Project description:Due to their surface characteristics, nanosized titanium dioxide particles (nTiO2) tend to adhere to biological surfaces and we thus hypothesize that they may alter the swimming performance and behavior of motile aquatic organisms. However, no suitable approaches to address these impairments in swimming behavior as a result of nanoparticle exposure are available. Water fleas Daphnia magna exposed to 5 and 20 mg/L nTiO2 (61 nm; polydispersity index: 0.157 in 17.46 mg/L stock suspension) for 96 h showed a significantly (p<0.05) reduced growth rate compared to a 1-mg/L treatment and the control. Using three-dimensional video observations of swimming trajectories, we observed a treatment-dependent swarming of D. magna in the center of the test vessels during the initial phase of the exposure period. Ensemble mean swimming velocities increased with increasing body length of D. magna, but were significantly reduced in comparison to the control in all treatments after 96 h of exposure. Spectral analysis of swimming velocities revealed that high-frequency variance, which we consider as a measure of swimming activity, was significantly reduced in the 5- and 20-mg/L treatments. The results highlight the potential of detailed swimming analysis of D. magna for the evaluation of sub-lethal mechanical stress mechanisms resulting from biological surface coating and thus for evaluating the effects of nanoparticles in the aquatic environment.

Project description:Ecosystem connectivity is an essential consideration for marine spatial planning of competing interests in the deep sea. Immobile, adult communities are connected through freely floating larvae, depending on new recruits for their health and to adapt to external pressures. We hypothesize that the vertical swimming ability of deep-sea larvae, before they permanently settle at the bottom, is one way larvae can control dispersal. We test this hypothesis with more than [Formula: see text] simulated particles with a range of active swimming behaviours embedded within the currents of a high-resolution ocean model. Despite much stronger horizontal ocean currents, vertical swimming of simulated larvae can have an order of magnitude impact on dispersal. These strong relationships between larval dispersal, pathways, and active swimming demonstrate that lack of data on larval behaviour traits is a serious impediment to modelling deep-sea ecosystem connectivity; this uncertainty greatly limits our ability to develop ecologically coherent marine protected area networks.

Project description:Larval dispersal is a key process determining population connectivity, metapopulation dynamics, and community structure in benthic marine ecosystems, yet the biophysical complexity of dispersal is not well understood. In this study, we investigate the interaction between disperser phenotype and hydrodynamics on larval dispersal pathways, using a temperate reef fish species, Trachinops caudimaculatus. We assessed the influence of larval traits on depth distribution and dispersal outcomes by: (i) using 24-h depth-stratified ichthyoplankton sampling, (ii) quantifying individual phenotypes using larval growth histories extracted from the sagittal otoliths of individual larvae, and (iii) simulating potential dispersal outcomes based on the empirical distribution of larval phenotypes and an advanced biological-physical ocean model. We found T. caudimaculatus larvae were vertically stratified with respect to phenotype, with high-quality phenotypes found in the bottom two depth strata, and poor-quality phenotypes found primarily at the surface. Our model showed high- and average-quality larvae experienced significantly higher local retention (more than double) and self-recruitment, and travelled shorter distances relative to poor-quality larvae. As populations are only connected when dispersers survive long enough to reproduce, determining how larval phenotype influences dispersal outcomes will be important for improving our understanding of marine population connectivity and persistence.

