Performance in even a simple perceptual task depends on mouse secondary visual areas.

ABSTRACT: Primary visual cortex (V1) in the mouse projects to numerous brain areas, including several secondary visual areas, frontal cortex, and basal ganglia. While it has been demonstrated that optogenetic silencing of V1 strongly impairs visually guided behavior, it is not known which downstream areas are required for visual behaviors. Here we trained mice to perform a contrast-increment change detection task, for which substantial stimulus information is present in V1. Optogenetic silencing of visual responses in secondary visual areas revealed that their activity is required for even this simple visual task. In vivo electrophysiology showed that, although inhibiting secondary visual areas could produce some feedback effects in V1, the principal effect was profound suppression at the location of the optogenetic light. The results show that pathways through secondary visual areas are necessary for even simple visual behaviors.

Mouse visual cortex areas represent perceptual and semantic features of learned visual categories.

Project description:Associative memories are stored in distributed networks extending across multiple brain regions. However, it is unclear to what extent sensory cortical areas are part of these networks. Using a paradigm for visual category learning in mice, we investigated whether perceptual and semantic features of learned category associations are already represented at the first stages of visual information processing in the neocortex. Mice learned categorizing visual stimuli, discriminating between categories and generalizing within categories. Inactivation experiments showed that categorization performance was contingent on neuronal activity in the visual cortex. Long-term calcium imaging in nine areas of the visual cortex identified changes in feature tuning and category tuning that occurred during this learning process, most prominently in the postrhinal area (POR). These results provide evidence for the view that associative memories form a brain-wide distributed network, with learning in early stages shaping perceptual representations and supporting semantic content downstream.

Control of visual adaptation depends upon task.

Project description:The visual system optimizes its functioning for a given environment through processes collectively called adaptation. It is currently unknown, however, whether adaptation is affected by the particular task the observer performs within that environment. Two experiments tested whether this is the case. Observers adapted to high contrast grating patterns, and the decay of adaptation was measured using a version of the tilt-aftereffect, while they performed two different secondary tasks. One task involved judging the luminance of a small circular spot at fixation, and was expected to be unaffected by adaptation. The other secondary task involved judging a low contrast grating, and adaptation was expected to make this task difficult by reducing the visibility of the grating. Identical displays containing both a fixation spot and a grating were used for both tasks. Tilt-aftereffects were smaller when subjects concurrently performed the grating task than when they performed the fixation task. These results suggest that the control of adaptation, in this case its decay, is sensitive to the nature of the task the observer is performing. Adaptation may attempt to optimize vision with respect to many different criteria simultaneously; task is likely one of the criteria included in this process.

Perceptual learning of task-irrelevant features depends on the sensory context.

Project description:The brain has evolved to extract behaviourally meaningful information from the environment. For example, it has been shown that visual perceptual learning (VPL) can occur for task-irrelevant stimulus features when those features are consistently paired with internal or external reinforcement signals. It is, however, unclear whether or not task-irrelevant VPL is influenced by stimulus features that are unrelated to reinforcement in a given sensory context. To address this question, we exposed participants to task-irrelevant and subliminal coherent motion stimuli in the background while they performed a central character identification task. A specific motion direction was consistently paired with the task-targets, while two other directions occurred only with distractors and, thus, were unrelated to reinforcement. We found that the magnitude of VPL of the target-paired direction was significantly greater when the distractor-paired directions were close to the target-paired direction, compared to when they were farther. Thus, even very weak signals that are both subliminal and unrelated to reinforcement are processed and exert an influence on VPL. This finding suggests that the outcome of VPL depends on the sensory context in which learning takes place and calls for a refinement of VPL theories to incorporate exposure-based influences on learning.

Perceptual integration rapidly activates dorsal visual pathway to guide local processing in early visual areas.

Project description:Rapidly grouping local elements into an organized object (i.e., perceptual integration) is a fundamental yet challenging task, especially in noisy contexts. Previous studies demonstrate that ventral visual pathway, which is widely known to mediate object recognition, engages in the process by conveying object-level information processed in high-level areas to modulate low-level sensory areas. Meanwhile, recent evidence suggests that the dorsal visual pathway, which is not typically attributable to object recognition, is also involved in the process. However, the underlying whole-brain fine spatiotemporal neuronal dynamics remains unknown. Here we used magnetoencephalography (MEG) recordings in combination with a temporal response function (TRF) approach to dissociate the time-resolved neuronal response that specifically tracks the perceptual grouping course. We demonstrate that perceptual integration initiates robust and rapid responses along the dorsal visual pathway in a reversed hierarchical manner, faster than the ventral pathway. Specifically, the anterior intraparietal sulcus (IPS) responds first (i.e., within 100 ms), followed by activities backpropagating along the dorsal pathway to early visual areas (EVAs). The IPS activity causally modulates the EVA response, even when the global form information is task-irrelevant. The IPS-to-EVA response profile fails to appear when the global form could not be perceived. Our results support the crucial function of the dorsal visual pathway in perceptual integration, by quickly extracting a coarse global template (i.e., an initial object representation) within first 100 ms to guide subsequent local sensory processing so that the ambiguities in the visual inputs can be efficiently resolved.

Perceptual learning depends on perceptual constancy.

