Immobilization-Free Electrochemical Sensor Coupled with a Graphene-Oxide-Based Aptasensor for Glycated Albumin Detection.

ABSTRACT: An immobilization-free electrochemical sensor coupled with a graphene oxide (GO)-based aptasensor was developed for glycated human serum albumin (GHSA) detection. The concentration of GHSA was monitored by measuring the electrochemical response of free GO and aptamer-bound GO in the presence of glycated albumin; their currents served as the analytical signals. The electrochemical aptasensor exhibited good performance with a base-10 logarithmic scale. The calibration curve was achieved in the range of 0.01-50 µg/mL. The limit of detection (LOD) was 8.70 ng/mL. The developed method was considered a one-drop measurement process because a fabrication step and the probe-immobilization process were not required. This simple sensor offers a cost-effective, rapid, and sensitive detection method, and could be an alternative approach for determination of GHSA levels.

Electrochemical Quantification of Glycated and Non-glycated Human Serum Albumin in Synthetic Urine.

Project description:A polymer-based electrode capable of specific detection of human serum albumin, and its glycated derivatives, is described. The sensor is constructed from a glass microscope slide coated with a synthesized, polythiophene film bearing a protected, iminodiacetic acid motif. The electrode surface is then further elaborated to a functional biosensor through deprotection of the iminodiacetic acid, followed by metal-affinity immobilization of a specific and high-affinity, albumin ligand. Albumin was then quantified in buffer and synthetic urine via electrochemical impedance spectroscopy. Glycated albumin was next bound to a boronic acid-modified, single-cysteine dihydrofolate reductase variant to quantify glycation ratios by square-wave voltammetry. The platform offers high sensitivity, specificity, and reproducibility in an inexpensive arrangement. The detection limits exceed the requirements for intermediate-term glycemic control monitoring in diabetes patients at 5 and 1 nM for albumin and its glycated forms, respectively.

Electrochemical Immunoassay for Determination of Glycated Albumin using Nanozymes.

Project description:We developed a new nanozyme-based electrochemical immunoassay method for the monitoring of glycated albumin (GA) known to reflect short-term glycaemic levels. For this study, we synthesized urchin-like Pt nanozymes (uPtNZs) and applied them to colorimetric and electrochemical assays for sensitive determination of GA in total human serum albumin (tHSA) using 3,3',5,5'-tetramethylbenzidine (TMB) and thionine as substrates, respectively. The uPtNZs showed peroxidase-mimic activity in the presence of hydrogen peroxide. Boronic acid (BA)-agarose bead was used to capture GA through specific cis-diol interactions. uPtNZs were modified with GA antibody (GA-Ab) to form sandwich complexes with GA/BA-agarose bead. The amount of Ab-uPtNZ/GA/BA-agarose bead complex increased with increasing percentage of GA in 50?mg/mL tHSA. The colorimetric assay exhibited linearity from 0.02 to 10% (10?µg/mL - 5?mg/mL) GA with an LOD of 0.02% (9.2?µg/mL). For electrochemical assay, GA was detected from 0.01 to 20% (5?µg/mL - 10?mg/mL) with an LOD of 0.008% (3.8?µg/mL). The recovery values of measured GA in human plasma samples were from 106 to 107%. These results indicate that electrochemical assay using uPtNZs is a promising method for determining GA.

Novel Aminosilane (APTES)-Grafted Polyaniline@Graphene Oxide (PANI-GO) Nanocomposite for Electrochemical Sensor.

Project description:Lead is a potentially toxic element (PTE) that has several adverse medical effects in humans. Its presence in the environment became prominent due to anthropogenic activities. The current study explores the use of newly developed composite materials (organic-inorganic hybrid) based on PANI-GO-APTES for electrochemical detection of Pb2+ in aqueous solution. The composite material (PANI-GO-APTES) was synthesized by chemical method and was characterized with SEM, XPS, XEDS, XRD, TGA, FTIR, EIS and CV. The result of characterization indicates the successful synthesis of the intended material. The PANI-GO-APTES was successfully applied for electrochemical detection of Pb2+ using cyclic voltammetry and linear sweep voltammetry method. The limit of detection of Pb2+ was 0.0053 µM in the linear range of 0.01 µM to 0.4 µM. The current response produced during the electrochemical reduction of Pb2+ catalyzed by PANI-GO-APTES was also very repeatable, reproducible and rapid. The application of PANI-GO-APTES-modified GCE in real sample analysis was also established. Therefore, PANI-GO-APTES is presented as a potential Pb2+ sensor for environmental and human health safety.

Disposable Electrochemical Aptasensor Based on Graphene Oxide-DNA Complex as Signal Amplifier towards Ultrasensitive Detection of Ochratoxin A.