Project description:Simple Summary New types of pesticides are increasingly found in surface waters around the world. Little is known about how they interact in mixtures, and how these mixtures might then affect aquatic organisms at risk of exposure. We looked at how two new pesticides, chlorantraniliprole (CHL) and imidacloprid (IMI), affected a sensitive aquatic organism, Daphnia magna. We exposed Daphnia to surface waters known to be contaminated by agricultural runoff at two time points: during an extended dry period and after the first seasonal storm event. In surface waters, the concentrations of CHL, IMI, and other pesticides of concern increased after the storm event. Exposure to these waters caused Daphnia to be hypoactive, and their response to light varied with the concentration of surface water. We then exposed Daphnia to each chemical individually and then to mixtures of the two chemicals, at concentrations that occur in polluted waterways. Daphnia exposed to CHL and IMI mixtures were more active in response to light stimuli than the control group. Daphnia swimming behavior is a sensitive way to measure the biological effects of CHL, IMI, and surface waters. Abstract Pesticides with novel modes of action including neonicotinoids and anthranilic diamides are increasingly detected in global surface waters. Little is known about how these pesticides of concern interact in mixtures at environmentally relevant concentrations, a common exposure scenario in waterways impacted by pesticide pollution. We examined effects of chlorantraniliprole (CHL) and imidacloprid (IMI) on the sensitive invertebrate, Daphnia magna. Exposures were first performed using surface waters known to be contaminated by agricultural runoff. To evaluate the seasonal variation in chemical concentration and composition of surface waters, we tested surface water samples taken at two time points: during an extended dry period and after a first flush storm event. In surface waters, the concentrations of CHL, IMI, and other pesticides of concern increased after first flush, resulting in hypoactivity and dose-dependent photomotor responses. We then examined mortality and behavior following single and binary chemical mixtures of CHL and IMI. We detected inverse photomotor responses and some evidence of synergistic effects in binary mixture exposures. Taken together, this research demonstrates that CHL, IMI, and contaminated surface waters all cause abnormal swimming behavior in D. magna. Invertebrate swimming behavior is a sensitive endpoint for measuring the biological effects of environmental pesticides of concern.

Project description:Dispersal is fundamental to population dynamics. However, it is increasingly apparent that, despite most models treating dispersal as a constant, many organisms make dispersal decisions based upon information gathered from the environment. Ideally, organisms would make fully informed decisions, with knowledge of both intra-patch conditions (conditions in their current location) and extra-patch conditions (conditions in alternative locations). Acquiring information is energetically costly, however, and extra-patch information will typically be costlier to obtain than intra-patch information. As a consequence, theory suggests that organisms will often make partially informed dispersal decisions, utilising intra-patch information only. We test this proposition in an experimental two-patch system using populations of the aquatic crustacean, Daphnia carinata. We manipulated conditions (food availability) in the population's home patch, and in its alternative patch. We found that D. carinata made use of intra-patch information (resource availability in the home patch induced a 10-fold increase in dispersal probability) but either ignored or were incapable of using of extra-patch information (resource availability in the alternative patch did not affect dispersal probability). We also observed a small apparent increase in dispersal in replicates with higher population densities, but this effect was smaller than the effect of resource constraint, and not found to be significant. Our work highlights the considerable influence that information can have on dispersal probability, but also that dispersal decisions will often be made in only a partially informed manner. The magnitude of the response we observed also adds to the growing chorus that condition-dependence may be a significant driver of variation in dispersal.

Project description:Conservation translocations are frequently inhibited by extensive dispersal after release, which can expose animals to dispersal-related mortality or Allee effects due to a lack of nearby conspecifics. However, translocation-induced dispersals also provide opportunities to study how animals move across a novel landscape, and how their movements are influenced by landscape configuration and anthropogenic features. Translocation among populations is considered a potential conservation strategy for lesser prairie-chickens (Tympanuchus pallidicinctus). We determined the influence of release area on dispersal frequency by translocated lesser prairie-chickens and measured how lesser prairie-chickens move through grassland landscapes through avoidance of anthropogenic features during their dispersal movements. We translocated 411 lesser prairie-chickens from northwest Kansas to southeastern Colorado and southwestern Kansas in 2016-2019. We used satellite GPS transmitters to track 115 lesser prairie-chickens throughout their post-release dispersal movements. We found that almost all lesser prairie-chickens that survived from their spring release date until June undergo post-translocation dispersal, and there was little variation in dispersal frequency by release area (96% of all tracked birds, 100% in Baca County, Colorado, 94% in Morton County, Kansas, n = 55). Dispersal movements (male: 103 ± 73 km, female: 175 ± 108 km, n = 62) led to diffusion across landscapes, with 69% of birds settling >5 km from their release site. During dispersal movements, translocated lesser prairie-chickens usually travel by a single 3.75 ± 4.95 km dispersal flight per day, selecting for steps that end far from roads and in Conservation Reserve Program (CRP) grasslands. Due to this "stepping stone" method of transit, landscape connectivity is optimized when <5 km separates grassland patches on the landscape. Future persistence of lesser prairie-chicken populations can be aided through conservation of habitat and strategic placement of CRP to maximize habitat connectivity. Dispersal rates suggest that translocation is better suited to objectives for regional, rather than site-specific, population augmentation for this species.