Project description:Perceptual learning refers to experience-induced improvements in the pick-up of information. Perceptual constancy describes the fact that, despite variable sensory input, perceptual representations typically correspond to stable properties of objects. Here, we show evidence of a strong link between perceptual learning and perceptual constancy: Perceptual learning depends on constancy-based perceptual representations. Perceptual learning may involve changes in early sensory analyzers, but such changes may in general be constrained by categorical distinctions among the high-level perceptual representations to which they contribute. Using established relations of perceptual constancy and sensory inputs, we tested the ability to discover regularities in tasks that dissociated perceptual and sensory invariants. We found that human subjects could learn to classify based on a perceptual invariant that depended on an underlying sensory invariant but could not learn the identical sensory invariant when it did not correlate with a perceptual invariant. These results suggest that constancy-based representations, known to be important for thought and action, also guide learning and plasticity.

Individual variability in functional connectivity predicts performance of a perceptual task.

Project description:People differ in their ability to perform novel perceptual tasks, both during initial exposure and in the rate of improvement with practice. It is also known that regions of the brain recruited by particular tasks change their activity during learning. Here we investigate neural signals predictive of individual variability in performance. We used resting-state functional MRI to assess functional connectivity before training on a novel visual discrimination task. Subsequent task performance was related to functional connectivity measures within portions of visual cortex and between visual cortex and prefrontal association areas. Our results indicate that individual differences in performing novel perceptual tasks can be related to individual differences in spontaneous cortical activity.

Visual Perceptual Load Attenuates Age-Related Audiovisual Integration in an Audiovisual Discrimination Task.

Project description:Previous studies confirmed that the cognitive resources are limited for each person, and perceptual load affects the detection of stimulus greatly; however, how the visual perceptual load influences audiovisual integration (AVI) is still unclear. Here, 20 older and 20 younger adults were recruited to perform an auditory/visual discrimination task under various visual perceptual-load conditions. The analysis for the response times revealed a significantly faster response to the audiovisual stimulus than to the visual stimulus or auditory stimulus (all p < 0.001), and a significantly slower response by the older adults than by the younger adults to all targets (all p ≤ 0.024). The race-model analysis revealed a higher AV facilitation effect for older (12.54%) than for younger (7.08%) adults under low visual perceptual-load conditions; however, no obvious difference was found between younger (2.92%) and older (3.06%) adults under medium visual perceptual-load conditions. Only the AV depression effect was found for both younger and older adults under high visual perceptual-load conditions. Additionally, the peak latencies of AVI were significantly delayed in older adults under all visual perceptual-load conditions. These results suggested that visual perceptual load decreased AVI (i.e., depression effects), and the AVI effect was increased but delayed for older adults.

Functional specialization of mouse higher visual cortical areas.

Project description:The mouse is emerging as an important model for understanding how sensory neocortex extracts cues to guide behavior, yet little is known about how these cues are processed beyond primary cortical areas. Here, we used two-photon calcium imaging in awake mice to compare visual responses in primary visual cortex (V1) and in two downstream target areas, AL and PM. Neighboring V1 neurons had diverse stimulus preferences spanning five octaves in spatial and temporal frequency. By contrast, AL and PM neurons responded best to distinct ranges of stimulus parameters. Most strikingly, AL neurons preferred fast-moving stimuli while PM neurons preferred slow-moving stimuli. By contrast, neurons in V1, AL, and PM demonstrated similar selectivity for stimulus orientation but not for stimulus direction. Based on these findings, we predict that area AL helps guide behaviors involving fast-moving stimuli (e.g., optic flow), while area PM helps guide behaviors involving slow-moving objects.

Functional specialization of seven mouse visual cortical areas.

Project description:To establish the mouse as a genetically tractable model for high-order visual processing, we characterized fine-scale retinotopic organization of visual cortex and determined functional specialization of layer 2/3 neuronal populations in seven retinotopically identified areas. Each area contains a distinct visuotopic representation and encodes a unique combination of spatiotemporal features. Areas LM, AL, RL, and AM prefer up to three times faster temporal frequencies and significantly lower spatial frequencies than V1, while V1 and PM prefer high spatial and low temporal frequencies. LI prefers both high spatial and temporal frequencies. All extrastriate areas except LI increase orientation selectivity compared to V1, and three areas are significantly more direction selective (AL, RL, and AM). Specific combinations of spatiotemporal representations further distinguish areas. These results reveal that mouse higher visual areas are functionally distinct, and separate groups of areas may be specialized for motion-related versus pattern-related computations, perhaps forming pathways analogous to dorsal and ventral streams in other species.

Reinforcement learning can account for associative and perceptual learning on a visual-decision task.

Project description:We recently showed that improved perceptual performance on a visual motion direction-discrimination task corresponds to changes in how an unmodified sensory representation in the brain is interpreted to form a decision that guides behavior. Here we found that these changes can be accounted for using a reinforcement-learning rule to shape functional connectivity between the sensory and decision neurons. We modeled performance on the basis of the readout of simulated responses of direction-selective sensory neurons in the middle temporal area (MT) of monkey cortex. A reward prediction error guided changes in connections between these sensory neurons and the decision process, first establishing the association between motion direction and response direction, and then gradually improving perceptual sensitivity by selectively strengthening the connections from the most sensitive neurons in the sensory population. The results suggest a common, feedback-driven mechanism for some forms of associative and perceptual learning.