Project description:Signal amplification is crucial in developing a reliable disposable screen-printed carbon electrodes (SPCEs)-based biosensor for analyte detection with a narrow detection window. This work demonstrated a novel label-free electrochemical aptasensor based on SPCEs for the ultrasensitive detection of ochratoxin A (OTA). The graphene oxide-DNA (GO-DNA) complex as a signal amplifier with easy preparation was investigated for the first time. The proposed aptasensor based on the SPCEs/GO/cDNA-aptamer/3D-rGO-AuNPs structure was formed through the hybridization of aptamer-linked 3D-rGO/AuNPs and its complementary DNA-linked GO (GO-cDNA). The presence of OTA was discerned by its specific aptamer forming a curled OTA-aptamer complex and releasing the GO-cDNA from the surface of SPCEs. The resulting OTA-aptamer complex hindered interfacial electron transfer on the sensing surface, leading to the decreased peak current. The GO-cDNA further amplified the peak current change. This electrochemical aptasensor showed a low limit of detection of 5 fg/mL as well as good reproducibility with the relative standard deviation (RSD) of 4.38%. Moreover, the detection result of OTA in the rice and oat samples was comparable with that of the enzyme-linked immunosorbent assay (ELISA) kit. In general, the OTA aptasensor used in this work with convenient preparation, low-cost, good selectivity, high sensitivity and acceptable reproducibility can be proposed as a reliable point-of-care (POC) technique for OTA determination.

Albuminuria detection using graphene oxide-mediated fluorescence quenching aptasensor.

Project description:A simple and sensitive graphene oxide-mediated fluorescence quenching aptasensor is developed to quantify albuminuria in urine samples. The developed aptasensor used the specific target binding property of aptamer and fluorescence quenching property of graphene oxide to determine the concentration of human serum albumin in urine. The limit of detection of the developed platform is 0.05 µg.mL-1 and the detection range is 0.1-600 µg.mL-1, which covers the albuminuria concentration range present in normal human urine and the urine of the patient with chronic kidney disease. This approach can be modified to measure albuminuria using a high-throughput quantification platform and portable point of care testing. In addition, the production cost for one reaction is cheaper than those for the standard automated method. Therefore, this aptasensor has significant potential for commercialization and public use.•Our protocol is customized by using the fluorescence quenching property of graphene oxide and specific binding property of human serum albumin aptamer to detect human serum albumin in urine sample•The limit of detection of our developed platform is 0.05 µg.mL-1•The detection range of our aptasensor is 0.1-600 µg.mL-1.

A Bifunctional Nanosilver-Reduced Graphene Oxide Nanocomposite for Label-Free Electrochemical Immunosensing.

Project description:A bi-functional material based on silver nanoparticles (AgNPs)-reduced graphene oxide (rGO) composite for both electrode modification and signal generation is successfully synthesized for use in the construction of a label-free electrochemical immunosensor. An AgNPs/rGO nanocomposite is prepared by a one-pot wet chemical process. The AgNPs/rGO composite dispersion is simply cast on a screen-printed carbon electrode (SPCE) to fabricate the electrochemical immunosensor. It possesses a sufficient conductivity/electroreactivity and improves the electrode reactivity of SPCE. Moreover, the material can generate an analytical response due to the formation of immunocomplexes for detection of human immunoglobulin G (IgG), a model biomarker. Based on electrochemical stripping of AgNPs, the material reveals signal amplification without external redox molecules/probes. Under optimized conditions, the square wave voltammetric peak current is responded to the logarithm of IgG concentration in two wide linear ranges from 1 to 50 pg.ml-1 and 0.05 to 50 ng.ml-1, and the limit of detection (LOD) is estimated to be 0.86 pg.ml-1. The proposed immunosensor displays satisfactory sensitivity and selectivity. Importantly, detection of IgG in human serum using the immunosensor shows satisfactory accuracy, suggesting that the immunosensor possesses a huge potential for further development in clinical diagnosis.

Gold Nanoparticles dotted Reduction Graphene Oxide Nanocomposite Based Electrochemical Aptasensor for Selective, Rapid, Sensitive and Congener-Specific PCB77 Detection.

Project description:Gold nanoparticles (AuNP) dotted reduction graphene oxide (RGO-AuNP) is used as a platform for an aptamer biosensor to selectively detect 3,3'4,4'-polychlorinated biphenyls (PCB77). By anchoring aptamers onto the binding sites of RGO-AuNP and making use of the synergy effect of RGO and AuNP, the RGO-AuNP based biosensor exhibits superior analytical performances to AuNP based biosensor in terms of sensitivity and repeatability. The sensitivity of RGO-AuNP based aptamers (RGO-AuNP-Ap) biosensor (226.8 μA cm-2) is nearly two times higher than that of Au based biosensors (AuNP-Ap/Au electrode, 147.2 μA cm-2). The RGO-AuNP-Ap/Au biosensor demonstrated a linear response for PCB77 concentrations between 1 pg L-1 and 10 μg L-1, with a low limit of detection (LOD) of 0.1 pg L-1. The superb LOD satisfies the exposure thresholds (uncontaminated water < 0.1 ng L-1) set out by International Agency for Research on Cancer (IARC) and the Environmental Protection Agency (EPA). The proposed biosensor can be a powerful tool for rapid, sensitive and selective detection of PCBs on site.