Project description:Swimming motility is considered a beneficial trait among bacterial species as it enables movement across fluid environments and augments invasion of tissues within the host. However, non-swimming bacteria also flourish in fluid habitats, but how they effectively spread and colonize distant ecological niches remains unclear. We show that non-motile staphylococci can gain motility by hitchhiking on swimming bacteria, leading to extended and directed motion with increased velocity. This phoretic interaction was observed between Staphylococcus aureus and Pseudomonas aeruginosa, Staphylococcus epidermidis and P. aeruginosa, as well as S. aureus and Escherichia coli, suggesting hitchhiking as a general translocation mechanism for non-motile staphylococcal species. By leveraging the motility of swimming bacteria, it was observed that staphylococci can colonize new niches that are less available in the absence of swimming carriers. This work highlights the importance of considering interactions between species within polymicrobial communities, in which bacteria can utilize each other as resources.

Project description:Dispersal provides a key mechanism for geographical range shifts in response to changing environmental conditions. For mangroves, which are highly susceptible to climate change, the spatial scale of dispersal remains largely unknown. Here we use a high-resolution, eddy- and tide-resolving numerical ocean model to simulate mangrove propagule dispersal across the global ocean and generate connectivity matrices between mangrove habitats using a range of floating periods. We find high rates of along-coast transport and transoceanic dispersal across the Atlantic, Pacific, and Indian Oceans. No connectivity is observed between populations on either side of the American and African continents. Archipelagos, such as the Galapagos and those found in Polynesia, Micronesia, and Melanesia, act as critical stepping-stones for dispersal across the Pacific Ocean. Direct and reciprocal dispersal routes across the Indian Ocean via the South Equatorial Current and seasonally reversing monsoon currents, respectively, allow connectivity between western Indian Ocean and Indo-West Pacific sites. We demonstrate the isolation of the Hawaii Islands and help explain the presence of mangroves on the latitudinal outlier Bermuda. Finally, we find that dispersal distance and connectivity are highly sensitive to the minimum and maximum floating periods. We anticipate that our findings will guide future research agendas to quantify biophysical factors that determine mangrove dispersal and connectivity, including the influence of ocean surface water properties on metabolic processes and buoyancy behavior, which may determine the potential of viably reaching a suitable habitat. Ultimately, this will lead to a better understanding of global mangrove species distributions and their response to changing climate conditions.

Project description:Freshwater zooplankton is increasingly used to study effects of dispersal on community and metacommunity structure. Yet, it remains unclear how zooplankton disperses. Clearly, birds and wind play a significant role as zooplankton dispersal agents, but they may not always be the main vectors. This experimental study shows that a cosmopolitan aquatic insect, Notonecta, can be an important vector of cladoceran resting eggs (ephippia). Dispersing Notonecta frequently transported ephippia during flight, with a bias towards smaller ephippia in two species. A similar trend was present at the species level: Daphnia species with smaller ephippia were more often dispersed, suggesting that Notonecta could generate specific colonist communities. In addition, buoyancy appeared a critical trait, as non-floating ephippia of Daphnia magna were never dispersed. Our data suggest that Notonecta could be important dispersers of Daphnia, and that knowledge of dispersal dynamics of Notonecta may be used to predict Daphnia dispersal, colonization and resilience to disturbance.

Project description:Using data from 83 isolates from a single population, the population genomics of the microcrustacean Daphnia pulex are described and compared to current knowledge for the only other well-studied invertebrate, Drosophila melanogaster These two species are quite similar with respect to effective population sizes and mutation rates, although some features of recombination appear to be different, with linkage disequilibrium being elevated at short ([Formula: see text] bp) distances in D. melanogaster and at long distances in D. pulex The study population adheres closely to the expectations under Hardy-Weinberg equilibrium, and reflects a past population history of no more than a twofold range of variation in effective population size. Fourfold redundant silent sites and a restricted region of intronic sites appear to evolve in a nearly neutral fashion, providing a powerful tool for population genetic analyses. Amino acid replacement sites are predominantly under strong purifying selection, as are a large fraction of sites in UTRs and intergenic regions, but the majority of SNPs at such sites that rise to frequencies [Formula: see text] appear to evolve in a nearly neutral fashion. All forms of genomic sites (including replacement sites within codons, and intergenic and UTR regions) appear to be experiencing an [Formula: see text] higher level of selection scaled to the power of drift in D. melanogaster, but this may in part be a consequence of recent demographic changes. These results establish D. pulex as an excellent system for future work on the evolutionary genomics of natural populations.
