ACT, and EF1α), commonly used as reference genes, were evaluated under nine different temperatures (27, 30, 33, 36, 39, 42, 45, 48, and 51°C) to assess their expression stability and further select the most suitable to be used on normalization of target gene expression data. Gene expression profiles were analyzed using geNorm, NormFinder, and BestKeeper software packages. The combined results demonstrated that the best-ranked reference genes for C. italicus are EF1α, GAPDH, and ACT under different thermal stress conditions. This is the first study that assesses gene expression analysis across a range of temperatures to select the most appropriate reference genes for RT-qPCR data normalization in C. italicus. These results should assist target gene expression analysis associated with heat stress in C. italicus.

Using the 13 PCGs and two rRNA of P. infumata, together with 10 other close-related and two outgroup species, we constructed Bayesian inference (BI) phylogenetic tree to validate the mitogenome sequences of P. infumata.

Molecular phylogeny and species delimitation of the genus Tonkinacris (Orthoptera, Acrididae, Melanoplinae) from China.

Project description:Tonkinacris is a small group in Acrididae. While a few species were occasionally sampled in some previous molecular studies, there is no revisionary research devoted to the genus. In this study, we explored the phylogeny of and the relationships among Chinese species of the genus Tonkinacris using the mitochondrial COI barcode and the complete sequences of ITS1 and ITS2 of the nuclear ribosomal DNA. The phylogeny was reconstructed in maximum likelihood and Bayesian inference frameworks, respectively. The overlap range between intraspecific variation and interspecific divergence was assessed via K2P distances. Species boundaries were delimitated using phylogenetic species concept, NJ tree, K2P distance, the statistical parsimony network as well as the GMYC model. The results demonstrate that the Chinese Tonkinacris species is a monophyletic group and the phylogenetic relationship among them is (T. sinensis, (T. meridionalis, (T. decoratus, T. damingshanus))). While T. sinensis, T. meridionalis and T. decoratus were confirmed being good independent species strongly supported by both morphological and molecular evidences, the validity of T. damingshanus was not perfectly supported by molecular evidence in this study.