Simple Synthesis of Large Graphene Oxide Sheets via Electrochemical Method Coupled with Oxidation Process.

Project description:In this paper, we report a simple two-step approach for the synthesis of large graphene oxide (GO) sheets with lateral dimensions of ≈10 μm or greater. The first step is a pretreatment step involving electrochemical exfoliation of graphite electrode to produce graphene in a mixture of H2SO4 and H3PO4. The second step is the oxidation step, where oxidation of exfoliated graphene sheets was performed using KMnO4 as the oxidizing agent. The oxidation was carried out for different times ranging from 1 to 12 h at ∼60 °C. Prepared GO batches were characterized using a number of spectroscopy and microscopy techniques such as X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FTIR), and UV-visible spectroscopy. Raman and thermogravimetric analysis techniques were used to study the degree of oxidation in the as-synthesized GO batches. The UV-visible absorption spectrum showed an intense peak at 230 nm and an adjacent band at 300 nm corresponding to π-π* and n-π* transitions in all samples. Normalized FTIR plots were used to calculate the relative percentages of oxygen-containing functional groups, which were found to be maximum in GO (6 h). Boehm titration was used to quantify the functional groups present on the GO surface. Overall GO sheets obtained after 6 h of oxidation, GO (6 h), were found to be the best. XRD pattern of GO (6 h) revealed a characteristic peak at 2θ = 8.88°, with the corresponding interplanar spacing between the layers being 0.995 nm, which is among the best with respect to the previous methods reported in the literature. Raman spectroscopy showed that the degree of defect (I D/I G) area ratio for GO (6 h) was 1.24, which is higher than that obtained for GO (1.18) prepared by widely used Marcano's approach.

A simple label-free electrochemical sensor for sensitive detection of alpha-fetoprotein based on specific aptamer immobilized platinum nanoparticles/carboxylated-graphene oxide.

Project description:A label-free electrochemical aptamer-based sensor has been fabricated for alpha-fetoprotein (AFP) detection. Platinum nanoparticles on carboxylated-graphene oxide (PtNPs/GO-COOH) modified screen-printed graphene-carbon paste electrode (SPGE) was utilized as an immobilization platform, and the AFP aptamer was employed as a bio-recognition element. The synthesized GO-COOH helps to increase the surface area and amounts of the immobilized aptamer. Subsequently, PtNPs are decorated on GO-COOH to enhance electrical conductivity and an oxidation current of the hydroquinone electrochemical probe. The aptamer selectively interacts with AFP, causing a decrease in the peak current of the hydroquinone because the binding biomolecules on the electrode surface hinder the electron transfer of the redox probe. Effects of aptamer concentration and AFP incubation time were studied, and the current changes of the redox probe before and after AFP binding were investigated by square wave voltammetry. The developed aptasensor provides a linear range from 3.0-30 ng mL-1 with a detection limit of 1.22 ng mL-1. Moreover, the aptamer immobilized electrode offers high selectivity to AFP molecules, good stability, and sensitive determination of AFP in human serum samples with high recoveries.

Sensitive detection of 4-nitrophenol based on pyridine diketopyrrolopyrrole-functionalized graphene oxide direct electrochemical sensor.

Project description:For highly sensitive detection of 4-nitrophenol (4-NP) in the environment, a novel pyridine diketopyrrolopyrrole-functionalized graphene oxide (PDPP-GO) composite was constructed for the first time by an improved Hummers' method. Herein, PDPP was completely dissolved in sulfuric acid (6 mol L-1) and reacted with GO, promoting PDPP evenly adhering to the GO surface. Moreover, the specific surface area increased from 15.51 to 22.033 m2 g-1. Infrared spectroscopy and X-ray photoelectron spectroscopy simultaneously demonstrated that PDPP was bound to GO by the strong intermolecular hydrogen bonding and π-π stacking conjugation. During the cyclic voltammetry test, the PDPP-GO coated glassy carbon electrode (PDPP-GO/GCE) direct electrochemical sensor gave expression to the best electrocatalytic activity for 4-nitrophenol detection than GO/GCE and bare GCE. Under optimization conditions, the as-prepared PDPP-GO/GCE sensor brought out remarkable sensitivities of 18.54 (0.5-50 μM) and 6.61 μA μM-1 cm-2 (50-163 μM) in the linear detection of 4-NP. Besides, a low detection limit of 0.10 μM, reliable long-term stability, excellent selectivity, and reproducibility were obtained. In the real sample test, the PDPP-GO/GCE demonstrated sensitive and reliable determination.
